    基于高通量测序探讨针刺结合补阳还五汤对颈动脉狭窄脑血流低灌注模型大鼠肠道菌群的影响

    查看更多>>摘要:目的 探究针刺结合补阳还五汤对颈动脉狭窄脑血流低灌注模型大鼠肠道菌群的影响。 方法 将40只大鼠按随机数字表法分为假手术组、模型组、中药组和针药组,每组10只。除假手术组外,其余各组采用针控线栓法制备脑缺血模型。中药组灌胃补阳还五汤化裁方水煎液100 mg/kg,1次/d,连续干预2周。针药组在中药组治疗基础上,选取“百会”及其左、右侧各旁开2 mm处进行干预,1次/d,连续干预2周。分别于给药后7、14 d进行神经功能评价和行为学功能评分。采用16S rRNA测序对各组大鼠粪便菌群结构和组成进行全面表征分析,采用线性判别效应量分析法(LEfSe)对多组间肠道细菌进行差异分析。 结果 给药后第7、14天,与模型组比较,中药组和针药组神经功能评分降低(P<0.05),行为学功能评分升高(P<0.05)。与模型组比较,中药组和针药组Shannon指数升高(P<0.05);厚壁菌门丰度升高(P<0.05),拟杆菌门和变形菌门丰度降低(P<0.05);梭菌纲丰度升高(P<0.05),丙型变形菌纲丰度降低(P<0.05);大肠杆菌-埃希氏菌和拟杆菌丰度降低(P<0.05);针药组乳酸杆菌丰度显著升高(P<0.05)。 结论 针刺结合补阳还五汤治疗对颈动脉狭窄脑血流低灌注模型大鼠的症状有改善作用,其机制可能与增加益生菌丰度有关。 Objective To explore the effects of acupuncture combined with Buyang Huanwu Decoction on intestinal flora in cerebral blood flow hypo perfusion model rats with carotid artery stenosis. Methods Totally 40 rats were randomly divided into sham-operation group, model group, TCM treatment group and acupuncture and drug combination treatment group, with 10 rats in each group. Except the sham-operation group, the other groups were prepared cerebral ischemia model by needle control and thread embolism method. TCM treatment group received Buyang Huanwu Decoction 100 mg/kg for gavage, once a day, and the intervention lasted for 2 weeks. In the acupuncture and drug combination group, based on the TCM treatment group, Baihui and its left and right sides of 2 mm were selected for acupuncture, once a day, and continuous intervention was performed for 2 weeks. Neurological function evaluation and behavioral function score were performed 7 and 14 days after administration, respectively. 16S rRNA sequencing was used to comprehensively characterize the structure and composition of fecal microflora of rats in each group. Linear discriminant analysis Effect Size (LEfSe) was used to analyze the difference of intestinal bacteria among groups. Result On the 7th and 14th day after administration, compared with the model group, the neurological function score in the TCM treatment group and the acupuncture and drug combination group decreased (P<0.05), and the behavioral function score increased (P<0.05). Compared with model group, the Shannon index of TCM treatment group and acupuncture and drug combination group increased (P<0.05). The abundance of Firmicutes increased (P<0.05), and the abundance of Bacteroidetes and Proteobacteria decreased (P<0.05) the abundance of Clostridia increased (P<0.05), and the abundance of Gammaproteobacteria decreased (P<0.05). The abundance of Escherichia-Shigella and Bacteroides decreased (P<0.05) the abundance of lactobacillus significantly increased (P<0.05). Conclusion Acupuncture combined with Buyang Huanwu Decoction can improve the symptoms of cerebral hypoperfusion model rats with carotid artery stenosis, and the mechanism may be to increase the abundance of probiotics.

