Study on antidepressant mechanism of Purslane oleracea L.based on inflammatory cytokines in mice
The study aimed to investigate the effect of Portulaca oleracea on antidepressant action in mice and to reveal the target and mechanism of action of Portulaca oleracea on antidepressants by detecting the biochemical indexes of TNF-α,IL-1β,and IL-6 in mouse brain tissues.The chronic unpredictable mild stimulation(CUMS)method was used to establish a depressed mouse model,and the success of the model was verified by the sugar-water preference test and the hanging tail immobility time test,and further analyzed the effects of different doses of Portulaca oleracea on the changes of depression-like behaviors as well as the levels of TNF-α,IL-1β,and IL-6 in the brain tissues of mice.The experimental results showed that the low,medium,and high dose groups of Portulaca oleracea can improve the behavioral indexes of CUMS mice.Among them,the middle and high dose groups could significantly reduce the levels of TNF-α and IL-6 in their brain tissues(p<0.05)but had no significant effect on IL-1β.It is inferred that Portulaca oleracea can improve depressive symptoms in CUMS model mice,which may be related to the inhibition of the expression levels of TNF-α and IL-6.
