The role of zinc enrichment in lipopolysaccharide and dexamethasone induced cleft palate in mice
Objective To investigate the role of zinc-enriched diet in lipopolysaccharide and dexamethasone in-duced cleft palate in mice,and to explore the possible mechanism.Methods 120 pregnant mice were divided in-to normal zinc control(C),lipopolysaccharide(LPS),dexamethasone(DEX),zinc-enriched diet(Zn),Zn+LPS(ZLPS),and Zn+DEX(ZDEX)groups.Starting from ED0.5,the first 3 groups were given normal zinc feed of 30 mg/kg,and the last 3 groups were given zinc-enriched diet of 100 mg/kg;LPS and ZLPS groups were injected with 0.15 mg/kg lipopolysaccharide in the neck on ED8.5,while DEX and ZDEX groups were in-jected with 6 mg/kg dexamethasone intraperitoneally for three consecutive days from ED 10.5.Mice were eutha-nized on ED 13.5 d,ED 14.5 d,and ED 15.5 d.The palates of the fetus were observed under microscope and the incidence of cleft palate was calculated;HE staining was used to observe the development status of the palatal process;the expression of Sp5,Wiz,Ptch1 and Gli3 in the palatal process was determined by immunohistochem-ical staining,RT-qPCR and WB.Results LPS and DEX produced cleft palate,and the incidence was higher with DEX;At ED 13.5 d,LPS increased the average optical density value and Sp5 protein expression in the pal-atal process,while Zn decreased the average optical density value and Sp5 and Wiz protein expression in the pal-atal process,while increased Wiz mRNA.The average optical density,Sp5 mRNA,and Sp5 protein expression of the palatal process in the ZLPS group were significantly lower than those in the LPS group,while the ZDEX group were opposite to those in the DEX group;the expression of Ptch1 mRNA and Ptch1 protein in LPS and Zn group increased,but the expression of Gli3 mRNA in LPS group decreased,while the expression of Gli3 mRNA in Zn group increased;The expression of Ptch1 mRNA and its protein in ZDEX group was significantly increased compared to DEX group,while the expression of Gli3 mRNA was reduced;The expression of Ptch1 mRNA and its protein in ZLPS group decreased compared to LPS group.Conclusion The zinc-enriched diet had antagonistic effects on cleft palate induced by LPS in pregnant mice,which was possibly related to the regulation of Sp5 and Wiz expression,but had no obvious antagonistic effects on DEX-induced cleft palate.
