Effects of total ginsenosides on IL-6 and IL-1β in mice with bleomycin-induced pulmonary fibrosis
Objective To study the effects of total ginsenosides on pro-inflammatory cytokines IL-6 and IL-1 β in lung tissues of mice with bleomycin-induced pulmonary fibrosis.Methods Sixty male Kunming mice were ran-domly divided into a normal control group,a model group,and a positive drug pirfenidone group(100 mg/kg)and total ginsenosides(40,80,160 mg/kg)groups;Except for the normal control group,the remaining five groups were constructed with a mouse lung fibrosis model by single-dose drops of bleomycin(3 mg/kg)through the oral endotracheal tube.The drug was administered by gavage on the next day of modelling,once a day for 14 consecutive days,and the lung tissues were taken to observe the pathological morphology,the degree of inflam-mation and fibrotic changes in the lungs.Type-Ⅰ collagen(COL Ⅰ),interleukin-6(IL-6)and interleukin-1 β(IL-1 β)protein levels were measured in the lung tissues of mice with pulmonary fibrosis.Results After 14 days of intraoral tracheal drip injection of bleomycin,the lung tissues of mice in the model group were obviously bruised and solid,alveolitis and pulmonary fibrosis were obvious,and the levels of COL Ⅰ,IL-6 and IL-1 β pro-tein in the lung tissues were increased significantly in comparison with those in the normal control group.When compared with the model group,total ginsenosides and pirfenidone effectively improved the degree of alveolitis and pulmonary fibrosis in the model mice,and significantly reduced the levels of COL Ⅰ,IL-6 and IL-1 β pro-tein in the lung tissues.Conclusion Total ginsenosides ameliorated bleomycin-induced pulmonary fibrosis in mice in association with the inhibition of inflammatory factors IL-6 and IL-1β protein levels.
