摘要

开发一批用于竹亚科(Bambusoideae)植物的EST-SSR分子标记,利用其判定样本亲缘关系,结合已有的纤维原料竹种的开发研究数据,给出竹亚科植物纤维资源评价方法与筛选建议.从纤维原料竹种毛竹(Phyllostachys edulis)和早竹(Phyllostachys violascens)转录组中挖掘EST-SSR位点并设计引物,利用PCR-PAGE对 50 对候选引物进行筛选验证.使用生物信息学软件对条带迁移信息进行处理,得到 17 份竹样本的亲缘关系.比对近缘种在文献中的竹纤维数据,得出竹亚科植物纤维资源评价方法和筛选建议.结果表明,筛选出 14 对高通用性和多态性引物;有 6 对竹样本有较近的亲缘关系,且亲缘关系较近的品种,其竹纤维性能也较为相似.得出结论,在开发竹亚科植物纤维原料新品种时,可以从备选品种中,优先考虑纤维性能优良的品种的近缘种.

Abstract

This study aimed to develop a set of EST-SSR(Expressed Sequence Tag-Simple Sequence Repeat)molecular markers for plants in the Bambusoideae subfamily,utilizing them to determine the genetic relationships among samples.By integrating these markers with existing research data on the development of fiber-producing bamboo species,we provided evaluation methods and screening recommendations for fiber resources within the Bambusoideae subfamily.EST-SSR loci were mined from the transcriptomes of two fiber-producing bamboo species,Phyllostachys edulis and Phyllostachys violascens,and primers were designed accordingly.Fifty candidate primer pairs were screened and validated using PCR-PAGE(Polymerase Chain Reaction-Polyacrylamide Gel Electrophoresis).Bioinformatics software was employed to process the band migration information,revealing the genetic relationships among 17 bamboo samples.By comparing the bamboo fiber data of closely related species from the literature,we formulated evaluation methods and screening suggestions for fiber resources in the Bambusoideae subfamily.The results indicated that 14 primer pairs with high universality and polymorphism were successfully screened.Among the 17 bamboo samples,six pairs were found to have close genetic relationships,and bamboo species with closer genetic ties exhibited similar fiber properties.Consequently,we concluded that when developing new varieties of fiber-producing bamboo in the Bambusoideae subfamily,priority should be given to the close relatives of those varieties with superior fiber properties.
