Experimental Study on the Effect of Traditional Chinese Medicine Combined with Acupuncture on Pre-Gastric Cancer Model Rats
Objective:To explore the effect of traditional Chinese medicine combined with acupuncture on pre-gastric cancer model rats.Methods:A hundred and twenty adult SD rats were randomly divided into control group,model group,traditional Chinese medicine group,and combination group,with 30 rats in each group.Except for the control group,the other groups were treated with N-methyl-N′-nitroso-guanidine comprehensive modeling method to create the spleen deficiency type pre-gastric cancer model.The traditional Chinese medicine group was gavaged with 5.4 g/kg traditional Chinese medicine suspension 2 ml;the combination group was gabaged with traditional Chinese medicine combined with acupuncture;and the control group and model group were given the same amount of normal saline in the same way,for 3 weeks.The changes of body mass,24 h food intake and organ coefficient were observed.The contents of TCRαβ+CD161a+,CD4+and CD4+/CD8+in peripheral blood of rats were detected.The levels of CEA,CA199 and CA724 in rat plasma were detected by electrochemiluminescence method.The expression of TLR-NF/κB signaling pathway protein in rat stomach was detected.Results:Compared with the control group,body mass,24 h food intake,spleen index,thymus index,TCRαβ+CD161a+,CD4+ratio and CD4+/CD8+ratio of rats in model group were significantly decreased(P<0.05);and the proportion of serum CD8+,CEA level,CA199 level,CA724 level,and the relative expression levels of TLR-4 and NF-κB p65 protein in gastric tissue were significantly increased(P<0.05).Compared with model group,body mass,24 h food intake,spleen index,thymus index,TCRαβ+CD161a+,CD4+ratio and CD4+/CD8+ratio of rats in traditional Chinese medicine group and combination group were significantly increased(P<0.05);while the proportion of serum CD8+,CEA level,CA199 level,CA724 level,and the relative expression levels of TLR-4 and NF-κB p65 protein in gastric tissue were significantly decreased(P<0.05).Compared with traditional Chinese medicine group,24 h food intake,spleen index,thymus index and CD4+level in combination group were significantly increased(P<0.05),while the levels of CEA,CA199 and CA724 in serum and the relative expression levels of TLR-4 and NF-κB p65 in gastric tissue were significantly decreased(P<0.05).Conclusion:Traditional Chinese medicine combined with acupuncture can reduce the level of serum tumor markers,regulate the activity of T lymphocytes and NK cells,and enhance immune regulation in rats with splen deficiency type precancerous gastric cancer,and its mechanism may be related to the regulation of TLR-NF/κB signaling pathway.
