    Characterization of the duration of immunity of Brucella abortus strain RB51 vaccination in cattle after experimental challenge

    查看更多>>摘要:? 2022Fifty-two, Hereford heifers were obtained from brucellosis-free herds and randomly assigned to Brucella abortus strain RB51 (RB51) vaccination (n = 32) or control (n = 20) treatments. Vaccinates received 1010 colony-forming units (CFU) of a commercial lyophilized RB51 vaccine. Immunologic responses after inoculation demonstrated significantly greater (P < 0.05) antibody, interferon-γ responses, and proliferative responses to RB51 antigens in cattle vaccinated with RB51 as compared to controls. A subgroup of control and vaccinated cattle were experimentally challenged at approximately 4, 5, and 6 years after inoculation with 107 CFU of B. abortus strain 2308 at 170–180 days gestation. After experimental challenge, 6 of 14 (43 %) control animals aborted at a higher rate (P < 0.05) when compared to RB51 vaccinates in years 4 and 5, but not year 6 (0 %, 10 %, and 50 %, respectively). When comparing recovery of Brucella from all tissues except head lymph nodes draining the site of challenge, RB51 vaccinates had reduced infection rates (P < 0.05) after experimental challenge at 4 years (14 %), but not at 5 or 6 years (78 % and 67 %, respectively) when compared to non-vaccinated cattle (93 %). Our data suggests that calfhood vaccination with RB51 does not induce lifelong immunity and suggests implementation of booster vaccination by 4–5 years of age should be utilized in endemic areas to maintain high levels of protection.

    Uptake of baits by wild badgers: Influences of deployment method, badger age and activity patterns on potential delivery of an oral vaccine

    查看更多>>摘要:? 2022In parts of the United Kingdom and Ireland, the European badger is a wildlife host for Mycobacterium bovis (the causative agent of bovine tuberculosis). Badger vaccination is one management option for reducing disease spread. Vaccination is currently achieved by parenteral vaccination of captured badgers, but an oral vaccine delivered in a bait may provide an additional approach in the future. We conducted two field experiments in wild badger populations to identify factors that influence uptake (% of individuals with evidence of bait consumption) of candidate oral vaccine baits. In both instances, baits containing the biomarker iophenoxic acid (as a proxy for the vaccine) were fed at burrows (setts) associated with badger social groups (study A = 48 groups, study B = 40 groups). Badgers were captured following a period of bait deployment to quantify uptake in relation to age, sex and social group. In addition, groups were allocated different treatments and the bait deployment protocol was varied to identify effects on uptake. Study A tested the effects of season, bait type, bait placement and packaging, while study B investigated the effects of bait quantity and badger activity levels. Overall bait uptake was low (Study A = 24 %, Study B = 37 %) but this varied among treatment groups (range 0–58 %). In both studies, bait uptake was significantly higher in cubs than in adults. Uptake was substantially higher where baits were placed directly into sett entrances (rather than under tiles near setts), and by badgers caught at main setts rather than at outlier setts. Season, bait type and packaging did not influence uptake, while increasing the quantity of bait available increased uptake by cubs but not by adults. Levels of badger activity at setts varied over time (suggesting potential disturbance), but were positively associated with levels of bait uptake.

    Bayesian estimation of sensitivity and specificity of fecal culture, fecal PCR and serum ELISA for diagnosis of Mycobacterium avium subsp. paratuberculosis infections in sheep

    查看更多>>摘要:? 2022 Elsevier B.V.The objective of the present study was to evaluate the diagnostic accuracy of the individual fecal culture (IFC), fecal PCR (FPCR), and serum ELISA for the detection of Mycobacterium avium subsp. paratuberculosis (MAP) infections in sheep from four governorates in Egypt, using a latent class model (LCM) fitted within a Bayesian framework. Furthermore, the within-governorate prevalence of MAP infection in sheep was estimated as a secondary objective. Fecal and blood samples were collected from 370 sheep in four Egyptian governorates. Fecal samples were analyzed by IFC and RT-PCR based on ISMav2 gene, while ELISA was performed on serum samples. The sensitivity (Se) and specificity (Sp) of the three diagnostic tests were estimated using a three-tests-four-populations Bayesian LCM to obtain posterior estimates [medians and 95% Bayesian credible intervals (95% BCI)] for each parameter. The median Se estimates (95% BCI) for IFC, FPCR, and serum ELISA were 31.8% (22.8–41.4), 49.7% (31.8–79.9), and 61.2% (39.8–81.4), respectively. The median Sp estimates (95% BCI) for IFC, FPCR, and serum ELISA were 97.7% (96.1–98.9), 97.7% (95.6–99.5), and 98.4% (96.9–99.3), respectively. The median within-governorate paratuberculosis prevalence (95% BCI) was 5.2% (1.1–13.6), 8.4% (2.9–17.7), 9.4% (3.0–20.7), and 18.2% (10.5–29.5) for the Gharbia, Menoufia, Qalyubia, and Kafr El-Sheikh governorates, respectively. In conclusion, at a ratio of the optical density (OD) sample/OD positive control threshold of > 45%, ELISA showed the highest Se among the three tests and comparable Sp to IFC and FPCR. The test ELISA evaluated in this study is an interesting alternative for detecting MAP in sheep due to its higher Se, lower cost, and shorter turnaround laboratory time compared to IFC and FPCR.

