Differential Analysis of Fresh and Dried Lilii Bulbus Using LC-MS Combined with Multivariate Statistical Analysis
Objective:To compare the differences in components between fresh and dried(freeze-dried and oven-dried)samples of different species of Lilii Bulbus(Lilium lancifolium Thunb.and L.brownii F.E.Brown var.viridulum Baker)based on ultra-performance liquid chromatography-quadrupole time-of-flight mass spectrometry(UPLC Q-TOF MS).Methods:UPLC Q-TOF MS technology was employed to analyze fresh and dried samples of different Lilii Bulbus species.Principal component analysis(PCA)and orthogonal partial least squares discriminant analysis(OPLS-DA)combined with single-factor analysis were employed to screen for differential compounds in fresh and oven-dried Lilii Bulbus samples.Results:The UPLC Q-TOF MS showed that the main components of fresh and dried Lilii Bulbus samples were similar.PCA results showed that fresh and freeze-dried Lilii Bulbus samples clustered into one group,while both differed significantly from oven-dried samples.This suggests that the overall chemical compounds of freeze-dried Lilii Bulbus samples are similar to fresh samples.In contrast,the chemical composition of oven-dried samples changed after blanching and drying,making them different from the former two.OPLS-DA results showed 387 differential components between fresh and oven-dried Lilium lancifolium Thunb.samples and 335 differential components between fresh and oven-dried Lilium Bulbus samples.8 components were preliminarily identified through database comparison.Conclusion:Compared to oven-dried Lilium Bulbus,the components of fresh and freeze-dried Lilii Bulbus are more similar,providing a basis for the clinical application and modern Chinese medicine formulation development of Lilii Bulbus.
