Mining of Biomarkers for the Ageing of Citri Reticulatae'Chachi'Pericarpium by Single Factor Design for Packaging Containers with Different Breathability
Objective:To study the biomarkers reflecting the ageing characteristics of Citri Reticulatae'Chachi'Pericarpium.Methods:First,the Citri Reticulatae'Chachi'Pericarpium samples with the same source were placed in the packaging containers with different breathability for three years to simulate the samples with different ageing levels.Then,high performance liquid chromatography-diode array detector(HPLC-DAD)was employed to perform full wavelength(200-400 nm)scanning on the samples,and the characteristic chromatographic peaks with differences between samples were analyzed.Finally,silica gel column chromatography and semi-preparative high-performance liquid chromatography were used for separation and purification,and the structures of compounds were identified by nuclear magnetic resonance spectroscopy and mass spectrometry.Results:The content of phloroglucinol-O-glucoside showed a negative relationship with the ageing level,which indicated that the sample with higher ageing level had lower content of this component.The content of 5-hydroxymethyl-2-furancarboxylic acid and vanillic acid was positively correlated with the ageing level,which indicated that the sample with higher ageing level had higher content of the two components.Conclusion:Phloroglucinol-O-glucoside,5-hydroxymethyl-2-furancarboxylic acid,and vanillic acid can serve as biomarkers reflecting the ageing characteristics of Citri Reticulatae'Chachi'Pericarpium.The results lay a foundation for elucidating the principle of the ageing process and quantifying the aging degree of this herbal medicine.
