    Identification of Active Components in Humic Acids Affecting Tomato Germination

    查看更多>>摘要:Humic acids can promote the germination of many vegetable seeds,but the key active components remain unclear.This study utilized nutrient content,cross polarization magic angle spin 13C solid magnetic resonance(CPMAS-13C-NMR)and ultra-high performance liquid chromatography-mass spectrometry(UHPLC-MS)to characterize the chemical components of humic acids.Tomato seed germination index(GI)was determined with the goal of screening the key active components of humic acids.Humic acids had a significantly higher nutrient content,except for the total nitrogen(TN)and the total phosphorus(TP)contents.Humic acids had a higher content of O-CH3/NCH,aromatic C-O and carbonyl C compared to weathered coal,with significantly lower anomeric C,aromatic C and O-alkyl C/alkyl C.There were 611 different compounds identified among the test materials using UHPLC-MS.Humic acids also had a significantly higher GI(158.0%and 153.1%)than weathered coal(85.5%).The organic matter(OM),TP and available potassium(AK)contents in humic acids were significantly positively correlated with GI,and available phosphorus(AP)was significantly negatively correlated.Among the carbon components,O-CH3/NCH,aromatic C-O and O-alkyl C/alkyl C were significantly positively correlated with GI,while anomeric C was significantly negatively correlated.Furthermore,among the top 10 positive and five negative correlation compounds,lipids and lipid-like molecules[armexifolin,boviquinone 4,3-methyladipic acid,lxocarpalactone A,monic acid,DG(20:1(11Z)/18:4(6Z,9Z,12Z,15Z)/0:0),and brassinolide]and organic acids and derivatives(N-acetylglutamic acid,8-hydroxy-5,6-octadienoic acid,acetyl-L-tyrosine,and hydroxyprolyl-methionine)in humic acids might be crucial active components for improving tomato seed germination.The results provided direct evidence for the identification of bioactive molecules of humic acids,and a scientific basis for the precise utilization of bioactive molecular components of humic acids in sustainable agricultural development.

    查看更多>>摘要:At the early stage,the transcriptome sequencing technique was used to detect the differentially expressed gene CsFK111 between vine cucumber and dwarf cucumber D0462.The gene was cloned,and bioinformatics software tools were used to analyze and predict the gene family and this gene.There were 30 members of the cucumber F-box gene family.The coding region of the cucumber CsFK111 gene was full-length 1 314 bp,which encoded 437 amino acids and was predicted to be located in the nucleus.The protein encoded by this gene was a non-transmembrane protein,and the prediction of the secondary structure showed that β-lamellar structure and irregular crimp were dominant.A comparison of the phylogenetic tree showed that it was closest to cantaloupe and belonged to the same branch.The results provided a basis for future study on the regulation mechanism of the CsFK111 gene on cucumber dwarfing and also laid a foundation for further study of FBK family proteins.

    查看更多>>摘要:As an important wild blueberry resource,Vaccinium uliginosum has attracted more and more attention.At present,the wild resources are under destruction.The conservation of wild Vaccinium uliginosum resources is imminent.However,there are few researches on the protection and preservation of its germplasm resources.In vitro preservation is an important method for germplasm conservation.In this study,one strain of wild Vaccinium uliginosum was used as material.The effects of temperature(25 ℃,15 ℃,10 ℃,or 0 ℃),media(WPM,1/2WPM or 1/3WPM),medium supplements(sorbitol or mannose),and photoperiod(8,10,12,or 14 h·d-1)on the growth,survival rate and rejuvenation rate of the plantlets were studied.The physiological changes of plantlets during preservation were analyzed.Methylation-sensitive amplified polymorphism(MSAP)analysis of genomic DNA methylation of plantlets was carried out to explore the genetic stability of the plantlets after preservation.The research results provided a theoretical basis for the germplasm preservation of Vaccinium uliginosum.

    Effects of Biochar-based Organic Fertilizer on Accumulation of Total Flavonoid Contents in Dandelion

    查看更多>>摘要:In order to study the effects of biochar-based organic fertilizer on the accumulation of the total flavonoid contents in dandelion after sowing,the experiment applied four levels of treatments of carbon-based organic fertilizer(0.15,0.30,0.45,and 0.60 kg·m-2),and the base fertilizer that only applied conventional chemical fertilizer was used as the control.The total flavonoid contents and physiological indices of dandelion in different growth periods were determined.The results showed that the accumulation of the total flavonoid contents in leaves and roots of dandelion improved significantly by applying biochar-based organic fertilizer(0.15,0.30,0.45,and 0.60 kg·m-2),with the highest total flavonoid occurred in the 0.45 kg·m-2 biochar-based organic fertilizer treatment.Along with dandelion growing,the total flavonoid contents in leaves were first elevated,and then decreased,with the maximum reached at 95 days after sowing(anthesis).While the total flavonoid contents in roots were elevated constantly.The total flavonoid contents accumulation in dandelion were positively correlated with net photosynthetic rate,stomatal conductance,transpiration rate,soluble sugar content,soluble protein,nitrate nitrogen,and available potassium,while negatively correlated with intercellular CO,concentration.

