    银杏内酯B对小鼠缺血性脑卒中后神经功能恢复及Wnt/β-catenin通路的影响

    查看更多>>摘要:目的 探究银杏内酯B对小鼠缺血性脑卒中后神经功能恢复及Wnt/β-连环蛋白(β-catenin)通路的影响。 方法 选取C57/BL6系实验小鼠55只,留取10只作为假手术组,其余45只小鼠构建缺血性脑卒中模型,最终有40只完成建模,将其随机分为模型组、短程给药(GB1w)组、长程给药(GB2w)组、长程给药+拮抗剂(GB2w+PRI-724)组,每组各10只。模型组大脑中动脉栓塞(MCAO)术后无药物干预;GB1w组MCAO术后1周内鼻饲银杏内酯B(10 mg/kg)0.1 ml;GB2w组MCAO术后2周内鼻饲银杏内酯B(10 mg/kg)0.1 ml;GB2w+PRI-724组MCAO术后2周内鼻饲银杏内酯B(10 mg/kg)0.1 ml,且第8~14天银杏内酯B给药前3 h,给予选择性拮抗剂PRI-724 20 mg/d。比较各组的神经功能评分、转棒上行走实验评分、转化生长因子-β1(TGF-β1)、成纤维细胞生长因子4(FGF4)、肿瘤坏死因子-α(TNF-α)、白细胞介素-6(IL-6)、丙二醛(MDA)、谷胱甘肽过氧化物酶(GSH-Px)、超氧化物歧化酶(SOD)、Wnt、β-catenin和糖原合成酶激酶-3β(GSK-3β)表达的变化情况。 结果 与假手术组相比,模型组、GB1w组、GB2w组和GB2w+PRI-724组的神经功能评分、转棒上行走实验评分、FGF4、TNF-α、IL-6和MDA水平及GSK-3β表达上升,TGF-β1、GSH-Px和SOD水平及Wnt、β-catenin表达下降,差异均有统计学意义(均P<0.001);与模型组相比,GB1w组、GB2w组和GB2w+PRI-724组的TGF-β1、GSH-Px和SOD水平及Wnt、β-catenin表达上升,神经功能评分、转棒上行走实验评分、FGF4、TNF-α、IL-6和MDA水平及GSK-3β表达下降,差异均有统计学意义(均P<0.001);与GB1w组相比,GB2w组和GB2w+PRI-724组的TGF-β1、GSH-Px和SOD水平及Wnt、β-catenin表达上升,神经功能评分、转棒上行走实验评分、FGF4、TNF-α、IL-6和MDA水平及GSK-3β表达下降,差异均有统计学意义(均P<0.001);与GB2w组相比,GB2w+PRI-724组的神经功能评分、转棒上行走实验评分、FGF4、TNF-α、IL-6和MDA水平及GSK-3β表达上升,TGF-β1、GSH-Px和SOD水平及Wnt、β-catenin表达下降,差异均有统计学意义(均P<0.001)。 结论 银杏内酯B可有效改善缺血性脑卒中小鼠神经功能,且可能与Wnt/β-catenin通路相关。 Objective To investigate the effects of ginkgolide B on neurological function recovery and the Wnt/β-catenin pathway after ischemic stroke in mice. Methods Fifty-five C57/BL6 mice were selected, of which 10 mice were kept as the sham group and the remaining 45 mice were constructed as the ischemic stroke model. There were 40 mice who finally completed the modeling, and then they were randomly divided into the blank control group (GB0w), short-course administration group (GB1w), long-term administration group (GB2w), and long-term administration+antagonist group (GB2w+PRI-724), with 10 mice in each group. There was no drug intervention after MCAO in GB0w. The mice in GB1w were given ginkgolide B (10 mg/kg) 0.1 ml within 1 week after MCAO in GB2w were given ginkgolide B (10 mg/kg) 0.1 ml within 2 weeks after MCAO and in GB2w+PRI-724 were nasally fed ginkgolide B (10 mg/kg) 0.1 ml within 2 weeks after MCAO and selective antagonist PRI-724 was given 3 h before administration of ginkgolide B on days 8 to 14. Neurological function scores, walking on rotor bar test scores, expression of transforming growth factor-β1 (TGF-β1), fibroblast growth factor 4 (FGF4), tumor necrosis factor-α (TNF-α), interleukin-6 (IL-6), malondialdehyde (MDA), glutathione peroxidase (GSH-Px), superoxide dismutase (SOD), Wnt, β-catenin, and glycogen synthase kinase-3β (GSK-3β) were compared among the groups. Results Compared with the sham group, the expressions of MDA, TNF-α, IL-6, FGF4, and GSK-3β in GB0w, GB1w, GB2w, and GB2w+ PRI-724 were increased, and the expressions of GSH-Px, SOD, TGF-β1, β-catenin, and Wnt were decreased (allP < 0.001). Compared with GB0w, the expressions of SOD, GSH-Px, TGF-β1, Wnt, and β-catenin were increased in GB1w, GB2w, and GB2w+PRI-724, and the expressions of MDA, TNF-α, IL-6, FGF4, and GSK-3β were decreased (all P < 0.001). Compared with GB1w, the expressions of GSH-Px, SOD, TGF-β 1, Wnt, and β-catenin were increased in GB2w and GB2w+PRI-724, and the expressions of IL-6, TNF-α, MDA, FGF4, and GSK-3β were decreased (all P < 0.001). Compared with GB2w, the neural function score, walking on the stick test score, and expressions of IL-6, TNF-α, FGF4, MDA, and GSK-3β were increased in GB2w+PRI-724, while the expressions of GSH-Px, TGF-β1, SOD, Wnt, and β-catenin were decreased (all P < 0.001). Conclusions Ginkgolide B can effectively improve the neurological function of ischemic stroke mice and may be related to the Wnt/β-catenin pathway.

