摘要

为了解郑州地区犬细小病毒(CPV)的流行特征及遗传变异情况,采集 81 份 2017-2022 年郑州市疑似感染CPV犬的粪便或肛门拭子进行 PCR检测.随机挑选出 21 份经 PCR检测的 CPV阳性样本,扩增其 VP2 基因,连接至载体并测序,用MegAlign软件对流行株进行同源性和氨基酸变异分析,并通过MEGA6.0 软件构建基因遗传进化树.结果显示:2017-2018 年郑州地区 CPV流行基因型为 New CPV-2a亚型,2019-2022 年则以 CPV-2c亚型为主导.21 株 CPV流行株与疫苗株的核苷酸同源性为 98.3%~99.0%,其中 CPV-79 流行株与疫苗株的核苷酸同源性最低,仅为 98.3%,且该毒株在 VP2 蛋白 GH 环区出现多位点氨基酸突变;部分 CPV-2c 亚型流行株第 5 和 370位氨基酸出现 A到 G和 Q到 R的变异.研究结果表明:郑州地区当前流行基因型为 CPV-2c亚型,虽出现多位点氨基酸突变,但未形成明显的国内进化分支.值得注意的是,New CPV-2a亚型在进化过程中形成了不同分支且存在抗原突变的风险.本研究丰富了郑州地区 CPV的分子流行病学调查数据,也为我国 CPV 流行疫苗株的研制奠定了基础.

Abstract

In order to understand the epidemiological characteristics and genetic variation of canine parvovirus(CPV)in Zhengzhou area,81 faecal or anal swabs suspected to be infected with CPV in Zhengzhou from 2017 to 2022 were collected for PCR detection.21 CPV positive samples were randomly selected by PCR,amplified VP2 gene,connected to vector and sequenced.Homology and amino acid variation of the epidemic strains were analyzed by MegAlign software,and genetic evolution tree was constructed by MEGA6.0 software.The results showed that the prevalent CPV genotype in Zhengzhou was New CPV-2a subtype from 2017 to 2018,and CPV-2c subtype was dominant from 2019 to 2022.The nucleotide homology between 21 CPV epidemic and vaccine strains was 98.3%~99.0%.The nucleotide homology between CPV-79 and vaccine strain was the lowest(98.3%),and the CPV-79 strain showed multi-site amino acid mutation in GH ring of VP2 protein.In addition,some CPV-2c subtype epidemic strains showed A to G and Q to R mutations in amino acids at positions 5 and 370.The results showed that the current prevalent genotype in Zhengzhou is CPV-2c subtype,although there are multi-site amino acid mutations,but there is no obvious domestic evolutionary branch.However,it is worth noting that the New CPV-2a subtype has evolved into different branches and is at risk of antigenic mutations.This study enriches the molecular epidemiological data of CPV in Zhengzhou area and lays a foundation for the development of CPV epidemic vaccine strains in China.
