RAPD Analysis of Genetic Variation of Tibetan Mastiff
[ Objective ] To investigate the genetic variation of Tibetan mastiff at the molecular level and provide a basis for the protection of germplasm resources. [Methods] A total of 20 Tibetan mastiff blood samples in different regions were collected and the genomic DNA swere then analyzed using RAPD technique. The effective number of alleles (Ne) , Nei's average expected gene heterozygosity (H) and Shannon' s information index (Ⅰ) in Tibetan mastiff group were determined with population genetics software PopGene32. [ Results] A total of 369 bands were amplified from all the blood samples, in which as many as 339 bands showed polymorphism. The percentage of polymorphic loci was 91.86%. The values of Ne, H and I were 1.751 2, 0.416 2 and 0.602 1, respectively. [Conclusion] There is an abundance of genetic variations in the Tibetan mastiff group.
