摘要
[目的]探讨槲皮素氧化前后对果蝇寿命和抗氧化活性的影响.[方法]以果蝇为试材,培养基添加氧化前后的槲皮素,(25±1)℃下培养5d后转移到(33±1)℃下培养到全部死亡,期间取材测定.分别采用氮蓝四唑(NBT)法、硫代巴比妥酸(TBA)法、流动注射化学发光方法和HPLC法,测定不同天数的果蝇体内超氧化物歧化酶(SOD)活性、丙二醛(MDA)含量、羟自由基的清除率及其体内槲皮素的含量.[结果]果蝇寿命和抗氧化活性是以槲皮素、氧化槲皮素、对照的顺序递减.[结论]光照、高温和高湿的环境使得槲皮素的抗氧化活性降低.
Abstract
[Objective] The aim was to discuss the effect of inoxidized and oxygenated quercetin on life-span and the antioxidant activity.[Method] With Drosophila melanogaster as the tested material,they were placed on the medium made by quercetin and oxygenated quercetin to be cultured at (25 ± 1)℃ for 5 days.After that transfer them to (33 ± 1)℃ until death wholly and we drawn them to determine.NBT method,TBA method,the flow injection chemiluminescence method and the HPLC method were used to determine the superoxide dismutase (SOD) activity,the malondialdehyde (MDA) content,the scavenging effect of hydroxyl radical and the content of rutin in D.melanogaster on different days.[Result] In order of quercetin,oxygenated quercetin,control,the life-span and the antioxidant activity of Drosophila melanogaster were decreased.[Conclusion] Light,high temperature and high humidity environment could reduce the antioxidant activity of quercetin.
