    New progress of novel COVID-19 variants and its effect on vaccine immune protection

    查看更多>>摘要:The COVID-19 pandemic caused by severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) poses a serious threat to human life and health as well as social and economic development. In order to deal with this public health event, scientific research institutions around the world have rapidly developed and put vaccines into urgent use, bringing hope to the victory over the epidemic. However, as the Novel Coronavirus continues to spread throughout the world, the virus genome has mutated to form a variety of variants. Among them, the Alpha, Beta, Gamma, Delta and Omicron variants show higher infectivity and higher resistance to vaccines and neutralizing antibodies, posing new challenges to the prevention and treatment of COVID-19. At present, the effect of variants on the effectiveness of developed vaccines has become a hot topic of global discussion. In this paper, we briefly review the new progress of novel Coronavirus variants and their effects on vaccine immune protection.

    Resveratrol ameliorates diabetic myocardial injury through HSF1-mediated ferroptosis

    查看更多>>摘要:Objective: To observe the effect of resveratrol on the injury of diabetic cardiomyocytes and its effect on HSF1 mediated iron death. Methods: the diabetic cardiomyopathy model was established by high glucose induced H9c2, and H9c2 was exposed to normal glucose concentration as a control. Then the intervention was performed with the corresponding drugs. The cell proliferation level was detected by CCK8 method, the concentrations of LDH, SOD, MDA and iron ions were detected by kit method, and the expression levels of HSF1, apoptotic proteins (Bax and Bcl-2) and iron death marker proteins (ACSL4, GPX4 and SLC7A11) were detected by Western blot; Results: compared with the blank group, the cell activity, SOD activity, the expression of HSF1, Bcl-2, GPX4 and SLC7A11 protein in the model group decreased significantly, and the LDH activity, MDA content, Bax and ACSL4 protein expression, Bax/Bcl-2 ratio and Fe2+ content increased significantly (P<0.01). Compared with the model group, the activity of H9c2 cells, the activity of SOD, the expression of HSF1, Bcl-2, GPX4 and SLC7A11 protein increased significantly, and the activity of LDH, the content of MDA, the expression of Bax and ACSL4 protein, the ratio of Bax/Bcl-2 and the content of Fe2+ decreased significantly in the resveratrol group (P<0.01). After the intervention, the activity of H9c2 cells, the activity of SOD, the expression of HSF1, Bcl-2, GPX4 and SLC7A11 protein decreased significantly, and the activity of LDH, the content of MDA, the expression of Bax and ACSL4 protein, the ratio of Bax/Bcl-2 and the content of Fe2+ increased significantly in si-hsf1 group (P<0.01); Conclusion: resveratrol can inhibit cell iron death and improve high glucose induced cardiomyocyte injury by up regulating the expression of HSF1.

    查看更多>>摘要:Objective: To investigate the effects of Qishen Yiqi dropping pills serum on KATP channel opening and PI3K/AKT signaling pathway of hypoxic/reoxygenated H9C2 cardiocytes. Methods: H9C2 cardiocytes cultured in vitro were randomly divided into five groups, A: H9C2 cell group B: H9C2 cells +H2O2 model group C:H9C2 cells +H2O2 model + Qishen Yiqi group D:H9C2 cells +H2O2 model + Qishen Yiqi +wort group E:H9C2 cells +H2O2 model + Qishenyiqi +5-HD group, the drug intervention is according to the corresponding conditions. CCK-8 method was used to detect the cell activity of each group; Western blot was used to detect the expression of AKT and P-Akt proteins in myocardial cells in each group. The current was recorded by the standard patch clamp whole cell recording method, and the current was collected and analyzed by Pclamp6.0 software. Results: CCK-8 test results showed that compared with group A, the activity of myocardial cells in group B was significantly decreased, and the difference was statistically significant (P<0.01); compared with group B, the difference in group C was statistically significant (P<0.01); compared with C, cardiomyocyte activity in D and E group were significantly decreased, and the difference was statistically significant (P<0.05); WB results showed that compared with A, p-Akt protein expression in B, C, D and E groups were significantly decreased, and the difference was statistically significant (P< 0.01); compared with group B, p-Akt protein expression in C, D and E group were significantly increased, and the difference was statistically significant (P< 0.01),but there was no significant difference in AKT expression among groups (P>0.05); The results of whole cell patch clamp experiment showed that the outward current of B was significantly increased compared with that of A, and the difference between groups was statistically significant (P<0.01); compared with group B, cardiomyocytes in group C further increased the outward current, and the difference between groups was statistically significant (P<0.01); compared with C, the current of D and E group were significantly decreased, with statistical significance between groups (P<0.01). Conclusion: QishenYiqi dropping pills can protect cardiomyocytes by activating p-Akt protein expression and KATP channel opening in H9C2 cardiomyocytes.

