Target gene prediction and bioinformatics analysis for hsa-miR-181a-5p
Objective To systematically analyze the bioinformatics of hsa-miR-181a-5p,predict its target gene,gene function and signaling pathway of hsa-miR-181a-5p,and annotate its biological function.Methods Data-base was used to locate hsa-miR-181a-5p gene and analyze its sequence conservation and expression in human tissues and organs.The target gene prediction analysis was carried out by using database.GO function analysis,KEGG pathway enrichment analysis and protein-protein interaction(PPI)network analysis were performed to predict the intersecting target genes.Finally,A549 cells were transfected with hsa-miR-181a-5p,and transcrip-tome sequencing was performed to verify the bioinformatics prediction results.Results The results showed that the genes were located in chr1 and chr9 chromosomes of human genome,and the mature sequences were highly conserved among species.There are 483 intersecting target genes in the three databases,including PTEN,MAPK8,AK73 and other genes,which are distributed in the cytoplasm and other cell components.The intersec-ting target genes,enriched in MAPK,PI3K-Akt,insulin resistance,miRNA in cancer and other signaling path-ways,with proteins encoded by them forming complex network maps,perform biological functions such as protein binding,and participate in biological processes such as signal transduction.After the experiment,the transcrip-tome sequencing results verified the accuracy of bioinformatics analysis and provided strong support for prediction and enrichment results.Conclusion hsa-miR-181a-5p plays an important role in a variety of biological proces-ses,especially in the occurrence and development of diseases of the nervous system,blood and immune system,and cancer,and it will become a new target for the pathogenesis and prevention of related diseases.
