查看更多>>摘要:This article presents the current status of the development of bioeconomy in the Czech Republic. Although the country has no unified strategy on bioeconomy, there are ambitious governmental innovation strategies and focused strategies for each region. Traditionally, the country has had a strong research performance in chemistry and biology, which together with developed agriculture, forestry and food industries, provides a good foundation for the development of locally based circular systems. Moreover, the government supports research on tools and applications of new plant breeding technologies, including genome editing, and there is a strong initiative from the research community calling to update EU regulatory policy in this area.

查看更多>>摘要:Complex recombinant glycoproteins produced as potential biopharmaceuticals in goat's milk have an aberrant pattern of N-glycosylation due to the lack of multi-antennary structures. Overexpression of glycosyltransferases may increase oligosaccharide branching of the desired glycoproteins. Here, human erythropoietin fused to human IgG Fc (EPO-Fc) was co-expressed with N-acetyl-glucosaminyltransferase-IVa (GnT-IVa) by adenoviral transduction in goat mammary gland to evaluate the in vivo modification of N-glycosylation pattern in this tissue. Adenoviral vectors, containing the EPO-Fc and GnT-IVa sequences were assembled for in vitro and in vivo expression in mammalian cell culture or in goat mammary gland. Protein detection was assessed by gel electrophoresis and western blot, and N-glycans were identified by HPLC and mass spectrometry. GnT-IVa overexpression and its colocalization with EPO-Fc in the Golgi apparatus of SiHa cells were demonstrated. N-glycan analysis of in vitro and in vivo expression of EPO-Fc modified by GnT-IVa (EPO-Fc/GnT-IVa) showed an increase in high molecular weight structures, which corresponded to tri- and tetra-antennary N-glycans in SiHa cells and mostly tri-antennary N-glycans in goat's milk from transformed mammary tissue. The results confirmed that successful modification of the goat mammary gland secretion pathway could be achieved by co-expressing glycoenzymes together with the glycoprotein of interest. This is the first report of modification of the N-glycosylation pattern in the goat mammary gland in vivo, and constitutes a step forward for improving the use of the mammary gland as a bioreactor for the production of complex recombinant proteins.

This also applies to the present century and its sustainability challenges at the planetary, regional and local levels. Therefore, it is of great importance and relevance to move forward on the journey which has been started globally to address the not insignificant number of challenges. It is however essential to go beyond descriptive work by continuing with novel, inspiring and interconnected steps to find solutions to overcome these challenges. As this huge task also requires multidimensional communication, understanding and actions across different regions, cultures, disciplines and knowledge areas, the development of a common conceptual framework such as the concept of bioeconomy has been accepted globally as very valuable. The momentum which has been created in more than 50 countries around the world by the growing number of activities, initiatives and strategies in bioeconomy is very encouraging. This offers great opportunities to mobilize even more stakeholders in science and industry, as well as society, to join the bioeconomy journey. Strategic high-level concepts such as preserving the value of the natural capital of planet earth, connecting economy and ecology, sustaining the boundary conditions and habitability of our biosphere are highly important. It is also essential on the bioeconomy journey to connect with highly specific and actionable missions, programs and plans towards sustainable economic growth under the boundary conditions of our planet.
查看更多>>摘要:The production, characterization and bioactivities of exopolysaccharides (EPSs) from a thermophilic bacterium Geobacillus sp. strain WSUCF1 were investigated. Using glucose as a carbon source 525.7 mg/L of exoproduct were produced in a 40-L bioreactor at 60 degrees C. Two purified EPSs were obtained: EPS-1 was a glucomannan containing mannose and glucose in a molar ratio of 1:0.21, while EPS-2 was composed of mannan only. The molecular weights of both EPSs were estimated to be approximately 1000 kDa, their FTIR and NMR spectra indicated the presence of alpha-type glycosidic bonds in a linear structure, and XRD analysis indicated a low degree of crystallinity of 0.11 (EPS-1) and 0.27 (EPS-2). EPS-1 and EPS-2 demonstrated high degradation temperatures of 319 degrees C and 314 degrees C, respectively, and non-cytotoxicity to HEK-293 cells at 2 and 3 mg/mL, respectively. In addition, both showed antioxidant activities. EPSs from strain WSUCF1 may expand the applications of microorganisms isolated from extreme environments and provide a valuable resource for exploitation in biomedical fields such as drug delivery carriers.
查看更多>>摘要:Bioeconomy has been proposed as a strategy to overcome many global and national challenges, from climate action to income diversification in rural areas. Therefore, the ultimate goal is not to measure the bioeconomy per se, but its sustainability. One way to ensure sustainability of bioeconomy strategies would be by linking its reporting with internationally-agreed targets of Sustainable Development Goals (SDGs) of the United Nations. Although there are case studies, scientific articles and technical reports in the literature of bioeconomy and other related strategies (e.g. green economy, circular economy) and their links to SDGs, the complete picture on this heterogeneous literature is missing. The objective of this work is to lay out the opportunities to couple monitoring and evaluation of bioeconomy and bioeconomy-related concepts to aid the countries to report their status on attaining SDGs. This study is the first to review and harmonize the results through meta-analysis of the available literature and technical reports on linking bioeconomy and SDGs. The results suggest that bioeconomy monitoring and evaluation can provide opportunities in terms of SDG reporting, in all three dimensions of sustainability. Bioeconomy strategies and practices were found to have strong potential in coupled reporting with several SDGs related to economic development, access to basic services, and sustainable consumption, followed by biodiversity conservation, waste re-use, equality, gender equality, inclusiveness and international cooperation.

