Objective:Screening efficient medium for inducing rice callus and adventitious bud differentiation culture system,laying the foundation for rice genetic engineering breeding.Methods:Using mature embryos of high-quality rice variety(Runzhuxiangzhan)as explants,studied the callus induction of mature embryos,the effects of different media and plant growth substance concentrations on callus induction and adventitious bud differentiation.Results:The callus induction rate of rice medium was N6>B5>MS>WPM,N6 was significantly different from B5、MS、WPM.The callus induction rate of 2,4-D concentration was B4>B3>B5>B6>B2>B1,significant difference between B4 and each treatment group.The ratio of plant growth substance concentration to Adventist bud differentiation of callus was T5>T7>T6>T1>T8>T3>T2>T4,significant difference between T5 and each treatment group.Conclusion:The optimal culture medium and 2,4-D concentration for inducing rice mature embryo callus are N6,2.0 mg/L.The optimal concentration of plant growth substances for adventitious bud differentiation is 6-BA,KT,and NAA at a ratio of 1∶1∶1,with the highest rate of adventitious bud differentiation.