    查看更多>>摘要:目的 研究改性柑橘果胶(MCP)对兔关节软骨细胞的糖酵解代谢的影响。 方法 分离、培养兔膝关节软骨细胞至第3代后,分别用含0、500 μg/ml MCP的培养液(MCP0和MCP500)培养3 d,设MCP0为对照组;连续传代培养软骨细胞,每代软骨细胞分别以MCP0和MCP500培养3 d;以白细胞介素-1β(IL-1β)处理软骨细胞1 d后,再分别用MCP0和MCP500培养3 d。以2-脱氧-葡萄糖(2DG)处理软骨细胞1 d后,再分别用MCP0和MCP500培养3 d;于培养3 d后,通过CCK-8方法检测软骨细胞的相对增殖情况;利用葡萄糖和乳酸检测试剂盒测定软骨细胞的葡萄糖摄取量和乳酸生成量;通过免疫荧光染色方法考察连续传代软骨细胞的Ⅱ型胶原α1(COL2A1)合成情况;采用实时荧光定量PCR(RT-qPCR)检测软骨细胞的COL2A1、蛋白聚糖(ACAN)、性别决定区Y框蛋白9(SOX9)、缺氧诱导因子-1α(HIF-1α)、葡萄糖转运蛋白-1(Glut-1)、丙酮酸激酶M2(PKM2)、乳酸脱氢酶A(LDHA)和葡萄糖转运蛋白-3(Glut-3)的基因表达情况。 结果 与对照组相比,MCP处理提高了软骨细胞的葡萄糖摄取量和乳酸生成量,上调了ACAN、HIF-1α、Glut-1和PKM2的基因表达水平;与对照组相比,连续传代过程中,MCP可以促进软骨细胞的增殖、维持软骨细胞的表型,上调COL2A1、ACAN、SOX9、HIF-1α、Glut-1、PKM2和LDHA的基因表达,提高软骨细胞COL2A1合成量和乳酸生成量;在经IL-β处理后,与对照组相比,MCP处理提高了软骨细胞的葡萄糖摄取量,上调了COL2A1、ACAN、HIF-1α和Glut-1的基因表达。在经2DG处理后,MCP处理提高了软骨细胞的葡萄糖摄取量,上调SOX9、HIF-1α、PKM2和Glut-3的基因表达。 结论 MCP能够增强软骨细胞的葡萄糖摄取能力,提高软骨细胞糖酵解代谢水平。 Objective To investigate the effect of modified citrus pectin (MCP) on the glucose metabolism of rabbit articular chondrocytes. Methods The third generation (P3) rabbit knee chondrocytes were extracted and cultured with 0 μg/ml (MCP0, control group) and 500 μg/ml of MCP (MCP500) for 3 days. Chondrocytes (P2-P7)were cultured continuously, and each generation of chondrocytes was treated with MCP0 and MCP500 medium for 3 days. Chondrocytes were treated with interleukin-1β (IL-1β) for 1 day and then treated with MCP0 and MCP500 medium for 3 days, respectively. Chondrocytes were treated with 2-deoxy-glucose (2DG) for 1 day and then treated with MCP0 and MCP500 medium for 3 days, respectively. After three days of culture, the proliferation of chondrocytes was calculated by CCK-8. Glucose uptake activity and lactate production of chondrocytes were measured by glucose and lactate detection kits. The synthesis of type Ⅱ collagen (COL2A1) in sequential chondrocytes was investigated by immunofluorescence staining. The gene expression of COL2A1, proteoglycan (ACAN), SOX9, hypoxia-inducible factor-1α (HIF-1α), glucose transporter-1 (Glut-1), pyruvate kinase M2 (PKM2), lactate dehydrogenase-A (LDHA) and glucose transporter-1 (Glut-3) were further detected by RT-qPCR. Results Compared with the control group, MCP treatment could increase the glucose uptake activity and lactate production of chondrocytes, and enhance the gene expression ability of HIF-1α, Glut-1, PKM2 and ACAN. Besides, MCP treatment could stimulate chondrocyte proliferation, maintain chondrocyte phenotype, increase lactate production, and upregulate the expression of COL2A1, ACAN, SOX9, HIF-1α, Glut-1, PKM2 and LDHA. After the treatment with IL-1β, MCP treatment could increase glucose uptake activity and upregulate the expression ofCOL2A1, ACAN, HIF-1α and Glut-1. After treatment with 2DG, MCP treatment could increase glucose uptake activity and upregulate the expression of SOX9, HIF-1α, PKM2 and Glut-3 genes. Conclusions MCP can enhance the glucose uptake capacity of chondrocytes and increase the level of chondrocyte glycolytic metabolism.

