    The latest research progress of the Corona Virus Disease 2019 mutant strain "Omicron"

    查看更多>>摘要:Coronavirus disease 2019 (COVID-19) that broke out at the end of December 2019 has been raging for 3 years, bringing unpredictable harm to the physical and mental health of all mankind and global economic development. The new type of coronavirus pneumonia is a new type of respiratory infectious disease with a high incidence and fatality rate caused by SARS-CoV-2. Up to now, hundreds of millions of people have been infected with new coronary pneumonia worldwide, and millions of people have died. Due to the specificity of the new coronavirus itself and its high mutation rate, a series of different new coronavirus variants have appeared, which has caused the new crown pneumonia epidemic to repeat. Recently, the World Health Organization (WHO) announced a new variant "omicron" (omicron, B.1.1.529), and declared that the mutant strain may be highly infectious, antibody tolerant and highly resistant to vaccines. This article briefly reviews the latest research progress of the "omicron" variants.

    Neuroprotective effect of Phyllanthus urinaria extract on ischemic brain injury in rats

    查看更多>>摘要:Objective: To investigate the neuroprotective effect of Phyllanthus urinaria on ischemic stroke and its primary mechanism. Methods: Adult SD rats were selected as the research object, and the right middle cerebral artery infarction rat model was established by the modified suture method(MCAO).Observe the neurological deficit score at 24h, 48h and 72h after the model is successfully prepared,then TTC staining method to detect the area of cerebral infarction, the content of superoxide dismutase (SOD), nitric oxide (NO) and endothelial NOS (eNOS) in brain tissue;Immunofluorescence method was used to detect the expression of Caspase-3 positive cells in brain tissue; Western blot method was used to detect the expression of PI3K and AKT protein in brain tissue.72 experimental animals were randomly divided into 4 groups, sham operation group, model group (MCAO), extract of Phyllanthus urinaria low dose group(PuL5 g/kg), high dose group (PuL10 g/kg), and make MCAO model after 7 days of continuous administration, and continue to infuse the medicine once/d until the material is obtained. Results: No neurological deficits in the sham operation group, The 24h, 48h and 72h of the modelling showed that the neurological impairment of the two doses of extract of Phyllanthus urinaria and the MCAO group was more severe than that of the sham operation group (P<0.01),however, with the prolongation of the modeling time, the neurological function scores of the two doses of extract of Phyllanthus urinaria were lower than those of the MCAO group, the most significant at 72h (P<0.01);The infarct size of the rats in the two dose groups of extract of Phyllanthus urinaria was lower than that of the MCAO group (P<0.01), and there was no dose dependence between the two groups,the content of SOD in the MCAO group was reduced, and the content of NO and eNOS was increased than the sham operation group(P<0.05),Compared with the MCAO group, the two administration groups significantly increased the content of SOD, decreased the content of NO and eNOS (P<0.05);Although the expression of Caspase-3 positive cells in the two administration groups was higher than that in the sham operation group, it was significantly lower than that in the MCAO group (Plow<0.05, Phigh<0.01);The expression of PI3K and AKT protein in the brain tissue of the MCAO group was significantly lower than that of the sham operation group (P<0.05),but the expression of PI3K and AKT protein in brain tissue of extract of Phyllanthus urinaria in high and low dose groups was significantly higher than that in MCAO group (P<0.05). Conclusion: Extract of Phyllanthus urinaria can improve neurological damage and cerebral infarction area in rats, and reduce the expression of Caspase-3 positive cells in MCAO rats, and then increase the expression levels of PI3K and AKT proteins to protect ischemic brain injury.

    Effect of Wenxin granule on Bcl-2/Bax/Caspase apoptosis pathway gene expression in rats with myocardial infarction

