Study on Content of Ursolic Acid and Oieanolic Acid in Sambucus chinensis
A determination method of ursolic acid and oleanolic acid in leaves, stems and roots of Sambucus chinensis in various collecting periods by reversed-phase high performance liquid chromtograph was established. The chromatographic column, Symmetry Shield RP18(3.9 mm× 150 mm,5 (μm), ethanenitrile-water-phosphoric acid (65:35:0.1, V/V/V) mobile phase with the flow rate of 1.0 ml/min, the detected wavelength (210 nm) and the column temperature(25 ℃) were adopted. The calibration curve of ursolic acid was linear under the content of 0.096-2.304 μg, the correlation coefficient was over 0.999 9, the detected limit was 0.3 mg/L (3σ). The calibration curve of oleanolic acid was linear under the content of 0.098~2.352 μg, the correlation coefficient was over 0.999 7, the detected limit was 0.2 mg/L(3σ).The average recovery of ursolic acid was 100.03 % and RSD was 1.5 %(n=5). The average recovery of oleanolic acid was 98.9 % and RSD was 1.2 %(N=5). The method was simple, accurate and had wider linear range. The method can be adapted to evaluate the quality of medicinal plants that contained ursolic acid and oleanolic acid.