    Effect of pooling family oral fluids on the probability of PRRSV RNA detection by RT-rtPCR

    查看更多>>摘要:? 2022 Elsevier B.V.Family oral fluids (FOFs) are an aggregate sample type shown to be a cost-efficient and convenient option for determining the porcine reproductive and respiratory syndrome virus (PRRSV) status of weaning age pigs. This study investigates the effect of pooling PRRSV-positive FOF samples with PRRSV-negative FOF samples at different levels (1/3, 1/5, 1/10, 1/20) on the probability of PRRSV RNA detection by reverse-transcription real-time polymerase chain reaction (RT-rtPCR). Mathematical models were built to assess how much the probability of RT-rtPCR PRRSV detection changed with increasing proportion of PRRSV-positive samples present within pools and how partially sampling a farrowing room influenced the probability of RT-rtPCR detection of PRRSV RNA in pooled samples at different prevalence scenarios. A general example of a guideline for FOF-based sampling under different prevalence scenarios to detect PRRSV RNA by RT-rtPCR with at least 95 % certainty is presented. At the sample level, the probability of detecting PRRSV RNA by RT-rtPCR decreased from 100 % to 87 %, 68 %, and 26 % when diluting up to 1/20 for PRRSV positive FOF having an initial Cycle threshold (Ct) below 34, between 34 and 36, or above 36, respectively. When PRRSV prevalence is near-zero (1 or 2 litters positive out of 56), the most cost-efficient farrowing room sampling strategy to detect PRRSV RNA with at least 95 % certainty was pooling FOF samples up to 1/10; at higher prevalence (≥ 3 of 56 litters positive), the most cost-efficient strategy was submitting samples in pools of 20. Subsampling a farrowing room for FOF pools was also demonstrated to be a valuable cost-saving strategy. Overall, based on the conditions of this study, pooling FOFs up to 1/20 is a valid option in situations of cost constraint and regardless of pooling level chosen, capturing as many litters as possible improves the probability of PRRSV detection.

    Prevalence and antimicrobial resistance of Salmonella enterica subsp. diarizonae serovar 61:k:1,5,(7) in Swiss sheep flocks

    查看更多>>摘要:? 2022 The AuthorsSalmonella (S.) enterica subspecies diarizonae (IIIb) serovar 61:k:1,5,(7) (S. IIIb 61:k:1,5,(7)) is considered to be sheep-associated, as it can be found in the intestine, tonsils and nose of clinically healthy sheep, but it has also been described in separate clinical disorders in sheep. In particular, S. IIIb 61:k:1,5,(7) is described as the causative agent of chronic proliferative rhinitis (CPR) in sheep. In Switzerland, CPR in sheep due to S. IIIb 61:k:1,5,(7) was first described in 2017 in a flock of Texel sheep. Therefore, we assessed the prevalence of S. IIIb 61:k:1,5,(7) within the Swiss sheep population using a representative sampling strategy. From May 2017 to June 2018 a total of 681 nasal swabs from individual clinically healthy sheep of 141 different flocks throughout Switzerland were taken. Swabs were analysed by selective enrichment for the presence of S. IIIb 61:k:1,5,(7). Additionally, antimicrobial resistance of the isolates was determined by broth microdilution. A total of 146 out of 681 nasal swabs tested positive for S. IIIb 61:k:1,5,(7), which corresponds to a prevalence on animal level of 21% (95%CI 18%?25%). In 73 out of 141 flocks tested, at least one sheep tested positive for S. IIIb 61:k:1.5,(7), resulting in a minimal prevalence on flock level of 52% (95%CI 43%?60%). Positive flocks were found in all cantons except the canton of Jura. Adults were significantly more affected than sheep under one year/lambs and positive sheep were found in several breeds. No microbiologically resistant isolates were detected, except for one isolate showing resistance against ampicillin. Because of its widespread occurrence in the Swiss sheep population, further research should focus on the pathogenic impact of S. IIIb 61:k:1,5,(7) on the health status of sheep.