    查看更多>>摘要:目的 借助网络药理学方法并结合动物实验预测鼻敏康颗粒治疗变应性鼻炎的分子机制。 方法 借助TCMSP、OMIM、GeneCards、TTD、DrugBank、PharmGKB数据库筛选鼻敏康颗粒的活性成分靶点及变应性鼻炎靶点;运用R语言软件对药物和疾病靶点映射取交集;采用Cytoscape 3.8.0软件、STRING数据库构建交集靶点PPI网络,并进行网络拓扑学分析;利用DAVID数据库进行GO功能富集和KEGG通路富集分析,并对主要活性成分与关键靶点进行分子对接验证。将32只大鼠按随机数字表法分为空白组8只和造模组24只。造模组大鼠经卵清蛋白诱导建立变应性鼻炎模型。将造模成功的24只SD大鼠按随机数字表法分为模型组、西药组、鼻敏康颗粒组,每组8只。西药组灌胃氯雷他定药液1 mg/kg,鼻敏康颗粒组灌胃鼻敏康颗粒药液4.1 g/kg,空白组与模型组大鼠灌胃等体积的生理盐水,1次/d,连续2周。观察并记录30 min各组大鼠鼻炎症状,采用HE染色观察大鼠鼻黏膜病理改变,ELISA法检测大鼠血清中IL-17、IL-6水平,Western blot法测定大鼠鼻黏膜组织中TNF、STAT3蛋白表达。 结果 预测到鼻敏康颗粒治疗变应性鼻炎靶点41个,PPI网络拓扑学分析得到TNF、STAT3等核心靶点,GO生物过程涉及药物反应、炎症反应、细胞因子反应等,KEGG通路主要富集于Th17细胞分化、脂质和动脉粥样硬化、IL-17、Toll样受体等通路。分子对接结果显示,主要活性成分与关键靶点结合活性良好。动物实验表明,鼻敏康颗粒可改善变应性鼻炎大鼠鼻炎症状,下调血清IL-17、IL-6表达,抑制TNF-α、STAT3蛋白表达。 结论 鼻敏康颗粒可通过多个活性成分、多靶点、多通路治疗变应性鼻炎,其机制可能与调控Th17细胞分化通路相关蛋白有关。 Objective To predict the molecular mechanism of Biminkang Granules in the treatment of allergic rhinitis using network pharmacological methods combined with animal experiments. Methods Active component targets and allergic rhinitis targets were screened from TCMSP, OMIM, GeneCards, TTD, DrugBank and PharmGKB databases R language software was used to map the intersection of drug and disease targets Cytoscape software and String platform were used to construct intersection target PPI network and conduct network topology analysis DAVID platform was used to perform GO enrichment and KEGG pathway analysis, and perform molecular docking verification on the main active components and key targets. 32 rats were divided into a blank group of 8 and a model group of 24 using a random number table method. Model rats were induced by ovalbumin to establish an allergic rhinitis model. 24 SD rats that were successfully modeled and were randomly divided into model group, Western medicine group, and Biminkang Granules group using a random number table method, with 8 rats in each group. The Western medicine group was gavaged with 1 mg/kg of loratadine solution, the Biminkang Granules group was gavaged with 4.1 g/kg of Biminkang Granules solution, and the blank group and model group rats were gavaged with the same volume of physiological saline once a day for 2 consecutive weeks. The symptoms of rhinitis in each group of rats for 30 minutes were observed and recorded, and the pathological changes of the rat nasal mucosa were observed using HE staining. ELISA method was used to detect the levels of IL-17 and IL-6 in rat serum, and Western blot method was used to determine the expressions of TNF and STAT3 proteins in rat tissues. Results A total of 41 target proteins of BiMinKang Dranule in the treatment of allergic rhinitis were predicted, and TNF, STAT3 and other core target proteins were obtained by PPI network topology analysis. The biological process of GO involved drug response, inflammatory response, cytokine response, etc.KEGG enrichment is involved in Th17 cell differentiation, lipid and atherosclerosis, IL-17, toll-like receptor and other pathways. Molecular docking results indicated that the main active components had good binding activity to key target proteins.Animal experiments showed that BiMinKang Dranule could improve the inflammatory symptoms of allergic rhinitis rats, down-regulate the expression of IL-17 and IL-6 in blood, and inhibit the expression of TNF and STAT3 proteins. Conclusion Biminkang Granules can treat allergic rhinitis through multiple active components, multiple target proteins and multiple pathways, and the mechanism may be related to the regulation of Th17 cell differentiation pathway related proteins.

