    Research on the Orientated Effective Components of Huangqi in Huangqi Jianzhong Tang Against Chronic Atrophic Gastritis Based on Multi-Spectrum-Effect Correlation

    查看更多>>摘要:Objective:This study aimed to investigate the orientated effective components of Astragali Radix Huangqi(HQ)in HQ Jianzhong Tang(HQJZ),a classical formula of traditional Chinese medicine(TCM)used for treating chronic atrophic gastritis(CAG),using HQ as a monarch medicine.Materials and Methods:The spectra of HQJZ containing different polar parts of HQ were obtained using ultra-high-performance liquid chromatography-Q-Exactive mass spectrometry.Furthermore,the efficacy of HQJZ,which contains different polar parts of HQ,in treating rats with CAG was evaluated using traditional pharmacodynamic and nuclear magnetic resonance-based metabonomics.Grey relation analysis and partial least squares analysis were applied to analyze the spectrum-effect relationship and to screen out the orientated effective components related to HQ in the treatment of CAG.Results:Spectrum-effect relationship analysis showed that 24 compounds identified from the fingerprint spectrum were strongly correlated with efficacy.Compounds 8(calycosin-7-O-glc-6"-O-acetate),9(3-hydroxy-9,10-dimethoxyptercarpan),and 22(astragaloside II)were ranked among the top three.Conclusions:This study showed that integrating metabolomics and spectrum-effect relationship analysis is a powerful tool for obtaining orientated effective components of Chinese medicine in a given TCM formula.

    查看更多>>摘要:Objective:Radix Bupleuri and Radix Paeoniae Alba are referred to as Chai-Hu and Bai-Shao,respectively,in Chinese.We used molecular docking and network pharmacological approaches to explore the active components of this Chinese herb combination which is commonly used for treating Alzheimer's disease(AD),the Chai-Hu and Bai-Shao pair(CBP).Materials and Methods:The active compounds and action targets of Chai-Hu and Bai-Shao were identified in the Traditional Chinese Medicine Systems Pharmacology database,and the Therapeutic Target Database,DrugBank,GenCards,DisGeNET,and Online Mendelian Inheritance in Man databases were used to identify therapeutic targets in patients with AD.To identify a common target of CBP in the treatment of AD,we used the Metascape tool to perform Gene Ontology function enrichment and Kyoto Encyclopedia of Genes and Genomes pathway enrichment analyses.In addition,CBP-AD protein-protein interaction(PPI)and herb-component-target networks were created using Cytoscape 3.8.2.Molecular docking was verified using AutoDock tools.Results:We identified 20 active compounds of CBP,202 active targets,1,323 disease targets,and 117 active compound disease intersection targets;the PPI network showed that the top eight key targets were AKT1,interleukin(IL)6,JUN,MAPK1,tumor necrosis factor,TP53,vascular endothelial growth factor,and epidermal growth factor.These act mainly on the fluid shear stress,atherosclerosis,IL-17,hypoxia-inducible factor-1,and nonalcoholic fatty liver disease pathways,among others,which control immune inflammation,cell proliferation,and apoptosis.Based on molecular docking results,the active components of CBP bind well to its key targets.Conclusion:Our results indicated that CBP was effective for treating AD by reducing the expression of inflammatory factors,promoting cell proliferation and differentiation,and preventing nerve cell death and anti-neuronal cell apoptosis.

    Glycine Attenuates Myocardial Fibrosis in Myocardial Infarction in Rats Partly through Modulating Signal Transducer and Activator of Transcription 3/Nuclear Factor-κB/Transforming Growth Factor-β axis

    查看更多>>摘要:Objective:Inflammation and fibrosis are strongly associated with each other.Glycine is present in various traditional Chinese medicines and exhibits anti-inflammatory activity.However,the effects of glycine on myocardial fibrosis(MF)in rats with myocardial infarction(MI)have not been reported.The purpose of this study is to investigate the effects of glycine therapy on MF and comprehend its underlying mechanisms.Materials and Methods:Left anterior descending artery ligation-induced MI in Sprague Dawley rats was leveraged to assess the therapeutic effects of Glycine.Rats received either normal saline or glycine(0.5 mg/g bodyweight)for 7 days.Results:Glycine upregulated cardiac ejection fraction and fractional shortening to improve cardiac function,as evaluated by echocardiography.Histological and immunohistochemical analyses demonstrated that glycine could decrease inflammatory cell infiltration and alleviate collagen deposition.Western blotting revealed that nuclear factor-κB(NF-κB)-mediated inflammatory signaling was also downregulated by glycine treatment.The expression of signal transducer and activator of transcription 3(STAT3),tumor necrosis factor-α,and transforming growth factor-β(TGF-β)was decreased significantly in the glycine-treated group compared to the model group.Thus,glycine plays a protective role against myocardial ischemia and subsequent MF.Conclusion:The protective effects of glycine were achieved partly through STAT3/NF-κB/TGF-β signaling pathway.
