Objective:To identify the pathogen (Enterobacter kobei) of the stem base rot of Tongling white ginger and its corresponding bacteriophage ,and to provide scientific basis for the prevention and control of stem base rot of Tongling white ginger.Methods:E.kobei SJ16 was isolated from diseased tissues of Tongling white ginger using both TTC and LB medium ,and bacteriophage KB-P2 was isolated from rhizosphere soil using semi-solid medium.The pathogenicity of this strain to Tongling white ginger was evaluated by two infection methods (rhizome inoculation and ginger slice inoculation ).The control effect of bacteriophage was measured using the injection method.Results:A total of 103 bacterial isolates were isolated and 10 strains were obtained by LB medium screening ,among which SJ16 was identified as E.kobei.Infection experiment showed that the strain was the pathogen that caused the blight of Tongling white ginger , and could cause root rot in the later stage.In addition , three bacteriophages of E.kobei were isolated and purified ,and KB-P2 bacteriophage with the strongest cleavage ability was screened by double-layer plate culture method.The infection experiment showed that the bacteriophage KB-P2 had greater control effect.Conclusion:SJ16 was one new pathogen causing stem base rot of Tongling white ginger ,while KB-P2 could effectively reduce the incidence of stem rot of Tongling white ginger.Therefore ,this study provided an important basis for further research on the prevention and control of stem rot of Tongling white ginger.
