    查看更多>>摘要:Since the outbreak of Novel coronavirus pneumonia(COVID-19), Novel coronavirus has undergone evolution and variation in the process of transmission, and has formed many variant strains. There are five variants of concern, Alpha, Beta, Gamma, Delta and Omicron. In November 9, 2021, South Africa firstly detected a variant strain (B.1.1.529) from a sample of cases. In November 26, the World Health Organization named it "Omicron variant strain", Currently, the Omicron variant strain has become the main epidemic variant strain in the world. Omicron variant strain consists of several subtypes, including BA.1, BA.2 and BA.3. The subtypes of Omicron variant strain have different mutation and infectivity. This article reviews the characteristics and immune escape mechanisms of each Omicron variant strain subtype.

    查看更多>>摘要:Objective: To investigate the effects of cagliazin, a sodium-glucose cotransporter 2 inhibitor (SGLT-2I), on ventricular remodeling in spontaneously hypertensive rats (SHR) through renin angiotensin system (RAS) and transforming growth factor -β1(TGF-β1). Methods: The experiment was divided into 4 groups: normal blood pressure control group, SHR group, cagliet net low-dose group (30mg/kg), cagliet net high-dose group (60mg/kg), once a day for 8 weeks. Normal blood pressure rats (WKY) were used as the control group to measure blood pressure with tail sleeve sphygmomanometer (BP) and blood glucose level was measured with glucose meter Cardiac function was evaluated by echocardiography, cell area of left ventricle was evaluated by histomorphology, real-time quantitative polymerase chain reaction and protein imprinting hybridization were used to detect TGF-β1 Smad4 renin from type I collagen (Col1a) type Ⅲ collagen (Col3a) matrix metalloproteinase 2(MMP-2) Expression results of angiotensin Ⅱ1 type receptor 1(AGTR1) and Angiotensin Ⅱ2 type receptor 2 (AGTR2).     查看更多>>摘要:Objective: The effect of Qingguangan Ⅱ on the transcription of RhoA mRNA, ROCK mRNA, Caspase-3 mRNA and Bcl-2 mRNA in the retina of DBA / 2J mice was observed. Methods: Forty-eight DBA/2J mice were randomly divided into six groups: model groups, Qingguangan Ⅱ decoction group, low concentration, medium concentration and high concentration group of Qingguangan Ⅱ effective ingredient and positive control group (Yimaikang tablet group), and eight C57BL / 6 mice were used as blank group, DBA/2J mice were fed until 38 weeks before forming a glaucoma model, The transcription of RhoA mRNA, ROCK mRNA, Caspase-3 mRNA and Bcl-2 mRNA in the retinal of DBA/2J mice was detected using real-time fluorescence quantitative PCR (Quantitative Real-time PCR) after 4 weeks of intervention. Conclusion: The high concentration of Qingguangan Ⅱ prescription probably attenuated Caspase-3 transcription in retinal ganglion cells by inhibiting the Rho / ROCK signaling pathway and activated Bcl-2 expression by inhibiting ROCK signaling, which attenuated apoptosis in retinal ganglion cells.

    Effect of HK3 on immune invasion, proliferation and invasion of colon cancer cells

    查看更多>>摘要:Objective: Investigate the effects of Hexokinase-3 on the proliferation, cell cycle, migration and immunologic invasion of RKO and HCT116. Methods: The expression levels of HK3 in tumor and normal colon samples were analyzed by bioinformatics. RKO and HCT116 were transfected with the control group (si-NC) and interference group HK3(si-HK3#1, si-HK3#2). CCK-8, Flow cytometry, wound healing and Transwell were used to examine the effects of interference with HK3 on the proliferation, cell cycle and migration of RKO and HCT116 cells. The expression levels of vimentin and E-cadherin, which are related to epithelial-mesenchymal transition(EMT), were detected by Western blot. Results: HK3 is associated with infiltration of multiple immune cells and co-expression with multiple immune checkpoints in colorectal cancer. Compared with the Control group (si-NC),the protein expression level of HK3 in interference group (si-HK3#1, si-HK3#2) was significantly decreased(P<0.05);the proliferation and migration of RKO and HCT116 cells were significantly inhibited(P<0.05);the cell cycle of RKO and HCT116 cells was arrested in S phase(P<0.05) and the expression of E-cadherin protein was increased, but the expression of vimentin protein was inhibited(P<0.05). Conclusion: HK3 affects the level of immunologic invasion of colon cancer. Interference with HK3 inhibits the proliferation, cell cycle and migration of colon cancer cells by inhibiting EMT behavior.

