Effects of Yishen Tiaojing Formula Regulating PI3K/Akt Pathway on Ovarian Function and Apoptotic Protein Expression in Rats with Early-Onset Ovarian Insufficiency
Objective:To investigate the effect of Yishen Tiaojing Formula(YTF)on premature ovarian insufficiency(POI)rats and its related mechanism.Methods:In 60 SD rats with normal estrous cycle,10 rats were randomly selected as blank group,and the remaining 50 rats were intraperitoneally injected cyclophosphamide(CTX)to induce POI rat model.After successful modeling,the rats were randomly divided into model group,control group,and Chinese medicine low,medium and high dose groups,and the corresponding drugs were gavaged for 14 days.After the treatment,the rats in each group were euthanized and their general condition,estrus cycle,ovarian index,and sex hormone levels were observed.The ovarian tissue structure was observed by HE staining,and the mRNA and protein expressions of PI3K,Akt,Bcl-2 and Bax were detected by RT-qPCR and immunohistochemistry.Results:Compared with blank group,in model group,the body mass of rats decreased significantly(P<0.000 1),estrous cycle was prolonged(P<0.000 1)and ovarian indices significantly decreased(P<0.000 1),E2 and AMH levels decreased(P<0.000 1,P<0.001),FSH and LH levels increased(P<0.000 1),mRNA and protein expressions of PI3K,Akt and Bcl-2 significantly decreased(P<0.000 1,P<0.001),Bax mRNA and its protein expression increased(P<0.000 1).Compared with the model group,the general condition of the rats in the drug administration groups was significantly improved,the body mass increased in different degrees(P<0.05,P<0.001,P<0.000 1),the estrous cycle was shortened(P<0.000 1),and the ovarian index increased(P<0.000 1).The levels of E2 and AMH were increased(P<0.000 1,P<0.01),the levels of FSH and LH decreased(P<0.01,P<0.001,P<0.000 1),and the mRNA and protein expressions of PI3K,Akt and Bcl-2 significantly increased(P<0.01,P<0.001,P<0.000 1),the expression of Bax mRNA and protein decreased significantly(P<0.01,P<0.000 1),and showed a significant dose dependence,in which the high dose group had the best improvement effect.Conclusion:Yishen Tiaojing Formula may improve the ovarian function of POI model rats by regulating PI3K/Akt pathway and apoptosis-related proteins.