    查看更多>>摘要:Objective: To explore the cardioprotective mechanism of Wenxin Granules regulating the expression of apoptosis-related genes in cardiomyocytes. Methods: A rat model of myocardial infarction was established and randomly divided into model group, Wenxin granule low-dose group, Wenxin granule high-dose group, metoprolol group and sham operation group. the left ventricular end systole anterior wall thickness(LVAWs), end systole inner diameter (LVIDs), end systole posterior wall thickness (LVPWs), end-diastolic anterior wall thickness (LVAWd), end-diastolic inner diameter (LVIDd), end-diastolic posterior wall thickness(LVPWd)and left ventricular ejection fraction (LVEF) were detected by echocardiography in each group after 2 weeks of treatment. Hematoxylin eosin (HE) staining was used to observe the changes in the cardiac structure of rats in each group. Real-time PCR (Real-time PCR) was used to detect the relative expression of mammalian B-cell lymphoma-2 (BCL-2), BCL-2 related X protein (BAX), Caspase-9 (Caspase-9), and Caspase-3 (Caspase-3) mRNA. TUNEL staining was used to detect changes in the apoptotic rate of rat cardiomyocytes in each group. Results: Compared with the sham operation group, the LVAWs, LVPWs, LVPWd and LVEF of the model group were significantly reduced (P<0.05, P<0.01), and LVIDs and LVIDd were significantly increased (P<0.05, P<0.01). Severe pathological ischemia injury of heart tissue. The relative expression of BCL-2 mRNA and the ratio of BCL-2/BAX in the model group were significantly reduced (P<0.01), while the relative expression of BAX, Caspase-9 and Caspase-3 mRNA was significantly increased (P<0.01). The apoptosis rate was significantly increased (P<0.01). In the low-dose and high-dose groups of Wenxin Granules and the Metoprolol group, LVAWs, LVPWs, LVPW d, and LVEF of rats in each administration group increased significantly (P<0.05, P<0.01), LVIDs , LVIDd was significantly reduced (P<0.05, P<0.01), the pathological damage of the heart tissue was improved, the expression of BCL-2 mRNA and the ratio of BCL-2/BAX were significantly increased (P<0.05, P<0.01), BAX, The expression of Caspase-9 and Caspase-3 mRNA was significantly reduced (P<0.05, P<0.01), and the apoptotic rate of myocardial cells was significantly reduced (P<0.01). Conclusion: Wenxin granule can play a cardioprotective role by regulating the gene expression of BCL-2/BAX/Caspase apoptosis pathway.

    Effect of Zhiyang Pingfu Liquid on epidermal growth factor receptor inhibitor-related skin lesion

    查看更多>>摘要:Objective: To verify the efficacy of the Chinese medicine "Zhiyang Pingfu Liquid" on the lesion associated with Epidermal Growth Factor Receptor Inhibitors (EGFRIs). Methods: Female BN rats were divided into Control group and Gefitinib group randomly. The Gefitinib group was administered gefitinib for 21 days. After 21 days, the rats in the Gefitinib group were grouped again and randomly divided into Model group, Gefitinib + ZY group, and Gefitinib + NS group. Starting from day 22, rats in Gefitinib+ZY or NS were given different drugs for 7 days besides the other conditions are as the same as before. Observe the morphological changes and histopathological changes of the skin during the research. The changes of inflammatory factors such as TNF-α and IL-6 in the serum of were detected by ELISA. Results: The application of "Zhiyang Pingfu Liquid" for 7 days could significantly reduce the skin inflammation whether in gross or pathological view. The concentration of TNF-α and IL-6 in Gefitinib+ZY is significantly lower than those in the Model group (P=0.002, P=0.002) and there is no significant changes compared with the Control group (P=0.279, P=0.165). Conclusion: Chinese herbal "Zhiyang Pingfu Liquid" can reduce the lesion and inflammatory caused by EGFRIs.

    Box-Behnken response surface method combined with fingerprint to optimize the extraction process of total anthraquinone from Cassia seeds

    查看更多>>摘要:Objective: The Box-Behnken response surface method combined with fingerprints was used to optimize the extraction process of total anthraquinone from Cassia seeds. Methods: A three-factor, three-level response surface test was conducted based on the single-factor test with comprehensive evaluation as the measurement index. The comprehensive evaluation indexes included the extraction rate of total anthraquinone of Cassia seeds or the equivalent amount of herbs per gram of total anthraquinone of Cassia seeds, the normalized value of peak areas of 5 index components such as aurantio obtusin in the fingerprint of each sample to 16 shared peaks and the similarity of fingerprints (the reference fingerprint was established by the extraction solvent for the determination of Cassia seeds content in the Chinese Pharmacopoeia 2020 edition) with the weights of 0.2, 0.5 and 0.3, respectively. Results: The best extraction process was obtained: the liquid-to-material ratio was 20:1 (mL g-1), the extraction solvent was mixture of 60% ethanol-ethyl acetate (2:1), and the extraction time was 15.12 min. The results of five sets of validation experiments showed that the overall evaluation of total anthraquinone of cassia seeds by the best process was 0.528 (RSD=0.45%), and the prediction result of response surface method model was 0.531, and the relative error with the prediction result was 0.531. The relative error of the predicted results was 0.56%, and the best extraction process was consistent with the model prediction, and the obtained best process could be used for the extraction of total anthraquinone from Cassia seeds. Conclusion: The Box-Behnken response surface method combined with the fingerprint technique can be used to find the best reaction conditions and examine the interactions among the factors in a comprehensive and accurate manner, which can provide reference for the optimization and evaluation of the extraction process of Chinese medicine.

