    Research progress on COVID-19 combined with digestive system diseases

    查看更多>>摘要:The corona virus disease 2019 (COVID-19) is an acute respiratory infection caused by the binding of SEVERE acute respiratory syndrome coronavirus 2 (SARS-COV-2) and the angiotensin-converting enzyme 2 (ACE2) receptor to the host target organ. New research suggests that COVID-19 may be a systemic infectious disease and a systemic inflammatory disease.Symptoms and signs of the digestive system are often found in patients with coVID-19 following acute infection.At present, despite the existence of some coronavirus-related digestive diseases (such as gastrointestinal diseases, liver injury, pancreatic injury, etc.), the pathogenesis of COVID-19 with digestive symptoms in existing studies is still unclear, and relevant guidelines and expert consensus are not enough for clinical diagnosis and treatment. Therefore, it is extremely important to understand the clinical manifestations, pathogenesis and prognosis of coVID-19 patients with digestive system.

    查看更多>>摘要:Objective: To explore the changes of cannabinoid receptor (CB1) and AMPA receptor subunit GluA1 in the medial prefrontal cortex (mPFC) of depression model rats induced by chronic unpredictable mild stress (CUMS). Methods: 30 rats were randomly divided into three groups. The three-week model group was given a three-week CUMS model, the four-week model group was given a four-week CUMS model, and the control group was given no treatment. The behavior of rats were detected in each group, and the expression of CB1 and GluA1 protein in synaptosomes in mPFC brain region was detected by Western blot. Results: Compared with the control group, the surcose preference decreased, the feeding latency increased in the novel inhibition feeding test, and the immobility time increased in the forced swimming test in the four-week model group. However, after three weeks of CUMS, there was no obvious change in the behavior of rats compared with the control group, suggesting that four-week model of CUMS was successful. CUMS can reduce the expression level of CB1 and GluA1 protein in synaptosomes of mPFC brain area in four-week model group (P<0.05), but there was no significant difference between three-week model group and control group. Conclusion: Four-week CUMS was more likely to lead to depressive-like behavior in rats, which may be closely related to the expression of CB1 and GluA1 in mPFC.

    Effects of acupuncture combined with Kaijingtongmai Decoction on ATP sensitive potassium channel related proteins Kir6.1 and Kir6.2 in myocardial infarction rats

    查看更多>>摘要:Objective: To study the effect of combination of acupuncture and medicine on the expression of ATP sensitive potassium channel related proteins Kir6.1 and Kir6.2 in rats with myocardial infarction, and to study the possible mechanism of combination of acupuncture and medicine on the improvement of myocardial infarction, so as to provide experimental data basis for the development of new treatment methods for myocardial infarction. Methods: 65 healthy male SD rats were randomly selected as the control group. The other rats were fed with high-fat food for three weeks. The rats in the control group were injected with normal saline subcutaneously, and the other rats were injected with isoproterenol hydrochloride in the same way. Through ECG comparison, 40 successful Mi rats were randomly divided into model group, acupuncture group, western medicine group and acupuncture drug combination group, with 10 rats in each group. After the corresponding treatment, the ECG changes of rats in each group were observed, the pathological changes of rat cardiomyocytes were observed by HE staining, and the expression of ATP sensitive potassium channel (Kir6.1, Kir6.2) protein was detected by Western blot. Results: compared with the control group, 40 experimental specimens in the experimental group showed significant changes in cardiomyocyte protein The expression of Kir6.1 and Kir6.2 increased, and the difference was statistically significant. After treatment, compared with the model group, the protein expression of Kir6.1 and Kir6.2 in cardiomyocytes of Western medicine group, acupuncture group and acupuncture drug combination group showed a downward trend, among which the decline degree of acupuncture drug combination group was the most obvious, and the difference was statistically significant. The decline degree of acupuncture group and Western medicine group was not significant, and there was no significant difference Conclusion: acupuncture combined with medicine has a significant effect on improving myocardial infarction in rats, which may be related to the expression of ATP sensitive potassium channel related proteins Kir6.1 and Kir6.2 in rat cardiomyocytes.