    查看更多>>摘要:Objective: To investigate the relationship between serum histone Deacetylase 3(HDAC3) , High Mobility Group B 1(HMGB-1) and the occurrence, development and structural remodeling of nonvalvular atrial fibrillation. Methods: Patients admitted to non-valvular atrial fibrillation (atrial fibrillation AF group, n=166) were selected and divided into paroxysmal atrial fibrillation group (paroxysmal AF group, n=42), persistent atrial fibrillation group (persistent AF group, n=63), and permanent atrial fibrillation group (permanent AF group, n=61).50 cases of sinus rhythm were selected as the control group, general clinical data, heart color ultrasound, NT-proBNP, and the levels of HDAC 3 and HMGB-1 were determined by enzyme-linked adsorption immunoassay (ELISA). Results: The levels of HDAC3, HMGB-1, LAD, and LVEDD were all higher than those in the control group (P <0.05); The levels of LAD, LVEDD, HDAC3, and HMGB-1 were all higher in the permanent AF groups than in the persistent AF versus paroxysmal AF groups (P <0.05), And the persistent AF group was higher than the paroxysmal AF group (P <0.05); According to the Pearson correlation analysis, HDAC3 vs. LAD (r=0.635), P<0.01), LVEDD(r=0.651, P<0.01), HMGB-1(r=0.757, P <0.01) showed a positive correlation; HMGB-1 was compared with LAD (r=0.658), P<0.01), LVEDD(r=0.569, P <0.01) showed a positive correlation; After a binary logistic regression analysis, HDAC3(OR= 1.712, 95%CI:1.070-2.739, P<0.05), HMGB-1(OR=1.287, 95%CI:1.008-1.634, P <0.05) was associated with whether atrial fibrillation occurred. Conclusion: The levels of HDAC3 and HMGB-1 are correlated to the occurrence and development of AF and are independent risk factors for the occurrence of AF; HDAC3 and HMGB-1 levels are related to the structural remodeling of atrial fibrillation.

    查看更多>>摘要:Objective:To explore the correlation between the expression of serum total IgE level and peripheral blood EOS count in patients with eczema and the severity of clinical eczema.Methods:The EASI and pruritus VAS score were used to evaluate the disease severity of 100 patients with eczema.According to the EASI score,patients with eczema were divided into three groups:mild(<7 points)、moderate(7-15 points)and severe(>15 points).At the same time,the serum total IgE level and peripheral blood EOS count were tested before and after the 4-week Chinese medicine treatment.Results:The serum total IgE level of 42%of eczema patients increased,and the peripheral blood EOS count increased in 26%.In the three groups of mild,moderate,and severe,as the severity of eczema increased,the level of serum total IgE and peripheral blood EOS count continued to increase,and the positive rate increased significantly(all P<0.05).In terms of correlation analysis,the study showed that in patients with eczema total IgE level and EASI score(r=0.421,P<0.01),EOS count and EASI score(r=0.326,P<0.01),EOS count and pruritus VAS score(r=0.529,P<0.01)all show relatively weak positive correlation;and a high proportion of patients had the same changes in IgE and EOS after treatment(82%of IgE is consistent with EASI,76%of EOS is consistent with EASI).Conclusion:This study shows that the total IgE level and EOS count of patients with eczema are positively correlated with the severity of eczema,but at the same time this correlation is relatively limited.In clinical practice,it is necessary to combine the dynamic changes of the individual's own indicators in order to diagnose,comprehensively treat and evaluate eczema.Provide greater reference value in other aspects.

    Study on key genes and pathways of myocardial fibrosis and prediction of effective traditional Chinese medicine

    查看更多>>摘要:Objectives:Bioinformatics was applied to screen the key genes of Myocardial fibrosis, explore its pathogenesis and predict the potential traditional Chinese medicines for the treatment of Myocardial fibrosis. Methods: Based on raw data of gene chip GSE59437 from gene expression database (GEO), myocardial tissue samples from 3 control mice and 3 mice treated with angiotensin Ⅱ-induced myocardial fibrosis were included.Using R language processing data and screening of gene express significant differences (DEG), use a database of DAVID and the R language finish Gene Ontology(GO) annotation and Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway enrichment for differences gene, using the STRING database structure protein protein interactions (PPI) networks, using Cytoscape software visualization and use the MCODE plug-in screening key function modules in the network.Coremine Medical database was used to map the key genes, construct the gene-Chinese medicine network, and screen the traditional Chinese medicines for the treatment of myocardial fibrosis. Results: 208 DEGs were screened, 94 of which were up-regulated and 114 were down-regulated.DEGs is mainly involved in a variety of biological processes such as extracellular matrix remodeling, collagen fiber deposition and lipid metabolism disorders. KEGG pathway enrichment involves Platelet activation,Oxytocin signaling pathway,Insulin secretion,ECM-receptor interaction,GnRH signaling pathway,TNF signaling pathway and other signaling pathways. Key modules of PPI network including: CTGF,TIMP1,SPP1,SERPINE1,COL3A1, POSTN and FOS. The potential traditional Chinese medicines for the treatment of myocardial fibrosis are Astragalus membranaceus (Fisch.), Lepidium apetalum Willd and Salvia miltiorrhiza Bge. Conclusion: Myocardial fibrosis is a complex pathological process, and the genes related to the imbalance of extracellular matrix synthesis and degradation and excessive deposition of collagen fibers play an important role in this process. This study provides a scientific reference for further exploring the pathogenesis of myocardial fibrosis, looking for therapeutic targets and potential therapeutic traditional Chinese medicines.

