    Dormancy of Amaranthus retroflexus L.Seeds and Physiological Response Seedlings to Acifluorfen Sodium

    查看更多>>摘要:Amaranthus retroflexus L.is a serious and widespread malignant weed in soybean fields in Heilongjiang Province.Exploring the dormancy characteristics of A.retroflexus L.seeds and the physiological response of its seedlings to acifluorfen sodium can provide a basis for further researches on its resistance mechanism.Using newly harvested and stored A.retroflexus L.seeds for one year as experimental materials,the effects of different concentrations of HCl,NaOH,water temperature,gibberellic acid(GA)and polyethylene glycol(PEG)on the dormancy and germination of A.retroflexus L.seeds were studied.The sensitivity of A.retroflexus L.to acifluorfen sodium was determined using bioassay.The effects on leaf chlorophyll content and target enzyme activity were studied at a normal dosage of 360 g a.i.hm-2 and a doubling dosage of 720 g a.i.hm-2 of acifluorfen sodium.Newly harvested seeds exhibiting dormancy were soaked in water of various temperatures and in different concentrations of NaOH and HCl,which were ineffective in breaking the seed dormancy.GA could break seed dormancy,and the highest seed germination rate reached 93.33%when they were soaked at 3 000 mg·L-1 for 72 h and 4 000 mg·L-1 for 48 h.The drought stress was simulated with a 15%-25%polyethylene glycol solution,which had no significant effect on the seed germination rate.The GR50 value of acifluorfen sodium for A.retroflexus L.was 705.7 g a.i.hm-2,which was 1.96 times the recommended dose in the field.After the application of different doses of acifluorfen sodium,the chlorophyll content of A.retroflexus L.reached its minimum value 3 days after treatment(DAT),and then gradually increased.The activity of the target enzyme protoporphyrinogen oxidase(PPO)reached the highest value at 7 DAT under different dosages,and gradually returned to normal levels thereafter.Soaking with gibberellin was an effective method to break seed dormancy.A.retroflexus L.seeds had certain drought resistance during the germination process.A.retroflexus L.was not sensitive to acifluorfen sodium and acifluorfen sodium ether,and could not effectively inhibit the PPO activity,indicating that A.retroflexus L.had target resistance to acifluorfen sodium.

    查看更多>>摘要:Soybean frogeye leaf spot(FLS)disease is a global disease affecting soybean yield,especially in the soybean growing area of Heilongjiang Province.In order to realize genomic selection breeding for FLS resistance of soybean,least absolute shrinkage and selection operator(LASSO)regression and stepwise regression were combined,and a genomic selection model was established for 40 002 SNP markers covering soybean genome and relative lesion area of soybean FLS.As a result,68 molecular markers controlling soybean FLS were detected accurately,and the phenotypic contribution rate of these markers reached 82.45%.In this study,a model was established,which could be used directly to evaluate the resistance of soybean FLS and to select excellent offspring.This research method could also provide ideas and methods for other plants to breeding in disease resistance.

    查看更多>>摘要:Salt stress is a major abiotic stress limiting plant growth and yield.In the present study,the effects of exogenous H2O2 on the reactive oxygen species(ROS)metabolism and the antioxidant system in leaves of Nitralia tangutorum Bobr.under salt stress were studied.N.tangutorum seedlings were subjected to 200 mmol·L-1 NaCl treatment with or without the exogenous application of H2O2 for 7 days.The results showed that NaCl stress significantly increased the relative conductivity,the contents of thiobarbituric acid reactive substances(TBARS)and ROS(H2O2 and O2·-),as well as promoted the activities of antioxidant enzymes including superoxide dismutase(SOD),peroxidase(POD),catalase(CAT),and ascorbate peroxidase(APX)in N.tangutorum leaves.In addition,exogenous H2O2 decreased the relative conductivity,the contents of TBARS,H2O2 and O2·-,while further enhanced the activities of antioxidant enzymes.These results indicated that H2O2 effectively alleviated the adverse effects of NaCl stress on N.tangutorum through the regulation of ROS metabolism.

    Key Physical Factors Affecting Spatial-temporal Variation of Labile Organic Carbon Fractions by Biochar Driven in Mollisols Region of Northeast China

    查看更多>>摘要:Biochar is widely used to improve soil physical properties and carbon sequestration.However,few studies focuse on the impact of maize stalk biochar on labile organic carbon(LOC)pool and the relationship between physical properties and LOC fractions.A field positioning experiment was performed in Mollisols region of Northeast China to evaluate the influence of maize stalk biochar on the spatial distribution and temporal changes of physical properties and LOC fractions.Maize stalk biochar treatments included C1(1.5 kg·hm-2),C2(3 kg·hm-2),C3(15 kg·hm-2),C4(30 kg·hm-2),and CK(0).The results showed that maize stalk biochar increased soil water contents(SWC)and soil porosity(SP),but reduced bulk density(BD).Maize stalk biochar reduced dissolved organic carbon(DOC)contents in the 0-20 cm soil layer,ranging from 0.25 g·kg-1 to 0.31 g·kg-1 in harvest period,while increased in the 20-40 cm soil layer.In addition,the application of biochar had a significant impact on the spatial distribution and temporal change of SWC,BD,SP,DOC,hot-water extractable carbon(HWC),acid hydrolyzed organic carbon(AHC I,II),and readily oxidized organic carbon(ROC).High amounts of maize stalk biochar up-regulated the contents of soil organic carbon SOC,HWC,AHC I,AHC II,and ROC.In addition,SWC and SP were the key physical factors to affect LOC fractions.In conclusions,maize stalk biochar could improve physical properties,and then influence LOC fractions,and maize stalk biochar could be used as an organic amendment for restoring degraded soils governed by their rates of addition.

