Study on Safflower Standard Decoction Based on HPLC Characteristic Chromatogram and Content Determination
Objective:This paper aims to establish the HPLC characteristic chromatogram of safflower standard decoction and determine the content of kaempferol and 6-methoxykaempferol.Methods:15 batches of safflower standard decoction were prepared,and the extraction rate was determined.The chromatogram of standard decoction was established by high performance liquid chromatography,and common characteristic peaks of standard decoction were analyzed by mass spectrometry and compared with reference standards.The similarity was evaluated.The contents and transfer rate of kaempferol and 6-methoxykaempferol were also determined.Results:The extraction rate of 15 batches of safflower standard decoction ranges from 26.0%to 36.5%,with an average of 31.7%.There are 12 common characteristic peaks in the chromatogram,and seven components are identified,namely,syringin(peak 1),hydroxysafflor yellow A(peak 3),tryptophan(peak 4),kaempferol-3-O-sophoroside(peak 7),rutin(peak 8),kaempferol-3-O-rutin(peak 9),and bidenoside C(peak 12).The similarity is greater than 0.915.The mass fraction of kaempferol in standard decoction is 0.39-1.00 mg·g-1,and the transfer rate is 45.9%-78.9%.The mass fraction of 6-methoxykaempferol is 0.20-1.15 mg·g-1,and the transfer rate is 41.2%-89.8%.Conclusion:The chemical components of 15 batches of safflower decoction pieces and standard decoction can be transferred well and stably,and the established methods are simple and reliable,which can provide a reference for the establishment and evaluation of quality standards.