    查看更多>>摘要:目的 探讨泻肺利水方治疗心力衰竭的有效成分及其潜在作用机制。 方法 采用超高效液相色谱串联四级杆飞行时间质谱(UPLC-Q-TOF-MS)技术分析鉴定泻肺利水方的活性成分。通过SwissTargetPrediction数据库进行药物靶点预测,并在GeneCards、DisGENET和TTD数据库收集疾病靶点,将药物靶点与疾病靶点取交集,采用STRING数据库构建PPI网络,筛选核心靶点,将核心靶点导入DAVID数据库进行GO功能富集和KEGG通路富集分析。将药物成分与对应的核心靶点进行分子对接验证。 结果 鉴定泻肺利水方10个活性成分,采用网络药理学筛选得到泻肺利水方治疗心力衰竭的潜在活性成分8个,所对应相关作用靶点160个,疾病相关靶点共收集到1 305个,两者交集靶点51个,17个核心靶点。GO功能富集和KEGG通路富集分析显示,泻肺利水方治疗心力衰竭的作用机制可能与蛋白结合、ATP结合、凋亡过程的负调控VEGF信号通路、T细胞受体通路等途径相关。分子对接结果显示,汉黄芩素与ESR1,毛蕊异黄酮与ESR1,以及槲皮素与AKT1、EGFR、IL2、ABCB1均显示较好的结合活性。 结论 泻肺利水方可能通过作用于ESR1、AKT1、EGFR等靶点,通过多条通路发挥治疗心力衰竭的作用。 Objective To explore the effective components and potential mechanisms of Xiefei Lishui Prescription in the treatment of heart failure. Methods Ultra high-performance liquid chromatography tandem four stage rod time of flight mass spectrometry (UPLC-Q-TOF-MS) technology was used to analyze and identify the active components of Xiefei Lishui Prescription. Drug targets were predicted through the Swiss Target Prediction database, and disease targets were collected from Gene Cards, Dis GENET, and TTD databases. The intersection of drug targets and disease targets was screened using a STRING database for protein interaction to identify core targets. The core targets were included in the DAVID database for GO enrichment and KEGG analysis. Finally, molecular docking validation was performed between the drug components and the corresponding core targets. Results The results identified 10 active components of Xiefei Lishui Prescription, and 8 potential active components were screened using network pharmacology for the treatment of heart failure with Xiefei Lishui Prescription, corresponding to 160 related action targets. A total of 1 305 disease-related targets were collected, and a total of 51 targets ad 17 core targets were included in the string database for protein interaction analysis. GO functional enrichment and KEGG analysis indicated that the mechanism of Xiefei Lishui Prescription in treating heart failure may be related to pathways such as protein binding, ATP binding, and negative regulation of the VEGF signaling pathway and T cell receptor pathway during apoptosis. The molecular docking results showed that baicalin exhibited good binding activity with ESR1, sorghum isoflavones with ESR1, and quercetin with AKT1, EGFR, IL2, and ABCB1. Conclusion Xiefei Lishui Prescription may exert therapeutic effects on heart failure through multiple pathways by targeting ESR1, AKT1, EGFR, and other targets.

    查看更多>>摘要:目的 采用超高效液相色谱法结合四极杆飞行时间质谱法技术(UPLC-Q-TOF/MS)与分子网络相结合的方法对黄芪及其蜜炙品成分进行分析,比较黄芪蜜炙前后主要成分含量变化。 方法 制备黄芪、炙黄芪水提物,采用UPLC-Q-TOF/MS检测其成分,采用全球天然产物分子网络分析平台(GNPS)进行分析鉴定,使用Cytoscape 3.9.1软件对生成的分子网络进行可视化分析。通过Masslynx 4.2软件根据化合物的二级碎片信息对化合物进行鉴别,分析黄芪炮制前后成分含量变化。 结果 从黄芪及其蜜炙品中鉴定出47个黄酮类和34个三萜皂苷类化合物,其中约87%黄酮类成分和82%皂苷类成分在蜜炙后含量下降。 结论 通过液质联用结合分子网络的方法可快速解析黄芪蜜炙前后的成分变化,黄芪经蜜炙后部分黄酮及皂苷类成分发生了水解反应,可能是炮制增效的物质变化基础。 Objective To analyse the composition of Astragali Radix and its honey-processed products through a combination of UPLC-Q-TOF/MS and molecular network To compare the changes in the main components of Astragali Radix before and after honey-frying. Methods The aqueous extracts of Astragali Radix before and after honey-frying were prepared, and the compositions were detected by UPLC-Q-TOF/MS, analyzed and identified by the Global Natural Products Molecular Network Analysis Platform (GNPS). The generated molecular networks were visualized and analyzed using Cytoscape 3.9.1 software. The compounds were identified by Masslynx 4.2 software based on the secondary fragmentation information of the compounds, and the changes in the content of the components before and after the processing of Astragali Radix were analysed. Results 47 flavonoids and 34 triterpenoid saponins were presumably identified from Astragali Radix and its honey-frying products using the above analytical methods, with about 87% of the flavonoids and about 82% of the saponins decreasing in content after honey-frying. Conclusions The compositional changes of Astragali Radix before and after honey-frying are rapidly resolved and visualised by liquid-quantity coupling combined with molecular network. It is found that some of the flavonoids and saponins components of Astragali Radix underwent hydrolysis after honey-frying and it may be the material change basis for processing efficiency enhancement.

