Diuretic Effects of Alcohol Extract from Malva Fructus Based on Rat Urine Biomarkers
Objective:To evaluate the diuretic mechanism of the alcohol extract of Malva fructus using the analysis method of urine metabolites.Methods:Fifty male SD rats were randomly divided into normal group(Z group),high dose group of alcohol extract of Malva fructus(DG group),medium dose group of alcohol extract of Malva fructus(DZ group),low dose group of alcohol extract of Malva fructus(DD group)and positive control group(Y group).After the rat water load model was prepared,urine was collected in a metabolic cage within 0-1,1-2,2-4,4-6,6-24 h.The contents of urine K+,Cl-,Na+and serum ADH,ANP,ALD,cAMP,cGMP,AQP1 and AQP2 were detected according to the requirements of the kit.Metabolomics method was used to detect endogenous metabolites in urine,and metabolic differences were characterized to search for differential metabolites in urine.Results:Compared with group Z,urine volume in group Y increased significantly at 0-1,1-2,2-4 h,and the difference was statistically significant(P<0.05,P<0.01).The urine volume of DD group increased significantly at 2 to 4 h and 6 to 24 h,and the difference was statistically significant(P<0.05,P<0.01).Urine volume of DZ group and DG group increased significantly at 2-4 h and 4-6 h,and the difference was statistically significant(P<0.05).Compared with Y group,urine volume in DD group,DZ group and DG group decreased significantly at 0-1 and 1-2 h,and the difference was statistically significant(P<0.05).Compared with group Z,the contents of Na+and Cl-in urine significantly increased in each group of the alcohol extract of Malva fructus,and the difference was statistically significant(P<0.01).Compared with Y group,Cl-content in DG group significantly increased,and Na+and Cl-contents in DZ and DD groups significantly increased,with statistical significance(P<0.05).Compared with group Z,the contents of ANP and AQP2 in DG group significantly increased,and the content of ADH significantly decreased,with statistical significance(P<0.05);the contents of cAMP and AQP1 in DD group significantly increased,with statistical significance(P<0.05).Compared with Y group,ADH content in DG group significantly decreased,and ANP content significantly increased,with statistical significance(P<0.05).Metabolomics analysis showed that compared with group Z,a total of 6 different metabolites in the DD group,8 in the DZ group and 10 in the DG group were screened in the urine samples,and the levels of these metabolites showed significant changes.Conclusion:Hydroxyproline,gluconic acid and catechol are urine biomarkers involved in the diuretic mechanism of alcohol extract of Malva fructus.