    查看更多>>摘要:Objective: To investigate the intervention effect and specific mechanism of Jisheng Shenqi Decoction plus Panax notoginseng and Turmeric on carbon tetrachloride (CCl4)-induced liver fibrosis in rats. Methods: SPF SD rats were randomly divided into control group, model group, and Chinese medicine treatment (low, medium and high dose) groups. The model group and Chinese medicine treatment group were given intraperitoneal injection of 40% CCl4 olive oil solution twice a week. A rat model of liver fibrosis was replicated by the method for 8 weeks. After successful modeling, each drug-administered group was gavaged with corresponding drugs, and the control group and model group were gavaged with equal volume of distilled water, once a day, for 4 weeks. After the last intragastric administration, HE staining and Masson staining were used to observe the pathological morphology of liver tissue, and liver function (ALT, AST) was detected; ELISA method was used to determine transforming growth factor β (TGF-β), angiotensin Ⅱ( Ang-Ⅱ), chitinase 3-like protein 1 (CHI3L1), interleukin-1 (IL-1), interleukin-6 (IL-6), tumor necrosis factor-α (TNF-α) content; The expression levels of type I, type Ⅲ collagen (Col-Ⅰ, Col-Ⅲ) and TGF-β mRNA were determined by RT-PCR. Results: ①HE and Masson staining showed that a large number of liver cells in the model group were inflamed and necrotic, the collagen fibers in the liver tissue were extensively proliferated, the fibrous septa were interconnected, and the pseudolobules were formed. Compared with the model group, the levels of ALT, AST, TGF-β, Ang-Ⅱ, CHI3L1, IL-1, IL-6 and TNF-α in the traditional Chinese medicine treatment group were decreased, while the levels of Col-Ⅰ, Col-Ⅲ The expressions of Ⅲ and TGF-β mRNA were decreased, and the effect was most obvious in the middle and high dose groups of traditional Chinese medicine (P<0.01). Conclusion: Jisheng Shenqi Decoction combined with Panax notoginseng and Biejia can significantly improve liver fibrosis induced by CCl4 in rats, and its mechanism may be related to the down-regulation of angiotensin-converting enzyme (ACE)-angiotensin Ⅱ(Ang-II)-angiotensin Ⅱ receptor 1 (AT1R) signaling pathway related gene expression, reducing the level of related cytokines, promoting the degradation of extracellular matrix and so on.

    查看更多>>摘要:Objective: To observe the effect of lipopolysaccharide (LPS) on cytoskeleton and the effect of Dahuang Lingxianfang on nF-KB /MAPK signaling pathway. Methods: Biliary epithelial cells of each group were stained with photolipin fluorescent staining, and the arrangement of cytoskeleton was observed under laser confocal microscope. Western blotting was used to detect the expression of f-actin. Results: After LPS intervention, the biliary epithelial cells showed nuclear shrinkage or damage, and the skeleton was broken or lumped. The cytoskeleton was partially repaired after the intervention of pathway blocking preparation combined with RHUbarb Lingxianfang. All the other groups had different degree of cytoskeleton fracture. Compared with normal group, the expression of F-actin protein in LPS group was decreased (P < 0.05); Compared with LPS group, the expression of F-actin in LPS+ TCM group, LPS+PDTC+ TCM group, LPS+SB203580+ TCM group and LPS+PDTC+SB203580+ TCM group was significantly increased (P < 0.05); Compared with traditional Chinese medicine group, the expression of F-actin in LPS+PDTC+ traditional Chinese medicine group, LPS+SB203580+ traditional Chinese medicine group and LPS+PDTC+SB203580+ traditional Chinese medicine group had no significant difference (P > 0.05). Conclusion: RHUbarb Lingxianfang can restore the sequence of biliary epithelial cytoskeleton and protect its microfilament structure under inflammation, and its mechanism may be related to the regulation of NF-KB /MAPK signaling pathway.