    Design and characterization of a bifunctional bybrid antibacterial peptide LLH for bactericidal/endotoxin neutralization effects

    查看更多>>摘要:Objective: To design a bi-functional antimicrobial peptide with bactericidal and endotoxin neutralization activity, and explore its bactericidal properties. Methods: The LBP (86-99) peptides and HLF (1-11) peptides were connected by a GGGS flexible 4-peptide linker to obtain the bi-functional antimicrobial peptide, which was named LLH. The secondary structure characteristics of LLH were analyzed by Emboss software. The minimum inhibitory concentration (MIC) and minimum bactericidal concentration (MBC) of LLH against Escherichia coli ATCC25922 and DH5 were determined by the microtiter broth dilution method. The bactericidal kinetics of LLH was characterized and its effect on endotoxin neutralization was determined. The hemolysis of LLH was evaluated. Results: LLH carried a positive charge of +9, exhibited β-folding and β-corner structure, and had strong hydrophobicity. The MIC of LLH against Escherichia coli ATCC25922 and DH5 was 4μM, and the MBC of LLH against Escherichia coli ATCC25922 and DH5 was 8 and 4μM, respectively. LLH showed rapid bactericidal effects and significantly neutralized the endotoxin released in the sterilization process as well as reducing release of endotoxin. LLH showed no significant hemolysis at concentrations up to 400μg/ mL. Conclusion: LLH produces dual effects of rapid sterilization and endotoxin neutralization, and does not induce significant hemolysis.

    Effect of Xifeng Capsule on blood stasis in patients with rheumatoid arthritis by regulating miR-126-VEGF/PI3K/AKT signaling pathway

    查看更多>>摘要:Objective: From the perspective of miR-126-vascular endothelial cytokine (VEGF)/phosphatidylinositol 3-kinase (PI3K)/ser-threonine protein kinase (AKT) signaling pathway, Xinfeng Capsule (XFC) can improve rheumatoid arthritis (RA) ) The mechanism of the patient's blood stasis state. Methods: Sixty RA patients meeting the diagnostic criteria were selected and divided into XFC treatment group 30 cases and Leflunomide (LEF) control group 30 cases according to the random number table method. The treatment group took Xinfeng Capsules (3 capsules each time, 3 times/d), and the control group took Leflunomide (1 capsule each time, 1 times/d). Observe the blood stasis symptom scores of RA patients, and detect the laboratory indicators erythrocyte sedimentation rate (ESR), c-reactive protein (CRP), rheumatoid factor (RF), anti-cyclic citrullinated peptide antibody (CCP), thrombin time (TT), part prothrombin time (APTT), prothrombin time (PT), D dimer (DD), fibrinogen (FBG), platelets (PLT), mean platelet volume (MPV), platelet distribution width (PDW) levels. Real-time fluorescent quantitative PCR method was used to detect the level of miR-126, and the ELISA method was used to detect the levels of tumor necrosis factor (TNF-α), interleukin-6 (IL-6), IL-35, VEGF, PI3K, and AKT. Spearman method was used to analyze the correlation between the total score of blood stasis symptoms, blood stasis-related indicators and disease activity indicators, cytokines, miR-126, VEGF, PI3K, and AKT in RA patients. Results: Comparing the two groups after treatment, the XFC group improved blood stasis symptoms, disease activity indicators, decreased miR-126, VEGF, PI3K, AKT, TNF-α, IL-6, D-D, FBG, PLT, and increased IL-35 The level was significantly better than the LEF group, with statistical significance (P<0.05, P<0.01). Correlation analysis showed that there was a certain correlation between the total score of blood stasis symptoms, blood stasis related indicators and disease activity indicators, cytokines, miR-126, VEGF, PI3K, and AKT in RA patients. Conclusion: There is blood stasis in RA patients. XFC may improve the cytokine network disorder of patients through miR-126-VEGF/PI3K/AKT signaling pathway, thereby improving the blood stasis status of RA patients.

    Efficacy and safety of traditional Chinese medicine in the treatment of coronary heart disease complicated with anxiety and/or depression after PCI: A systematic review and meta-analysis