    查看更多>>摘要:The rice Dongfu 159 was used for the potting experiment to simulate soil cadmium(Cd)stress,and four silicon preparations,Si-50-G and Si-60-G at a concentration of 0.20%and Si-T-G and Si-E-G at a concentration of 0.50%,were applied at the tillering stage and booting stage of rice,respectively.The cadmium content,root vitality,root malondialdehyde(MDA)content,and antioxidant enzyme activity of each part of rice plants were determined,in order to provide a theoretical reference for exploring the mechanism of silicon fertilizer on the mitigation of Cd-stressed plants.The results showed that under the condition of cadmium stress,silicon application could significantly increase the root vitality and antioxidant enzyme activity,reduce the content of MDA,and reduce the accumulation of cadmium in various parts of rice.The treatment of Si-T-G applied at the tillering stage was the most effective in increasing the root vitality of rice,which was significantly increased by 63.00%compared with the CK.The treatment of Si-50-G applied at the stage of booting was the most effective in reducing MDA content of rice roots,which was significantly reduced by 30.16%compared with the CK.The treatment of Si-60-G applied at the stage of booting was the most effective in increasing the root superoxide dismutase(SOD)activity,which was significantly increased by 39.36%compared with the CK.The treatment of Si-60-G applied at the tillering stage was the most effective in increasing the root peroxidase(POD)and catalase(CAT)activities,which were significantly increased by 64.66%and 51.52%,respectively,compared with the CK.The treatment of Si-T-G applied at the tillering stage was the most effective in reducing Cd content of rice roots,stems and grains,which were significantly reduced by 39.53%,61.19%and 43.41%,respectively,compared with the CK,and the treatment of Si-60-G was the most effective in reducing Cd content of leaves,which was significantly reduced by 53.40%compared with the CK.

    查看更多>>摘要:Avian influenza virus is one of the main pathogens causing avian influenza.In this experiment,the avian influenza hemagglutinin(HA)gene was transferred into tobacco by Agrobacterium tumefaciens-mediated method using kanamycin as a resistance marker to produce HA type edible vaccine designed for avian influenza hemagglutinin protein.The fusion of cholera toxin B subgene(CTB)and avian influenza HA was initiated by CaMV35S promoter,and then transformed into Nicotiana benthamiana to induce callus and adventitious bud differentiation,bud elongation,and growth and root induction.The total genomic DNA of 28 transgenic tobacco plants was extracted,and the CTB sequence of the CTB-HA fusion gene was used as a template to design primers for PCR amplification.Eight of them were positive,and four of them were expressed at the RNA level after RNA extraction and RT-PCR amplification.In western blot detection of protein extraction,two strains were shown at the position of the target protein.The results provided material support for the preparation of a transgenic plant oral vaccine against HA infection,and provided a theoretical basis for accelerating the development of a transgenic plant vaccine.

    查看更多>>摘要:This study aimed to evaluate the effects of dietary yeast culture(YC)on lipopolysaccharide(LPS)-induced oxidative stress,immune and inflammatory response in P.ussuriensis.The fish were randomly assigned into three groups as the control group,LPS group and YC+LPS group.The fish in the control were fed diet with no YC supplementation and no LPS challenge,and the fish in the LPS group or YC+LPS group were fed diet supplemented with no YC or 20 g·kg-1 YC,and with LPS challenge,respectively.The results showed that compared with the control group,intestinal total antioxidant capacity(T-AOC)level and superoxide dismutase(SOD)activity were significantly decreased,while intestinal malondialdehyde(MDA),plasma aspartate transaminase(AST)and alanine transaminase(ALT)levels were significantly increased in the LPS group(P<0.05).Besides,lower plasma alkaline phosphatase(ALP),alternative complement pathway(ACH50)activity and the albumin(ALB)level,as well as higher lysozyme(LZM)activity,were also found in the LPS group.However,dietary 20 g·kg-1 YC supplementation could relieve the above LPS-induced changes in Pseudobagrus ussuriensis.Furtherly,LPS challenge could significantly up-regulate gene expression of interleukin-8(IL-8),heat shock protein(HSP70)and NF-κBp65 except for toll-like receptors 2(TLR2),while dietary 20 g·kg-1 YC supplementation suppressed the increased expression of NF-κBp65 and IL-8 induced by LPS in P.ussuriensis.In summary,LPS challenge could induce immune impairment,oxidative stress and hepatic damage,and the protective effect of dietary 20 g·kg-1 YC supplementation on LPS-induced immune impairment and oxidative stress was observed in the present study,which was associated with the enhanced levels of antioxidant enzymes and immune parameters.Also,dietary 20 g·kg-1 YC supplementation could suppress LPS-induced inflammatory response by down-regulating NF-κBp65,HSP70 and IL-8 gene expression.

    Basic Terms in Shiology—Revealing the Connotation and Extension of the Core Concept of Shiance Cognition

    查看更多>>摘要:Shiology,abbreviated from the study of shiance,stands as a"disciplinary category"level knowledge system.Within the Chinese educational system's subject catalog,it is an extension of agricultural science(09),and it is a knowledge system that integrates the eater needs,food acquisition and shiance order.Cognition of previously unrecognized objects requires new concepts,and the construction of a new knowledge system cannot be separated from the support of new concepts.Shiance knowledge system is a huge collection of shiance concepts,and this research limits its discussion to the connotations and extensions of four core groups of concepts:shiance,food,eaters,and eating.These core concepts of shiance knowledge also serve as the foundational terms for constructing the shiology framework.Establishing the foundational terms and definitions of shiology is a fundamental research endeavor.If one were to liken the shiology framework to a building,then its foundational terms are akin to the"bricks";defining these terms is comparable to setting standards for these"bricks",facilitating the construction of a more robust structure.The basic words discussed in this research include 34 in the category of"shiance",31 in the category of"food",seven in the category of"eaters"and 16 in the category of"eating".These basic words play a fundamental and leading role in the process of constructing the system of shiology system,and they are used in the names of 40 subjects in the 53 basic shiology systems.The absence and narrowing of cognitive concepts are at the root of the lack of systematic governance of human shiance issues.