    查看更多>>摘要:目的 以五倍子中没食子酸为研究对象,探讨Image J软件用于薄层色谱定量的可行性。 方法 采用硅胶GF254薄层板,以三氯甲烷-甲酸乙酯-甲酸(5∶5∶1)为展开剂,紫外光灯(254 nm)下拍照;采用聚酰胺薄膜,以75%乙醇-冰醋酸(50∶1)为展开剂,1%三氯化铁乙醇溶液为显色剂,日光下拍照。将图像导入Image J软件进行分析,以外标两点法计算五倍子中没食子酸含量。采用HPLC法,以甲醇-0.1%磷酸溶液(15∶85)为流动相,在273 nm波长下测定五倍子中没食子酸含量。 结果 硅胶GF254薄层板没食子酸定量限为0.401 6 μg,线性范围2.855~9.515 μg(r 2=0.996 0),回收率为105.12%(RSD=3.48%);聚酰胺薄膜没食子酸定量限为0.363 4 μg,线性范围1.427~4.758 μg(r 2=0.991 5),回收率为103.75%(RSD=4.60%);HPLC法没食子酸的定量限为4.42 ng,线性范围0.122~0.977 μg(r 2=0.999 9),回收率为98.30%(RSD=1.40%),3种方法的精密度、重复性、稳定性RSD值均<5%。使用Image J测得的没食子酸含量与HPLC测定结果比较,最大相对误差9.30%,最小相对误差1.62%。 结论 Image J软件可用于薄层色谱定量分析,可作为中药质量控制方法的补充。 Objectives To discuss the feasibility of Image J in quantitative analysis on thin layer chromatography (TLC) using gallic acid in Galla Chinensis as research object. Methods Silica gel GF254 thin-layer plate was used with chloroform-ethyl formate-formic acid (5:5:1) as the developing solvent and the images were taken under ultraviolet light (254 nm). Polyamide film was used with 75% ethanol-glacial acetic acid (50:1) as the developing solvent and 1% ferric chloride ethanol solution as the chromogenic reagent, and the images were taken under sunlight. Images obtained from the above conditions were imported into Image J to analyze and calculate the content of gallic acid in Galla Chinensis by using external standard two-point method. High-performance liquid chromatography (HPLC) was used with a mobile phase of methanol-0.1% phosphoric acid solution (15:85) at a wavelength of 273 nm to determine the gallic acid content in Galla Chinensis. Results The quantitation limit of gallic acid on silica gel GF254 thin-layer plate was 0.401 6 μg, the linear range was 2.855 - 9.515 μg (r2 = 0.996 0), and the average recovery was 105.12% (RSD=3.48%) the quantitation limit of gallic acid on polyamide film was 0.363 4 μg, the linear range was 1.427 - 4.758 μg (r2 = 0.991 5), and the average recovery was 103.75% (RSD=4.60%). The HPLC method had a quantitative limit of 4.42 ng, a linear range of 0.122-0.977 μg (r2 = 0.999 9), and a recovery rate of 98.30% (RSD = 1.40%). The accuracy, repeatability and stability of RSD were all <5%. The gallic acid content measured using Image J showed a maximum relative error of 9.30% and a minimum of 1.62% compared to the HPLC results. Conclusions Image J is feasible for quantitative analysis of TLC and can be used as a complementary method for quality control of Chinese materia medica.