    Study on the mechanism of Shengjiang Xiexin Decoction in the treatment of chemotherapy-induced diarrhea based on network pharmacology and molecular docking

    查看更多>>摘要:Objective: To explore the key targets and mechanism of Shengjiang Xiexin Decoction in the treatment of chemotherapy-induced diarrhea based on network pharmacological methods. Methods: The effective components and corresponding target proteins of Shengjiang Xiexin Decoction were screened by TCMSP, and the target of chemotherapy-induced diarrhea were screened by the GeneCards. R software was used to obtain the common targets of drugs and diseases, and the "component-target-disease" network diagram was constructed by Cytoscape3.8.0 software. The string datebase was used to draw the protein interaction (PPI) network, and the Bioconductor software was used to perform GO function and KEGG pathway enrichment analysis on effective targets. Result: The result showed that 216 components were screened and 276 effective targets were screened. There were 1764 chemotherapy-induced diarrhea targets. The 173 common targets were obtained through venn diagram. The GO function analysis found 2427 items of biological process, 168 items of molecular function and 79 items of cellular component. The KEGG pathway analysis found 169 items. Conclusion: The PPI network found that STAT3、AKT1、MAPK3、JUN、MAPK1、RELA、IL6、etc. may be the key targets for Shengjiang Xiexin Decoction in treatment of chemotherapy-induced diarrhea. GO biological processes include DNA-binding transcription factor activity, cytokine receptor binding, cytokine activity, response to lipopolysaccharide, cellular response to chemical stress and so on. The KEGG pathways involved mainly include Toll-like receptor signaling pathway, TNF signaling pathway, inlfuenza A signaling pathway、hepatitise B signaling pathway and other pathways, that Play the role of anti-inflammatory and repair barrier.

    Clinical effect of governor meridian moxibustion on treatment of lumbar disc protrusion: A meta analysis

    查看更多>>摘要:Objective:To explore the clinical effect of governor meridian moxibustion on lumbar intervertebral disc protrusion. Methods:Randomized controlled trials on treatment of lumbar intervertebral disc protrusion with governor meridian moxibustion and combined governor meridian moxibustion were retrieved from CNKI,VIP,Wan Fang Data,CBM,Web of ScienceTM Core Collection,EBSCO Medline,PubMed,OVID,the Cochrane Library,and other databases. The time was from the inception of the database until October 2021. The studies were screened according to the inclusion and exclusion criteria,extracted,and evaluated for quality assessment. The data were analyzed with Review Manager 5.3 software. Results:A total of 24 randomized controlled trials(2379 cases in total)were included. Meta-analysis results showed that compared with control groups such as acupuncture,electroacupuncture,and massage,the governor meridian treatment improved the clinical efficacy[RR=1.17,95%CI(1.13,1.20),P<0.001],relieved patients' pain[MD=-1.56,95%CI(-1.89,-1.22),P<0.001],and improved patients'dysfunction to a certain extent[MD=4.84,95%CI(3.87,5.82),P<0.001]. Funnel plot analysis showed that the included literature may be potentially biased in publication. Conclusion:Governor meridian moxibustion has a significant effect on the treatment of lumbar disc protrusion and is worthy of widespread use. However,given the small number of high-quality literature included in the study,the small sample size,and the short follow-up time,high-quality randomized controlled trials are needed to further study the long-term therapeutic effect of governor meridian moxibustion.

    查看更多>>摘要:Tic disorder is one of the common neuropsychiatric disorders that affect children's physical and mental health. If it is not tested, diagnosed and treated early, it will have a serious impact on children themselves, their families, and even the whole society. Simple western medicine treatment methods include drugs, nerve regulation, psychological and behavioral therapy, but the results are not ideal. Traditional Chinese medicine takes systematic treatment based on syndrome differentiation and it uses methods of traditional Chinese medicine, massage and acupuncture to regulate five viscera and six bowels in order to harmonized Yin and Yang. These methods make up for the deficiencies of western treatment. In recent years, the clinical treatment of children with Tic disorders involving a variety of combined therapy has achieved significant results. As a result, the rehabilitation of children with Tic disorders has been significantly improved. By consulting the available literature at home and abroad in recent years, the author elaborates the etiology, pathogenesis and treatment of tic disorder in children, in order to further guide the clinical diagnosis and treatment.
