Methylation Level of SLCO1B3 Gene and Eggshell Color Analysis
This study systematically investigated the changes in eggshell color, the expression levels, and the methylation status of the SLCO1B3 gene in 162 healthy Lu's green-shelled laying hens during different egg-laying periods, and analyzed the correlation between these three factors. The aim is to reveal the molecular mechanisms behind the variation in eggshell color depth and to provide theoretical support for achieving eggshell color consistency in production.Measurements were conducted using NR color difference meter,qRT-PCR,and bisulfite sequencing on eggs produced at various ages(25-65 weeks). Results showed that eggshell color gradually lightened with increasing laying age.Expression levels of the SLCO1B3 gene were significantly positively correlated with eggshell color, while gene methylation levels exhibited a significant negative correlation with both gene expression and eggshell color. The study indicates that as the laying age of Luoyang green-shelled hens increases, eggshell color lightens,a phenomenon closely associated with decreased SLCO1B3 gene expression and increased gene methylation levels.