    Experimental study on the effect of cryoablation on lung cancer mice based on MAPK/ERK signaling pathway

    查看更多>>摘要:Objective: To study the regulatory effect of cryoablation on MAPK/ERK signaling pathway in mice with lung adenocarcinoma. Methods: Lewis mouse lung adenocarcinoma cell line was used to establish subcutaneous transplanted tumor model in C57BL / 6 mice. Ten mice were randomly divided into two groups: sham operation group and cryoablation group, with 5 mice in each group. The cryoablation group was treated with double circulation-rewarming ablation, and the sham operation group was treated with incision and suture at the transplanted tumor. The tumor tissues were taken 14 days after operation. Detect the effect of cryoablation on MAPK/ERK pathway related proteins by Western blot, such as KRAS, RAF1, MEK1, ERK1/2, P-RAF1, P-MEK1, P-ERK1/2. The expression of KRAS gene was further verified by qRt-PCR. Results: Compared with the sham operation group, the phosphorylated proteins P-RAF1, P-MEK1 and P-ERK1/2 in tumor tissue after cryoablation were decreased (P< 0.05 ), and the key molecule KRAS in MAPK/ERK pathway was decreased in protein and gene expression (P< 0.05 ). Conclusion: Cryoablation can negatively regulate MAPK / ERK signaling pathway by down-regulating KRas expression.

    Effect of pestle intervention in type 2 diabetic peripheral neuropathy on Keap1 / Nrf2 / ARE pathway and the relationship with oxidative stress

    查看更多>>摘要:Objective: To investigate the effect of pestle needle treatment on Nrf2 pathway and the relationship with oxidative stress in diabetic peripheral neuropathy. Methods: Patients with DPN who met the inclusion criteria were randomly divided into control and test groups with 30 patients in each group in a 1:1 allocation ratio. Both groups were given basic treatment, and the pestle group was treated with needle pestle therapy 5 times a week for a total of 4 weeks of intervention. Serum SOD and GSH PX levels were examined by colorimetry before and after intervention; Serum Keap1/Nrf2/ARE signaling pathway related factors expression levels were measured by ELISA; Keap1 and Nrf2 mRNA expression was determined by RT-PCR. Results: Compared with the control group, SOD and GSH-Px in the test group were significantly increased, Keap1 expression was decreased, Nrf2 expression was increased, Keap1 mRNA expression was significantly decreased, and Nrf2 mRNA expression was significantly increased. Conclusions: the pestle needle may enhance the body's antioxidant capacity by modulating the Keap1/Nrf2/ARE signaling pathway to enhance the production of its downstream antioxidant enzymes SOD and GSH Px, thereby protecting and repairing the damaged peripheral nerves in DPN patients.

    Network pharmacological study and molecular docking verification of Capparis spinosa in the treatment of systemic sclerosis

    查看更多>>摘要:Objective: To explore the molecular mechanism of Capparis spinosa in the treatment of systemic sclerosis ((SSC)) based on network pharmacology. Methods: GEO, Genecards, Pharmgkb, TTD and Drugbank databases were used to obtain SSC targets, related literatures and Swisstargetprediction databases were used to obtain the main components of Citrus and their corresponding targets, and intersection was used to obtain prediction targets. Log in to the String database to analyze the protein interaction of the prediction target(PPI), further used Cytoscape to obtain the core gene by network topology analysis, and the core gene was docked with the main components of Capparis spinosa. The prediction targets were analyzed by gene ontology (GO) analysis and Kyoto encyclopedia of genes and genomes (KEGG) pathway analysis using R software. Results: A total of 15 active components and their targets were obtained, 3171 SSC targets were obtained, and 66 predicted targets were obtained by intersection. Ten PPI core genes such as VEGFA, TNF, AKT1, PTGS2 and MMP9 were obtained by topological analysis. GO analysis involved many biological processes such as reactive oxygen species metabolic process、protein kinase B signaling、regulation of inflammatory response、phosphatidylinositol 3-kinase signaling and so on. KEGG pathway analysis showed PI3K-Akt signaling pathway, Proteoglycans in cancer, Focal adhesion, Rap1 signaling pathway and other signaling pathways. Conclusion: The molecular mechanism of Capparis spinosa in the treatment of SSC is predicted by the method of network pharmacology, which provides theoretical basis and data support for the basic research of Citrus officinalis in the treatment of SSC.