查看更多>>摘要:Bioeconomy is becoming the main driver transforming European agri-food value chains towards global sustainability in the food supply chain. Intensive horticultural production systems based on medium and low-tech greenhouses are suitable scenarios implementing bioeconomy strategies to achieve sustainability targets. Since the publication of the European Strategy of Bioeconomy in 2012, policy measures intended to boost bioeconomy are responsible for changing what are now considered outdated production systems to more high-tech models capable of responding to climate-change challenges. This article describes the potential for the agri-food supply chain to drive the transition of medium and low-tech intensive greenhouse systems to biobased, circular economy value-chains. Key areas of impact relate to waste valorisation and management, new inputs based on biotechnological innovations, building clusters of innovative delivery partners within the sector, and the increase in public awareness of the impact of the bioeconomy through socio-economic analysis.

There is still no common international method for determining, measuring and comparing the extent of sustainability. The aim of this research is to develop a methodology for the assessment of bioeconomy-influencing factor interlinkages, and creation of benchmarks through a top-down approach. The main output is the assessment of factor interlinkages that could be further used for composite index creation. A case of triple factor nexus is presented: policy, research and innovations, and technology nexus for EU countries. As a result, the empirical model presents the mathematical description of policy, research and innovation, and technology link benchmark.
查看更多>>摘要:The parasitic fungus Claviceps purpurea has been used for decades by the pharmaceutical industry as a valuable producer of ergot alkaloids. As the biosynthetic pathway of ergot alkaloids involves a common precursor L-tryptophan, targeted genetic modification of the related genes may improve production yield. In this work, the S76L mutated version of the trpE gene encoding anthranilate synthase was constitutively overexpressed in the fungus with the aim of overcoming feedback inhibition of the native enzyme by an excess of tryptophan. In another approach, the dmaW gene encoding dimethylallyltryptophan synthase, which produces a key intermediate for the biosynthesis of ergot alkaloids, was also constitutively overexpressed. Each of the above manipulations led to a significant increase (up to 7-fold) in the production of ergot alkaloids in submerged cultures.
查看更多>>摘要:Bio-based solutions are expected to ensure technological circularity in priority areas such as agriculture, biotechnology, ecology, green industry or energy. Although Poland, unlike the other EU member states, has not yet adopted a precise political strategy to promote bioeconomy, it has taken several actions to enable smart, sustainable and inclusive growth. This goal can be achieved by developing selected bioeconomy-related areas such as the biogas industry together with novel technologies implemented to optimize treatment of municipal sewage and management of organic solid waste. Here, the relatively strong status of the Polish biogas sector is presented. The widely used practice of sewage sludge biomethanation has led to construction of numerous complex installations combining biological wastewater treatment plants with anaerobic digesters. Based on physico-chemical processing of biostabilized sludges, a novel method for efficient granulated soil fertilizer production is elaborated, in line with the concept of circular economy and the notion of "waste-to-product". It is also shown that anaerobic fermentation of sewage sludges can be optimized by co-digestion with properly selected co-substrates to increase bioprocess yield and improve the resultant digestate fertilizer quality. The problem of post-fermentation eutrophic sludge liquors, environmentally hazardous waste effluents requiring proper treatment prior to discharge or field application, is addressed. Attempts to optimize biological treatment of digestate liquors with complex microbial consortia are presented. The Polish innovations described show that the "zero waste" path in circular bioeconomy may bring advantageous results in terms of transformation of waste materials into commercial, added-value products together with recovery of water resources.
