Establishment and evaluation of airway remodeling model induced by IL-1β in chronic obstructive pulmonary disease
Objective To establish and systematically evaluate an IL-1β-induced airway remodeling model in mice with chronic obstructive pulmonary disease (COPD).Methods Fifteen C57BL/6J mice were divided into control group,7-days and 14-day groups.Ad-IL-1βwas injected intratracheally in the 7-day and 14-day groups,and Ad-LacZ was injected intratracheally in the control group.Samples were collected from the control group af-ter 14 days,and samples were collected from the experimental group at 7 days and 14 days,respectively,to de-termine whether the model was successfully established.The general condition and pulmonary function of mice was recorded,the lung tissue was observed after Masson and HE staining,and inflammatory factors in peripheral blood was detected by ELISA.Results Compared with the control group,the pulmonary function of mice in the 7-day group and 14-day group was significantly decreased,a large number of inflammatory cells infiltrated around the small airways,collagen deposition was significant,and inflammatory factors in the peripheral blood were sig-nificantly increased (P<0.05).Conclusion The airway remodeling model of chronic obstructive pulmonary dis-ease can be successfully established by intratracheal administration of IL-1β.