    Research on the possible molecular mechanism of Fructus cnidii to improve sleep based on network pharmacology

    查看更多>>摘要:Objective: To study the mechanism of cnidium cnidium improving sleep by network pharmacology. Methods: The active components of Fructus cnidii and the targets of active components were obtained by systematic pharmacology database and analysis platform (TCMSP).With the help of Uniprot database;the corresponding target genes of active components were searched, and the targets related to insomnia were retrieved in drugbank and GeneCards databases.Cytoscape 3.7.2 was used to construct the hypnotic active component-insomnia - target network.String database to build PPI network;On the basis of Metascape database, GO enrichment analysis and KEGG enrichment analysis were carried out to predict the mechanism of action. Results: A total of 19 active compounds such as geranyl lignin, osthole, and omellae were screened from Osthole, 55 effective targets were predicted, 962 insomnia related targets were screened, and 27 intersection targets for drug insomnia were made.Among them, PTGS1, PIK3CG, GABRA1, CHRM1, CHRM3, ESR1, etc., may be the key targets of cnidii to improve insomnia. Biological function and pathway enrichment analysis showed that the main active components of Cnidium were involved in chemical synaptic transmission, G-protein-coupled receptor signaling pathway, membrane potential regulation, neurotransmitter transport, monoamine transport, ion transport regulation and other biological processes.Sleep regulation was most likely by regulating Neuroactive ligand-receptor interaction, signaling pathway, Cholinergic synapse, gMP-PKG signaling pathway, it addiction, Serotonergic synapse, and Dopaminergic synapse. Conclusion: The network pharmacology analysis shows that fructus cnidii can improve the characteristics of sleep multi-component, multi-target and multi-pathway, which provides a certain theoretical basis for the research of mechanism of fructus cnidii regulating sleep.

    Study on the mechanism of Shenling Baizhu Powder in the treatment of diarrhea-type irritable bowel syndrome based on network pharmacology and molecular docking

    查看更多>>摘要:Objective: Explore the possible intervention mechanism of Shenling Baizhu Powder on IBS-D from network data through network pharmacology and molecular docking simulation technology. Methods: Establish a complete regulatory network of Shenling Baizhu powder on IBS-D after obtaining the active components and drug targets of Shenling Baizhu Powder on the TCMSP platform and obtaining the disease targets of IBS-D from GeneCards and OMIM databases. Then screen out the potential best target of Shenling Baizhu Powder in IBS-D intervention network combining with the PPI network constructed in the STRING database, and screen the potential core of compounds through the molecular docking simulation and comparison with pivecurium bromide. After that,the related gene ontology functions and pathways of Shenling Baizhu Powder in treating IBS-D will be obtained though gene ontology(GO) enrichment analysis through DAVID database and Kyoto encyclo-pedia of genes and genomes(KEGG) pathways enrichment analysis through Bioconductor database. Results: The number of IBS-D disease-related targets was 2,724. And there were 184 active compounds in Shenling Baizhusan, among which 175 could act on IBS-D. After screening, four potential best targets of PTGS2, ESR1, AR and MAPK14 were obtained, and four core compounds of quercetin, luteolin, kaempferol and isorhamnetin were obtained by molecular docking simulation.GO enrichment analysis results showed that there were 92 biological processes, 18 cellular components and 21 molecular functions associated with shenling Baizhu Powder intervention IBS-D process. The results of KEGG pathway enrichment analysis showed that shenling Baizhu Powder had 172 pathways involved in the intervention of IBS-D. Conclusion: The intervention effect of Shenling Baizhu powder on IBS-D was better than pivecurium bromide in terms of the number of targets and the ease of binding to the targets.Its intervention process of IBS-D may be closely related to inflammation, cancer, endocrine disorders and other related reactions and pathways, among which quercetin, luteolin, kaempferol and isorrine are the most potential core compounds in the intervention network, and the four target proteins of PTGS2, ESR1, AR and MAPK14 are the potential best target options in the intervention network.