    基于共同空间模式与迁移学习的脑电识别研究

    查看更多>>摘要:目的 针对目标用户的脑电识别问题,提出了基于共同空间模式(CSP)与迁移学习的脑电识别方法。 方法 首先对脑电数据进行预处理,选择事件相关去同步(ERD)现象存在明显差异的时间窗0.5~2.5 s及包含α和β波的宽频带8~30 Hz脑电,然后对多用户利用CSP分别提取特征得到特征向量,最后利用迁移学习进行目标用户的脑电识别。 结果 用户aa在Cz通道处想象右手的ERD高于想象双脚的ERD。用户aa、al、av、aw、ay的分类正确率分别为93.8%、100.0%、84.2%、94.6%、94.4%,平均分类正确率为92.4%,优于常用分类器SVM和期望最大化EM,仅比竞赛第一名清华大学所采用的方法低1.8%。 结论 基于CSP与迁移学习的脑电识别方法能够利用非目标用户提高目标用户的脑电识别性能,对运动想象脑-机接口系统的研究具有重要意义。 Objective Aiming at the problem of target user electroencephalogram (EEG) recognition, an EEG recognition method was presented based on common spatial patterns (CSP) and transfer learning. Methods Firstly, preprocess was adopted on the original EEG data, and time windows 0.5~2.5 s and broad frequency band 8~30 Hz EEG signals, which contained α and β wave, were selected. Here event-related desynchronization (ERD) phenomenon existed significant differences. Afterwards, by CSP preprocessed EEG signals of multi-user were conducted to extract feature and feature vectors were obtained, respectively. Finally, by transfer learning target user EEG recognition was completed. Results In channel Cz, ERD of right hand motor imagery was higher than ERD of foot motor imagery. The classification accuracy of users aa, al, av, aw, and ay were 93.8%, 100.0%, 84.2%, 94.6%, and 94.4%, respectively. The average classification accuracy was 92.4%, which was better than the commonly used classifiers SVM and EM. The method was only lower than the method of the first winner in the competition adopted by Tsinghua University 1.8%. Conclusions EEG recognition method based on CSP and transfer learning increased target user EEG recognition performance by using non-target users and had important implications for the study of motor imagery brain-computer interface.

In the digital era, the amount of data in clinical practice is increasing, which requires healthcare workers to integrate and interpret the various information generated during clinical work. With the help of artificial intelligence techniques, especially machine learning techniques, researchers in cardiovascular medicine have developed a variety of predictive models to improve the efficiency of clinical work and treatment outcomes. The types of machine learning models were introduced, and the current prediction models of cardiovascular diseases using machine learning technology were summarized. The purpose of this paper is to facilitate accurate diagnosis of cardiovascular diseases and to provide a clearer direction for future development of cardiovascular disease prediction models using machine learning techniques.

    查看更多>>摘要:"健康中国2030"战略背景下,跑步成为了最常见的运动方式。健身跑是一种耐力有氧运动,与游泳、健身操等其他活动度高、强度大的运动相比,下肢的过劳损伤风险较高。介绍了跑步相关损伤的危险因素,分析了跑步对关节的影响,并发现髋内收、屈膝和踝关节异常是跑步时造成主要损伤的3种关节相关因素,同时总结出4种康复干预手段,患者可以通过训练生成个性化的康复治疗方案。 Under the backdrop of the "Healthy China 2030" strategy, running has become the most common form of exercise. Fitness running is a kind of endurance aerobic exercise. Compared with swimming, aerobics, and other sports with high activity and intensity, the risk of lower limb fatigue injury during fitness running is higher. In this review paper, the risk factors for running-related injuries were summarized by consulting and analyzing the database and focuses on discussing and analyzing the impact of running on joints. The results showed that hip adduction, knee bending, and ankle joint abnormalities are the three joint-related factors that cause the main injuries during running. Four rehabilitation intervention methods for running-related injuries were proposed, that can guide patients to generate personalized rehabilitation treatment plans through training.