    Differential Expression Analysis of Proteins Regulated by Rare Earth Cerium in Soybean Leaves at Seedling Stage

    查看更多>>摘要:This study was to explore the functional mechanism of rare earth regulating soybean leaves and the characteristics and functions of differentially expressed proteins under the regulation of rare earth.In this study,Dongnong 42 was used as material,and 30 mg·L-1 CeCl3 solution was sprayed on soybean leaves at the seedling stage.Tandem mass tag(TMT)quantitative proteomics technique and bioinformatics analysis were used to identify soybean leaf proteins.A total of 8 510 proteins were identified,and 127 differentially expressed proteins(DEPs)in response to rare earth cerium regulation were identified,among which 64 were up-regulated and 63 were down-regulated.The gene ontology(GO)annotation indicated that DEPs were mainly involved in metabolic process,cellular process,response to stimulus,biological regulation,and response to a stimulus;DEPs in cell module categories were mainly involved in cells,cell part,organelle,membrane,membrane part,organelle par,and protein-containing complex;DEPs in molecular functional categories were mainly involved in catalytic activity,binding and antioxidant activity.Kyoto encyclopedia of genes and genomes(KEGG)pathway significantly enriched starch and sucrose metabolism,glycolysis/gluconeogenesis,galactose metabolism,pentose phosphate pathway,and MAPK signaling pathway-plant.These DEPs were mainly involved in photosynthesis,glucose metabolism and stress response.Forty-six differential protein interaction networks were identified by protein interaction network analysis.This experiment provided a reference for studies of the mechanism of rare earth cerium regulating soybean leaf function from the proteomic perspective.

    查看更多>>摘要:In order to optimize the ultrasonic extraction technique for the total flavonoid of leaf yellows plus,the contents of 21 leaf yellows plus total flavonoid from four regions in Heilongjiang Province were comparatively analyzed.The ultrasonic extraction technology was optimized by Box-Behnken response surface method,and the total flavonoid content of 21 kinds of Acanthopanax senticosus(Rupr.et Maxim.)Harms from different producing areas were analyzed by cluster analysis.The optimal process conditions were determined as ultrasonic time 30 min,solid-liquid ratio 1:12 and ultrasonic power 250 W,and the average yield of the total flavonoid was 1.453 mg·g-1.By optimizing the ultrasonic-assisted extraction method,the total flavonoid content from different producing areas was compared in the experiment,which provided certain data support for the optimization of the extraction process in the future and laid a certain theoretical foundation for the quality analysis of Chinese medicinal materials.

    Cookie Baking Process Optimization and Quality Analysis Based on Food 3D Printing

    查看更多>>摘要:In order to obtain better quality cookies,food 3D printing technology was employed to prepare cookies.The texture,color,deformation,moisture content,and temperature of the cookie as evaluation indicators,the influences of baking process parameters,such as baking time,surface heating temperature and bottom heating temperature,on the quality of the cookie were studied to optimize the baking process parameters.The results showed that the baking process parameters had obvious effects on the texture,color,deformation,moisture content,and temperature of the cookie.All of the roasting surface heating temperature,bottom heating temperature and baking time had positive influences on the hardness,crunchiness,crispiness,and the total color difference(ΔE)of the cookie.When the heating temperatures of the surfac and bottom increased,the diameter and thickness deformation rate of the cookie increased.However,with the extension of baking time,the diameter and thickness deformation rate of the cookie first increased and then decreased.With the surface heating temperature of 180℃,the bottom heating temperature of 150℃,and baking time of 15 min,the cookie was crisp and moderate with moderate deformation and uniform color.There was no burnt phenomenon with the desired quality.Research results provided a theoretical basis for cookie manufactory based on food 3D printing technology.

    查看更多>>摘要:Soybean is one of the important crops in China.Soymilk,a traditional neutral plant-based protein drink,is rich in high quality proteins.Although soybean milk is rich in nutrients,its marketing among consumers,especially those in Western countries who are used to peaceful flavor,has been limited due to the adverse flavor impact brought by its special composition.In recent years,with the increasing attention to the nutritional value of soymilk,the flavor of soymilk has become a popular research object for scholars at home and abroad.The flavor components of soymilk are mainly volatile small molecular compounds produced by enzymatic reactions catalyzed by lipoxygenase(LOX).After formation,they interact with protein macromolecules to form the overall flavor of soymilk.At present,there are many methods to control the off-odor of soymilk at home and abroad,including physical heating methods,chemical methods,biological enzymatic digestion methods,mask methods,and a variety of breeding methods.These methods effectively reduce the off-odor of soymilk,but all of them have shortcomings.Currently,the sensory characteristics of the beany odor in soymilk are evaluated mainly by traditional human sensory scoring along with the assistance of modern instrument analysis of volatile flavor substances using headspace solid phase microextraction(SPME)gas chromatography coupled with-mass spectrometry(GC-MS).This paper summarized the research results of volatile flavor substances in soymilk in recent years and the sensory evaluation methods of soymilk at home and abroad,and looked forward to the future development direction,hoping to provide some theoretical bases and reference detection methods for solving the problem of soymilk flavor in the future.
