Extraction of Red Pigment with Inhibitory Effect on COX-2 from Kadsurae Caulis
Objective:To optimize the extraction conditions of red pigment from Kadsurae Caulis,examine the basic properties and the inhibitory effect of the red pigment on cyclooxygenase-2(COX-2),and provide a reference for the clinical application of Kadsurae Caulis and the development of related preparations.Methods:With absorbance as the indicator,single factor and orthogonal experiments were carried out to optimize the liquid-to-solid ratio,extraction time,and ethanol concentration for the extraction of red pigment from Kadsurae Caulis.Furthermore,the light,thermal,and pH stability of the red pigment was examined,and the COX-2 Inhibitor Screening Kit was used to determine the inhibitory effect of the red pigment on COX-2.Results:The absorbance of the red pigment was the largest when the extraction was carried out with 50%ethanol and at the liquid-to-solid ratio of 110 for 12 h.The extracted pigment had good stability at room temperature and pH 1-9.Moreover,the extracted pigment inhibited the activity of COX-2,with the half maximal inhibitory concentration(IC50)of only 2.04 ng·mL-1 crude drug.Conclusion:This study optimized the extraction conditions and examined the stability of red pigment from Kadsurae Caulis.The extracted red pigment demonstrated strong inhibitory effect on COX-2.The findings lay a foundation for the comprehensive utilization and development of the resources and the clinical application of Kadsurae Caulis.
