Influence of Electron Beam Irradiation on Sterilization Effect and Active Component Contents of Mabaosongtang Powder
Objective:This paper aims to investigate the effect of electron beam irradiation on the main active components and sterilization effect of Tibetan Medicine Mabaosongtang Powder.Methods:The samples of Mabaosongtang Powder were irradiated with electron beams at 0,2,4,6,8,and 10 kGy respectively,and the appearance characteristics,thin-layer chromatography,microscopic characteristics,the content of Lac dye,acetylshikonin,and rubicin,fingerprint similarity,and microbial levels of the samples were compared before and after irradiation.Results:After irradiation,the sample odor has no obvious change,and the total color difference(∆E*)<0.5.A large number of multicellular non-glandular hair,catheters,cork cells,and pigment blocks can be seen under the microscope,and in the thin-layer chromatography,spots in the same color show in the same position as the control such as Radix Onosmatis,Radix et Rhizoma Rubiae,and Lacca.There are no significant changes in the contents of active components including acetylshikonin,rubicin,and Lac dye,and the similarity of high-performance liquid chromatography(HPLC)fingerprints is 0.998 or above.The contents of bacteria,mold,and yeast are significantly decreased(P<0.01).When the electron beam irradiation reaches 4 kGy,the microbial levels comply with the provisions of the fourth part of the 2020 edition of the Pharmacopoeia of the People's Republic of China.Conclusion:Electron beam irradiation has no significant effect on the appearance characteristics,microscopic characteristics,thin-layer chromatography,content,and chemical components of the main active components of Mabaosongtang Powder and achieves a good sterilization effect at 4 kGy.It provides a reference for the application of electron beam irradiation technology in the sterilization of Mabaosongtang Powder.
