Polymorphism of RIP140 gene and its relationship with litter size of Jining Grey goat
According to the Bos taurus RIP 140 mRNA sequence, five pairs of primers were designed to am plify whole CDS sequence of RIP 140 gene in five randomly selected individuals from Jining Grey goats and Liaoning Cashmere goats, respectively. Through DNA sequencing and alignment, the whole CDS sequence of the goat RIP140 gene was obtained. Meanwhile, one nucleotide transition (C822T) was identified in goat RIP140 gene. According to the CDS sequence of the goat RIP 140 gene, primer P6 was designed to detect C822T mutation of RIP 140 gene in high prolificacy breeds (Jining Grey and Guizhou White goats) and low prolificacy breeds (Liaoning Cashmere, Boer and Inner Mongolia Cashmere goats) by PCR-RFLP. The results showed that three genotypes (CC, CT and TT) were detected in Jining Grey, Boer, Liaoning Cashmere and Guizhou White goats and only two genotypes (CT and TT) were detected in Inner Mongolia Cashmere goat. In Jining Grey goat, genotype frequency of CC, CT and TT was 0.056, 0.309 and 0.635, respectively. Jining Grey goat does with genotype CC or CT had 0.81(P<0.05) or 0.65 (P<0.05) kids more than those with genotype TT, respectively. The Jining Grey goat does with genotype CC had 0.16 (P>0.05) kids more than those with genotype CT. These results preliminarily indicated that allele C of RIP140 gene is a potential DNA marker for improving litter size in goats.
