Determining Aroma Genes in Rice by Using BADH Premier and Its Application in the Breeding of Fragrant Rice
[Objective] The research aimed to determine the aroma genes in rice and provide basis for its application in the breeding of fragrant rice. [Method] After DNA was extracted from backcross progeny materials BC3F2 of Thailand fragrant rice and Chinese fragrant rice, BADH premier was added for PCR amplification and determine the aroma genes by polyacrylamide gel electrophoresis. At the same time, aroma of leaf blade was identified on the individual plants contained aroma genes by using KOH solution. Two kinds of the detection results were compared. [Result] The electrophoresis results showed that among 94 samples, 32 samples contained homozygous aroma genes and 19 samples were heterozygotes of aroma genes, which was accordant with the results of determining aroma by KOH solution method. The PCR amplification results of BADH premier showed that the progeny materials that aroma rooted in Xinxiang B could produce obvious DNA amplification bands, whose aroma genes and the aroma genes in KDLM105 and Basmati fragrant rice varieties all lay on chromosome 8 and belonged to alleles. [Conclusion] In the breeding of fragrant rice varieties, using BADH premier could determine the plants contained aroma genes exactly and shorten the breeding period of fragrant rice varieties significantly.