    Meta-analysis of effectiveness and safety of Chinese herbal medicine treatment for obese patients

    查看更多>>摘要:Objective: To systematically review the efficacy and safety of traditional Chinese medicine for weight loss. Methods: We used standard methodologies to search, review, analyse and synthesize published data on the efficacy, safety. Search randomized controlled trial(RCT) from CNKI、WanFang Data,VIP Data, Sinomed,The Cochrane Library,Pubmed,MEDLINE and Web of Scinece.Two reviewers independently screened literature,extracted data and assessed the risk of bias of included trials according to the Cochrane handbook. Then use review manager5.3 completed meta-analysis. Results: A total of 16 RCT were included.The results of meta-analysis showed that compared with placebo or other anti-obesity medicine, CHM is effective and safe in reducing theweight[MD=-1.99,95%CI(-2.94,-1.02),p<0.0001], BMI[MD=-0.78,95%CI(-1.02,-0.54), p<0.00001],waist circumference[MD=-2.71,95%CI(-3.81,-1.59), p<0.00001],fat mass[MD=-1.23,95%CI(-2.37,-0.18), p=0.02],fat mass percentage [MD=-2.71,95%CI(-3.81-1.59), p<0.00001] of patients.There were no significant changes in glucose and lipid metabolism. 6 RCTs reported adverse events (gastrointestinal reactions and headaches), 18 cases (1.40%) in the treatment group and 7 cases (0.54%) in the control group; no serious adverse reactions were seen. Conclusion:Chinese herbal medicine has define effect and safety, but that is uncertain about inregulating the glucose and lipid metabolism, and more high-quality literature is needed for further verification.

    Systematic review of Shufeng Jiedu capsules for acute attack of chronic bronchitis

    查看更多>>摘要:Objective: To systematically evaluate the efficacy and safety of Shufeng Jiedu Capsules in the treatment of acute attacks of chronic bronchitis. Methods: CNKI, VIP, CBM, WanFang, PubMed, EMbase, Cochrane Library and ChiCTR were searched by computer, and randomized controlled trials of Shufeng Jiedu capsules in the treatment of AECB were integrated.The retrieval time was from the date of establishing the database to December 31, 2020. Meta-analysis of outcome indicators [including effective rate, C-reactive protein (CRP), lung function [FEV1% and/or FEV1/FVC], interleukin-8 (IL-8), tumor necrosis factor-α (TNF-α)] were carried out by RevMan 5.3 software, and the effective rate was tested by sequential analysis (TSA), and the quality of evidence was evaluated according to GRADE standards. Results: A total of 680 cases in 8 articles were included. Shufeng Jiedu Capsules improved the effective rate of clinicaltreatment [RR=1.20, 95%CI (1.13, 1.28)], improved lung function [FEV1:MD=0.33, 95% CI (0.22, 0.45), FEV1/FVC:MD=10.17,95%CI(8.15,12.19) ],reduced CRP[MD=-7.32, 95%CI(-8.42,-6.22)], IL-8[MD=-63.39, 95%CI(-73.49,- 53.29)], TNF-α[MD=-7.44, 95%CI (-8.35, -6.53)] levels. The above differences were statistically significant (P<0.05), and no serious adverse reactions were reported in all studies.The results of TSA analysis showed that the experimental group had definite evidence for improving the efficiency. According to the GRADE evaluation system, the efficiency, TNF-αwere medium-quality evidences, FEV1 was low-quality evidence, and FEV1/FVC, CRP, and IL-8 were extremely low-quality evidence. Conclusions: The curative effect of Shufeng Jiedu capsule combined with western medicine in the treatment of AECB was better than that of single western medicine. Nevertheless, considering the limited sample size and the quality of included articles,higher quality RCTs are still needed to further confirm its effectiveness and safety.