    Clinical effect of enriching qi, activating blood circulation, clearing away dampness and heat combined with western medicine in the treatment of idiopathic membranous nephropathy: A meta-analysis

    查看更多>>摘要:Objective: To systemically review the efficacy and safety about the method of enriching qi, activating blood circulation, clearing away dampness and heat combined with western medicine in the therapy of patients with idiopathy membranous nephropathy (IMN). Methods: We collected the randomized controlled trials (RCTs) of enriching qi, activating blood circulation, clearing away dampness and heat for the treatment of IMN from Pub Med, The Cochrane Library, Medline, China National Knowledge Infrastructure (CNKI), China Biology Medicine disc (CBM), Wanfang Data and Wiper Databases. Bias of risk of retrieval literature was evaluated according to Cochrane Collaboration standard, and Review Manager 5.3 software was used for statistical analysis. Results: Fourteen trials (836 participants) were included in the meta-analysis. This kind of traditional Chinese method combined with western medicine in the treatment of IMN exerted statistical differences in reducing 24-hour urinary protein [WMD=-0.97, 95%CI(-1.30,-0.65), Z=5.86(P<0.00001)] and elevating serum albumin [WMD=3.83, 95%CI(2.10, 5.57), Z=4.33, P<0.0001], lowering serum cholesterol [WMD=-0.82, 95%CI(-1.08, -0.56), Z=6.18, P<0.00001], triglycerides [WMD=-0.39, 95%CI(-0.67, -0.11), Z=2.77, P=0.006] and reducing the risk of adverse events[OR=0.29, 95%CI[0.16, 0.50], Z=4.35, P<0.0001] in the patients with IMN as compared with controls. However, there was no statistically significant difference between the method and controls when combining all trials in serum creatinine [WMD=-5.52, 95%CI(-18.06, 7.03), Z=0.86, P=0.39], when combining all trials in urea nitrogen[WMD=-0.90, 95%CI(-2.22, 0.41), Z=1.35, P=0.18]. Conclusion: The method of enriching qi, activating blood circulation, clearing away dampness and heat combined with western medicine exerts certain advantages and better safety in treating patients with IMN. However, for the inferiority of the included studies, the conclusion still needs high-quality and large-sample prospective randomized controlled trials to verify.

    Exploring the molecular biological mechanism of Shugan Jianpi Decoction in the treatment of depression-related breast cancer based on network pharmacology

    查看更多>>摘要:Objective: To explore the molecular biological mechanism of Shugan Jianpi Decoction in treating depression-related breast cancer based on network pharmacology. Methods: The active compounds of Shugan Jianpi Decoction were explored to obtain their drug targets, and the venny network, "TCM-component-target" network, PPI network, "pathway-gene" network, GO and KEGG enrichment results were constructed using relevant softwares. Results: A total of 121 active ingredients , 250 genes, 2357 GO functions and 184 KEGG pathways were obtained. Conclusion: The active ingredients of Shugan Jianpi Decoction are quercetin||、kaempferol、luteolin、naringenin.The key targets are MAPK1, MAPK3, RELA, and AKT1. GO functions include cellular response to hormone stimulus、cellular response to lipid、cellular response to oxidative stress and so on.KEGG mainly involves PI3K-Akt, microRNAs, and FoxO signaling pathways.This study preliminarily explores the molecular biological mechanism of Shugan Jianpi Decoction in the treatment of depression-related breast cancer, and speculates that it mainly acts by arresting the cell cycle, inhibiting cancer cell proliferation, mediating cancer cell apoptosis, and preventing its recurrence and metastasis, which provides new evidence and new direction for subsequent clinical application and experimental research.

