Differential Analysis of Ajania purpurea C.Shih Based on HPLC Fingerprint
Objective:To establish a high-performance liquid chromatography(HPLC)fingerprint and multi-component content determination method for Ajania purpurea C.Shih.,and compare and analyze 19 batches of samples collected from different locations and times,as well as medicinal parts,to assess their differences.Methods:HPLC was used with 0.1%phosphoric acid solution and acetonitrile as the mobile phase,gradient elution,and a detection wavelength of 320 nm.A fingerprint was established.Based on similarity evaluation,combined with cluster analysis,principal component analysis(PCA),and orthogonal partial least squares discriminant analysis(OPLS-DA),the content of chlorogenic acid and isochlorogenic acid in A.purpurea was determined simultaneously.Results:The fingerprint spectra for the 19 batches of A.purpurea samples were established,with similarity ranging from 0.810 to 0.997.Fourteen common peaks were identified,with chlorogenic acid and isochlorogenic acid recognized as key components,which can serve as quality markers for evaluating A.purpurea from different sources and collection times,and medicinal parts.Cluster analysis and PCA of the 14 common peaks showed minimal component differences between different locations,but significant chemical differences between different collection times.Quantitative analysis revealed statistically significant differences in the chemical component content of A.purpurea medicinal parts and those from different locations,with higher levels of chlorogenic acid and isochlorogenic acid in samples collected in August.Conclusion:The establishment of the HPLC fingerprint and content determination method for A.purpurea is feasible and can provide a reference for the overall quality evaluation of A.purpurea medicinal materials.
