摘要

[目的]从延边黄牛和韩延牛血液中分离Exosome(外胞体),研究2种外胞体对延边黄牛垂体细胞中GH含量的影响.[方法]通过超高速离心等方法,从延边黄牛和韩延牛血液中分离Exosome,并以100 ng/mL的浓度分别转染给原代培养的延边黄牛垂体细胞培养12、24和48 h,对细胞上清液中GH含量进行检测.[结果]电镜下延边黄牛与韩延牛血液Exosome均呈圆形或椭圆形的双层膜小囊泡,直径均为30 ~ 100 nm,膜结构完整.韩延牛Exosome处理组的延边黄牛垂体细胞GH mRNA表达显著高于延边黄牛Exosome处理组(P>0.05).韩延牛血液Exosome处理组垂体中GH的含量显著高于延边黄牛血液Exosome处理组(P>0.05).[结论]延边黄牛和韩延牛血液中的Exosome对延边黄牛垂体细胞中GH含量的影响有明显差异.

Abstract

[Objective] To study the effects of blood Exosome in Yanbian yellow cattle and Hanyan cattle on the GH content in pituitary cells of Yanbian yellow cattle.[Method] Exosome was isolated from Yanbian yellow cattle and Hanyan cattle by ultra high speed centrifugation method,and then was transfected to primary pituitary cell of Yanbian yellow cattle at a concentration of 100 ng/mL for 12,24 and 48 h,respectively.Content of GH in cell supernatant was detected by kit.[Result] Electron microscope observation showed Exosomes from Yanbian cattle and Hanyan cattle blood were both a round or oval global double membrane vesicle.Their diameters were both 30-100 nm,and membrane structure was both complete.The expression of GH mRNA in the pituitary cells of Yanbian yellow cattle treated by Hanyan cattle Exosome was significantly higher than that treated by Yanbian yellow cattle Exosome(P > 0.05).The content of GH in the pituitary of the Hanyan cattle group treated by blood Exosome was significantly higher than that of the Yanbian yellow cattle(P > 0.05).[Conclusion] GH content in pituitary cells of Yanbian yellow cattle treated by blood Exosome of Yanbian yellow cattle shows significant differences with that treated by Hanyan cattle blood Exosome.