    Potential impacts to antibiotics use around the dry period if selective dry cow therapy is adopted by dairy herds: An example of the western US

    查看更多>>摘要:? 2022Mastitis is a prevalent and expensive disease in dairy herds worldwide. Blanket dry cow therapy (BDCT), in which all quarters of all cows are infused with antimicrobials at the dry-off, is a cornerstone for mastitis control in many countries. An alternative approach is the use of selective dry cow therapy (SDCT), in which only cows with high risk for intramammary infection (IMI) at dry-off receive antimicrobials. Our objectives in this cross-sectional study were to estimate the potential reduction in the use of antimicrobials if SDCT was adopted in the US by using cow-level dairy herd data and to describe the factors associated with cows being classified as high-risk for an IMI at dry-off. Besides, we aimed to describe the seasonality in IMI at dry-off. We used cow-level somatic cell score (SCS) test-day data from herds in the western US (DHIA, Dairy Herd Improvement Association, AgriTech, Visalia, CA) to create five scenarios to classify cows as high risk for IMI at dry-off. Associations between cow-level data and state were also used in logistic regression models. We also calculated the average animal-defined daily dosage of antimicrobials per cow per year around the dry period if a BDCT or SDCT approach is used, adjusting for the risk of cases of clinical mastitis in the next lactation for the SDCT approach. The point prevalence of IMI at the last test-day before dry-off varied between 15.0 % and 54 % for primiparous and 34.0 % and 85 % for multiparous cows, depending on the scenario. By extrapolating our results obtained from using data from dairies enrolled in the DHIA testing program for the western US, we demonstrated that regardless of the criteria used to classify cows as high risk of IMI at dry-off (scenarios 1–5), if selective dry cow therapy is adopted in the US, the dairy industry could reduce the use of antimicrobials around the dry-off between 31 % and 66 %. Multiparous cows had greater odds of being classified as high-risk than primiparous cows. Cows dried off in the spring, summer, and fall had lower odds of being classified as high-risk compared to cows dried off in the winter. Advanced days in milk at dry-off was associated with greater risk of IMI at dry-off. Greater milk yield and higher protein percentage at the last test-day before dry-off were associated with decreased odds of a cow being classified as high-risk at dry-off, cows in small herds had greater odds of being classified as high-risk at dry-off, and a variation among states was observed.

    Molecular survey of porcine respiratory disease complex pathogens in Brazilian wild boars

    查看更多>>摘要:? 2022 Elsevier B.V.Wild boar (Sus scrofa) is an exotic invasive species in Brazil and may be a reservoir for several pathogens, including those related to the porcine respiratory disease complex (PRDC), a critical infectious disease in pig production. The objective of this study was to investigate viral and bacterial pathogens related to PRDC in free-living wild boars from Brazil. Eighty animals were examined in search of genomes of porcine circovirus 2 (PCV2), Torque teno Sus virus 1a (TTSuV1a) and 1b (TTSuV1b), Influenza A virus (IAV), Actinobacillus pleuropneumoniae, Glaesserella parasuis, Pasteurella multocida, and Mycoplasma hyopneumoniae. The results demonstrated that 57.5% (46/80) of the animals had at least one detected pathogen, and 11.3% of them (9/80) were co-infected. TTSuV1a was the most prevalent genome, for which risk factors were associated with increased contact between wild boars and other animals. The other pathogens were detected at much lower frequencies or not detected (M. hyopneumoniae and IAV). An additional IAV serology search identified H1N1pdm09 antibodies in 35.5% (16/45) of the wild boars, bringing concern related to public health. In conclusion, wild boars are infected with pathogens that cause swine diseases, so their eventual contact with domestic pigs might risk animal production in Brazil.

