Effects of Different Mucor circinelloides Strains and Microalgae on Seed Germination and Seedling Growth of Polygala tenuifolia
Objective:To explore the effects of Mucor circinelloides,microalgae,and their combinations on seed germination and seedling growth of Polygala tenuifolia Willd.Methods:This study designed six treatments:sterile water(control),microalgae,M.circinelloides DF20,M.circinelloides zh,M.circinelloides DF20+microalgae,and M.circinelloides zh+microalgae.The effects of different treatments on the germination rate,germination potential,seed vitality index,seedling growth parameters(such as radicle length,young shoot length,and fresh weight),and soluble protein content in the seedlings of P.tenuifolia were evaluated.The colonization of M.circinelloides in the radicle of P.tenuifolia was observed by acid fuchsin staining.Results:Both M.circinelloides DF20 and M.circinelloides zh could colonize the radicle of P.tenuifolia seeds and form mycelia and sporangia.Treating the seeds with M.circinelloides DF20 alone showed the strongest promoting effect on germination,increasing the germination potential and germination rate by 12.15%and 10.26%,respectively,compared with the control.Soaking the seeds with microalgae increased the vitality index of P.tenuifolia seeds by 53.62%(P<0.05),and this index was positively correlated with the radicle length,young shoot length,and fresh weight of seedlings(P<0.001).Both microalgae alone and in combination with each of the two strains of M.circinelloides significantly promoted the growth of P.tenuifolia seedlings,and microalgae alone demonstrated the best promoting effect,increasing the radicle length,young shoot length,and fresh weight by 56.39%,90.51%,and 36.84%,respectively,compared with the control(P<0.05).The soluble protein content in P.tenuifolia seedlings increased after the seeds were soaked in the culture of M.circinelloides.Particularly,when M.circinelloides zh was combined with microalgae,the soluble protein content increased by 33.05%(P<0.05).Conclusion:M.circinelloides can promote the accumulation of soluble proteins in P.tenuifolia seedlings,and soaking the seeds with microalgae can significantly improve the seed vitality of P.tenuifolia.Both microalgae alone and in combination with M.circinelloides can promote seedling growth.However,the combined application of M.circinelloides and microalgae did not show the expected synergistic effect.Therefore,it is recommended to solely employ microalgae at the early stage in the cultivation of P.tenuifolia.
