Objective:The effects of brassinolide(BR)on morphogenesis and physiological indexes of tissue culture seedlings of Baihuayushizi pomegranates were investigated.Methods:The effects of applying different concentrations(0,0.01,0.05,0.10,0.50 mg/L)of BR on proliferation,root growth,growth potential and physiological indices were investigated by means of tissue culture techniques.Results:After the application of 0.5 mg/L BR,pomegranate histocultured seedlings were induced to 100%,the highest multiplication factor of 9.67,the highest plant height of 6.96 cm,the heaviest fresh weight and the largest relative chlorophyll value.The rooting rate showed a gradual increase with increasing BR concentration,but the addition of BR inhibited the growth and elongation of plant roots.Superoxide dismutase activity was strongest at a BR concentration of 0.10 mg/L,peroxidase activity was strongest at a BR concentration of 0.50 mg/L,and catalase activity was strongest at a BR concentration of 0.05 mg/L,soluble sugar content was the lighest at a BR concentration of 0.50 mg/L,and soluble protein content was the highest at a BR concentration of 0.10 mg/L.BR treatment inhibited the Malondialdehyde content of pomegranate histocultures,which was significantly(P<0.05)lower than that of control in pomegranate histocultures treated with different concentrations of BR.Conclusion:BR promoted in vitro regeneration of Baihuayushizi pomegranates tissue seedling,the best results were obtained under 0.50 mg/L BR treatment,and it would affect the growth of pomegranate histocultured seedlings if the concetrocting was too low.