    Resistance to extended-spectrum cephalosporins in Escherichia coli and Salmonella enterica isolated from food-producing animals: Ecological study from selected national surveillance programs

    查看更多>>摘要:? 2022Extended-spectrum cephalosporins (ESC) are categorized by World Health Organization as critically important antimicrobials with limited therapeutic alternatives for the treatment of severe bacterial infections in humans. Preserving the effectiveness of ESC requires continuous monitoring of resistance and comparison of associated data across national surveillance programs in the face of globalization. In this ecological study, we compared ESC resistance in Escherichia coli and Salmonella enterica isolated from food-producing animals from 2003 to 2019 between nine countries (Canada, Denmark, Finland, Japan, Netherlands, Norway, Sweden, United Kingdom, and the United States). Using the beta-regression model, compared to Canada, non-selective ESC-R Salmonella enterica was less likely isolated from food producing animals in other eight countries (Odds ratio range: 0.07–0.76). We observed an interaction between the country and the year with a significantly decreased proportion (P < 0.05) of non-selective ESC-R Escherichia coli from the Netherlands, the United Kingdom, and the United States compared to Canada over the years. There was a linear correlation between non-selective ESC-R Escherichia coli and ESC use from Netherlands (Spearman's ρ = 0.91, P < 0.0001). For the six European countries, the interaction between the country and year showed a significant decrease in the proportion of selective ESC-R Escherichia coli over the years for the Netherlands compared to Denmark (P = 0.002). While there were variations in the proportion of beta-lactamase genes reported over the years, blaCTX-M and blaCMY-2 genes were commonly detected among the selective ESC-R Escherichia coli. This study reveals variability in the recovery of ESC-resistant bacteria among the countries that seems likely influenced by the individual country policy on the use of critically important antimicrobials and resistance surveillance programs. However, there is a need for harmonization and consistency in food animal sources of bacterial isolates used in surveillance programs within and between the countries for easy comparability.

    Seroprevalence of pathogenic Leptospira spp. in domestic dogs from southern Chile and risk factors associated with different environments

    查看更多>>摘要:? 2022 The AuthorsLeptospirosis is a neglected and severe zoonotic bacterial disease that affects canine populations worldwide, and it is present in animals from different settings, such as urban and rural environments. Therefore, a cross-sectional study was conducted on owned domestic dogs from urban and rural origins in southern Chile. The study aimed: 1) to estimate the true prevalence of pathogenic Leptospira in dogs from urban and rural environments in southern Chile, 2) to determine the serovars circulating in an endemic area of Chile, 3) to assess potential risk factors associated with seropositivity in dogs from urban and rural environments. Blood samples from 706 canines were collected, and the serum was tested with Micro-Agglutination-Test (MAT), using a panel of 13 serovars. A Bayesian approach was applied to estimate True prevalence (TP). In addition, dog owners answered a questionnaire that had prompts regarding dog characteristics and potential risk factors. The effect of these factors on the risk of being infected was analysed using a logistic regression model. The TP for urban dogs was 9.4% and 9.1% for rural animals. However, seroprevalence was higher in dogs with different individual, lifestyle, and habitat characteristics regarding their origin. For example, in urban animals, those living in places with rodents roaming near houses and dogs with rodent hunting habits had decreased risk for the seropositivity in MAT. On the other hand, in rural dogs, those of a recognized breed, with contact with cattle and sheep, living with more than one cat at home and living in places close to natural water sources have more risk to seropositivity in MAT. The presence of anti-Leptospira antibodies is widespread in pet dogs in urban and rural environments in southern Chile, and individual, lifestyle and habitat characteristics of the animals, according to the origin, are associated with the seropositivity in MAT. Therefore, the adoption of preventive measures is urgent, in addition to increase the awareness either in public health institutions, practitioners and companion animals’ owners, considering the zoonotic potential of Leptospira infection and the close contact between people and their pets.

    Adverse event classification and signal detection of data from the customer service and pharmacovigilance of a multinational veterinary pharmaceutical company

    查看更多>>摘要:? 2022 Elsevier B.V.The pharmacovigilance of a veterinary company may include the analysis of spontaneous reports of adverse events (AE) related to its products. The present study developed an AE classification flowchart to analyze AE notified to the customer service and pharmacovigilance department of a multinational veterinary pharmaceutical company in Brazil. The product-AE binomials using the flowchart were characterized in terms of their frequencies and subsequently, three signal detection models were used: Reporting Odds Ratio, Bayesian confidence propagation neural network, and Gamma Poisson Shrinker. The signals detected with the three methods were classified according to their intensity, always with the most intense signal in the first position. Among the signals detected by the three methods, the positions of each signal were summed to obtain an aggregated classification that considered the results of the three methods and allowed a serial interpretation. Among the 531 reports, 20 types of AE; 88 product-AE binomials were identified. From the total of reports, seven were signs identified by the three methods. The classification of AE following explicit criteria and the combined use of more than one signal detection method enhances spontaneous-reports-based pharmacovigilance.