    查看更多>>摘要:Objective: The clinical efficacy and safety of traditional Chinese medicine in the treatment of anxiety and/or depression after PCI for coronary heart disease were systematically evaluated. Methods: The databases of CNKI, Wanfang, VIP, CMB, PubMed, Web of Science, PubMed, Web of Science and Cochrane Library were searched by computer, and the Chinese and English randomized controlled trials(RCTs)of traditional Chinese medicine in the treatment of anxiety and/or depression after PCI of coronary heart disease published from the database to November 2020 were included. The Cochrane bias risk tool and the modified Jadad scale were used to evaluate the quality of the included literature, and the RevMan 5.3 and Stata 16.0 software were used for Meta-analysis. Results: A total of 23 RCTs were included, including 1837 subjects, including 917 cases in the treatment group of traditional Chinese medicine, 920 cases in the control group, the baseline situation between groups was consistent, comparable. Meta-analysis results show that TCM treatment improves the total clinical effective rate[OR=2.34,95%CI(1.61,3.40),P<0.00001],and the efficacy of TCM syndrome[OR=5.19,95%CI(2.74,9.86),P<0.00001],reduce Hamilton anxiety scale HAMA score[SMD=-0.96,95% CI(-1.78,-0.14),P=0.02],Hamilton Depression Scale HAMD score[SMD=-1.10,95% CI(-1.79,-0.41),P=0.002],SAS self-rating scale score[SMD=-1.20,95% CI(-1.75,-0.65),P<0.0001],SDS self-rating scale score[SMD=-1.14,95% CI(-1.62,-0.65),P<0.00001] and other indicators are better than the control group, and the difference is statistically significant (P<0.05). Conclusion: The combination of traditional Chinese medicine intervention and conventional western medicine treatment after PCI can further improve the clinical efficacy of patients with anxiety and/or depression after PCI of coronary heart disease and has good safety. Due to the general quality of the literature included in this study, the above conclusions need to be further confirmed by high-quality randomized controlled double-blind clinical trials.

    Study on medication rules of traditional Chinese medicine for Meniere's disease based on data analysis

    查看更多>>摘要:Objective: Study on medication rules of traditional Chinese medicine in treating Meniere 's disease based on data mining technology. Methods: Computer retrieval with since establishment of the"CNKI", "WF", "VIP", there have been literatures on the treatment of Meniere syndrome with Traditional Chinses medicine. Memory preprocessing in accordance with inclusion criteria. Then, EXCLE 2010, SPSS Statistics (ver. 25) and SPSS Modeler (ver. 18.0) were adopted respectively for frequency analysis, cluster analysis and association rules analysis. Result: A total of 133 references were included in this study, 146 prescriptions, 192 kinds of drugs, and the total frequency of drugs was 1702. The high frequency Meniere syndrome types were spleen deficiency and phlegm dampness syndrome, wind phlegm syndrome, gallbladder depression and phlegm disturbance syndrome, hyperactivity of liver yang syndrome, phlegm and blood stasis syndrome. The efficacy of drug frequency 25 were summarized as follows, antiasthmatic drugs for relieving phlegm and relieving cough, diuresis-removing dampness, Tonic drugs, etc. High frequency meridian of drugs are: lung, liver, spleen, etc. High frequency drug properties are: temperature, cold, flat etc. High frequency drug taste: bitter, sweet, pungent, etc. Core drugs: Pinellia, Atractylodes, Poria, Gastrodia, etc. The main prescriptions werealisma soup, Banxia Baizhu Tianma Decoction, Zhengan Xifeng Decoction and Xiaochaihu Decoction. Conclusion: In this study, data mining was used to sort out the treatment of Meniere ' s disease by traditional Chinese medicine. It was found that the treatment of Meniere ' s disease was mainly to calm the liver and strengthen the spleen, supplemented by regulating qi and activating blood, clearing heat and opening orifices, tonifying deficiency and tranquilizing mind.

    查看更多>>摘要:Objective: To dig deeper into the traditional Chinese medicine treatment rules of chemotherapy-induced diarrhea with traditional Chinese medicine (TCM) inheritance computing platform system. Methods: Taking "traditional Chinese medicine", "chemotherapy" and "diarrhea" as the theme words, comprehensive search of the database of CNKI, Wanfang and VIP from its inception to November 2020 was conduct-ed. The formulas of external treatment of traditional Chinese medicine for chemotherapy-induced diarrhea were included and the association rule, clustering and factor analysis were carried out. Results: A total of 145 papers, 57 prescriptions meeting the inclusion criteria were collected, among which high-frequency drugs including Baizhu (Atractylodis macrocephalae rhizoma), Fuling (Poria), Dangshen (Codonopsisradix), Huanglian (Coptidis rhizoma), Zhigancao (Glycyrrhizae radix et rhizoma praeparata cum melle) were the most commonly used. The confidence level was set as 0.7 and the support level was set as 10, 12 core compatibility groups were obtained, and 6 categories were cluster analyzed. Conclusion: The principle of external treatment of chemotherapy-induced diarrhea is mainly "restore deficiency and benefiting qi", "benefiting water infiltration and dampness", "clearing heat" and "inducing astringency". Prescription combination and new prescription combination based on traditional Chinese medicine (TCM) inheritance computing platform system can be used as reference for clinicians and applied in primary hospitals.