    查看更多>>摘要:Objective: To analyze the rule of prescribing traditional Chinese medicine for treating pneumoconiosis, so as to provide reference for differential diagnosis and treatment of pneumoconiosis as well as for the development of new drugs for treatingthe disease. Methods: We searched China National Knowledge Infrastructure, Wanfang Database and VIP Chinese PublicationDatabase to retrieve relevant literatures which were then screened according to the enrollment criteria to establish a prescriptiondatabase of traditional Chinese medicine for the treatment of pneumoconiosis. The inheritance calculation platform of traditionalChinese medicine was used to analyze the prescribing rule of traditional Chinese medicine in the treatment of pneumoconiosisbased on association rules, k-means clustering algorithm and regression model analysis. Results: A total of 131 related literature were preliminarily selected, from which 97 prescriptions of traditional Chinese medicine with a total of 195 herbs were included.The most frequently prescribed herbs included Radix astragali, Platycodon grandiflorum, Pinellia ternata, licorice, Codonopsispilosula, Salvia miltiorrhiza, bitter almond etc. A total of 14 association rules, 13 high-frequency herb pairs were found and 5groups of formulas were revealed by cluster analysis. Conclusion: The prescriptions for the treatment of pneumoconiosis are mainly composed of herbs for tonifying deficiency, resolving phlegm, relieving cough and asthma, activating blood circulation and removingblood stasis, which are supplemented with herbs for clearing heat, relieving appearance, regulating qi, promoting waterand permeating dampness, etc. , The prescribing rules reflect the basic pathological characteristics of lung deficiency and collateral arthralgia in pneumoconiosis, which provides some ideas for the clinical differentiation and treatment of pneumoconiosis in traditionalChinese medicine. It also provides reference for the research and development of new treatment methods.

    Study on the medication rules of traditional Chinese medicine in the treatment of sleep disorder after stroke based on data mining

    查看更多>>摘要:Objective: To explore the medication rule of Traditional Chinese Medicine(TCM) in the treatment of sleep disorder after stroke by using data mining technology. Methods: A computer search was used to search the electronic database of clinical literature on the treatment of sleep disorders after stroke by TCM from January 2000 to January 2021. Excel was used to establish the database, and the prescription information was described and analyzed statistically. Using IBM SPSS Modeler 18.0 software, Apriori algorithm was used for TCM association analysis, and IBM SPSS 22.0 software was used for systematic cluster analysis of high-frequency TCM. Results: A total of 67 literatures were included, covering 131 traditional Chinese medicines.The medecines with a higher frequency of sodium use include Ziziphi Spinosae Semen(Suanzaoren), Angelicae Sinensis Radix(Danggui), Ligusticum (Chuanxiong), liquorice(Gancao), Poria cocos(Fuling), and so on. From the effect point of view, deficiency-tonifying medicine, sedative medicine and blood-activating and stasis-removing medicine are commonly used. The medicinal properties are mainly cold, mild and warm. The main medicine flavor are sweet and bitter. The medicines mostly belong to the liver, heart and spleen Meridian. Thirty-three association rules were obtained for medicine pairs and medicine groups from the correlation analysis, and the core combinations were "Ziziphi Spinosae Semen(Suanzaoren)-Tuber fleeceflower stem(Yejiaoteng)", "Ziziphi Spinosae Semen (Suanzaoren)-Polygala(Yuanzhi)", "Ziziphi Spinosae Semen(Suanzaoren)-Cortex albiziae(Hehuanpi)" and "Angelicae Sinensis Radix(Danggui)-Radix bupleuri(Chaihu)-Radix Paeoniae Alba(Baishao)" and so on. Seven medicine aggregation groups were obtained by medicine cluster analysis. Conclusion: In the treatment of sleep disorder after stroke by TCM, the main method is to calm the heart and mind. Meanwhile, according to different syndrome types, the treatment methods of tonifying the heart and spleen, nourishing the liver and kidney, soothing the liver and softening the liver, clearing heat and resolving phlegm, nourishing the blood and promoting blood circulation are selected, which provide certain reference for clinical treatment.

Modern studies have shown that the abnormality of brain-gut peptides is closely related to the occurrence of irritable bowel syndrome. This article starts with vasoactive intestinal peptide, substance P, serotonin, neuropeptide Y, corticotropin releasing factor, calcitonin gene-related peptide, cholecystokinin and other brain-gut peptides and their correlation with intestinal flora, to summarize the treatment of irritable bowel syndrome with traditional Chinese medicine in recent years.