    查看更多>>摘要:目的 基于数据挖掘及知识图谱技术探究周楣声教授医案手稿中灸法治疗流行性出血热(EHF)的治疗规律。 方法 收集并整理周教授医案手稿中砀山县人民医院传染科1985年12月16日-1987年12月25日住院患者灸治EHF诊疗记录,应用Graphpad Grism 8.0软件进行描述性分析,通过PHP 5.4程序代码进行关联规则分析,运用SPSS Statistics 26.0进行聚类分析,使用Neo4j Community 3.5.25数据库进行证型权值图谱分析。 结果 共纳入处方205首,频次>40的症状有21个,其中恶寒、发热、发疹、烦躁出现频率为100%。治疗主要采用的灸法有灸架熏灸法、万应点灸笔灸法、火针代灸灸法等,挑治法为频次最多的中医辅助操作。频次>25的穴位有29个,高频腧穴有中脘、肾俞、命门等。关联规则分析表明,三阴交-中脘-肺俞-肾俞-至阳关联性较高。聚类分析得出6个有效灸治EHF取穴聚类组合。权值图谱得出目标证型关联性较高的前30条关系。 结论 周教授将“热证贵灸”思想运用于EHF的治疗中,以“因症选穴”法作为灸法治疗EHF的主要选穴思路,临证多以灸法配合中医辅助操作治疗EHF,可取得较好疗效。 Objective To explore the therapeutic law of moxibustion in Professor Zhou Meisheng's medical manuscripts for epidemic hemorrhagic fever (EHF) based on data mining and knowledge map technology. Methods The manuscript data of Professor Zhou Meisheng's moxibustion treatment of EHFwere collected from Infectious Diseases Department of Dangshan County People's Hospital from December 16, 1985 to December 25, 1987. Graphpad Grism 8.0 software was used for descriptive analysis. PHP 5.4 program code was used for association rule analysis. SPSS Statistics 26.0 was used for clustering analysis. Neo4j Community 3.5.25 database was used to analyze the syndrome-weight graph. Results 205 prescriptions were included. There were 21 symptoms with frequency>40, in which the frequency of aversion to cold, fever, rash and irritability was 100%. The main types of moxibustion methods used in the treatment included moxibustion frame fumigation moxibustion, Wanying acupoint moxibustion pen moxibustion, and fire needle instead of moxibustion. There were 29 acupoints with a frequency of >25, including Zhongwan (CV12), Shenshu (BL23) and Mingmen (DU4), etc. Association rules showed that Sanyinjiao (SP6)-Zhongwan (CV12)-Feishu (BL13)-Shenshu (BL23)-Zhiyang (DU9) had the highest correlation. Six effective clustering combinations of moxibustion for EHF were summarized by clustering analysis. The weight graph can obtained the first 30 relationships with high correlation of target syndromes. Conclusions Professor Zhou applied the idea of "moxibustion for heat syndrome" to the treatment of EHF, and took the method of "acupoint selection according to symptoms" as the main acupoint selection idea for moxibustion treatment of EHF. In clinical practice, moxibustion combined with auxiliary operation of TCM is often used to treat EHF, which can achieve good results.

    查看更多>>摘要:目的 分析国家专利中药复方治疗PCOS的用药及配伍规律,为治疗PCOS的中药新方提供参考。 方法 检索国家知识产权局中国专利公布公告网站建库至2023年4月20日发布的治疗PCOS的中药复方专利,采用SPSS 25.0、IBM SPSS Modeler 18.0及Cytoscape 3.8.0等软件对数据进行频次统计、关联规则分析,构建中药核心复杂网络,并进行系统聚类分析。 结果 纳入复方专利126项,涉及中药392味,总频次为1 709次,用药药性以温性为主,药味以甘味为主,归经以肝经为主;高频药物有当归(46次)、菟丝子(44次)、香附(42次)、熟地黄(41次)、丹参(37次)等;常用药对有茯苓-苍术(支持度16.67%,置信度76.19%)、当归-白芍(支持度15.87%,置信度80.00%)等,常用角药有香附-半夏-茯苓(支持度12.70,置信度81.25%)等;核心处方有“茯苓、半夏、陈皮、香附、丹参、淫羊藿、白术、巴戟天、皂角刺、石菖蒲”等;潜在处方有“麦芽、谷芽、大血藤、葛根、茺蔚子”等。 结论 中药复方专利治疗PCOS以燥湿健脾、补肾阳并滋肾阴、活血化瘀消癥等治法为主。 Objective To analyze the medication and compatibility law of TCM compounds with national patents for the treatment of polycystic ovary syndrome (PCOS) To provide reference for the new TCM prescriptions for the treatment of PCOS. Methods TCM compound patents for PCOS were retrieved from the China National Intellectual Property Administration's Chinese Patent Announcement Website from the establishment of the website to April 20, 2023. SPSS 25.0, IBM SPSS Modeler 18.0, and Cytoscape 3.8.0 were used to carry out frequency statistics, association rule analysis on the data, and a TCM core complex network and systematic clustering analysis were built. Results 126 compound patents were included, involving 392 kinds of Chinese materia medica, with a total frequency of 1 709 times. The medicinal property was mainly warm, the taste was mainly sweet, and the meridian was mainly liver meridian. High frequency drugs included Angelica Sinensis (46 times), Cuscutae Semen (44 times), Cyperi Rhizoma (42 times), Rehmanniae Radix Praeparata (41 times), Salviae Miltiorrhizae Radix et Rhizoma (37 times), etc. Commonly used medicinal pairs included Poria-Atractylodis Macrocephalae Rhizoma (support 16.67%, confidence 76.19%), Angelica Sinensis-Paeoniae Radix Alba (support 15.87%, confidence 80.00%), etc. The triple drug combinations were Cyperi Rhizoma-Pinelliae Rhizoma-Poria (support 12.70%, confidence 81.25%).
