摘要
[目的]研究新鲜紫穗槐果实挥发油的化学成分及其抗氧化和细胞毒活性.[方法]水蒸气蒸馏法提取挥发油后,采用GC-MS法检测其成分组成;采用DPPH和β-胡萝卜素法检测挥发油的抗氧化活性,采用SRB法检测挥发油对BGC-823、A-549和HeLa等3种肿瘤细胞株的细胞毒活性.[结果]从挥发油中共鉴定出40个成分,占挥发油总量的88.65%,主要有γ-杜松烯(19.43%)、α-毕澄茄烯(10.24%)、表圆线藻烯(8.99%)和α-蒎烯(7.22%)等;紫穗槐果实挥发油对DPPH自由基清除率的IC.值为18.25 mg/mL,β-胡萝卜素-亚油酸法检测1.8 mg/mL紫穗槐果实挥发油的抑制率为25.08%;对BGC-823、A-549和HeLa等3种肿瘤细胞的细胞毒IC.值分别为37.597、46.733和48.656 μg/mL.[结论]紫穗槐果实挥发油的抗氧化活性较弱,但具有较好的肿瘤细胞毒活性.
Abstract
[Objective] To study the chemical composition,antioxidant and cytotoxic activity of the volatile oil from fresh Amorpha fruticosa fruits.[Method] The volatile oil was isolated by distillation extraction,the chemical compositions were identified by GC-MS method;the antioxidant activity of the volatile oil was assayed by DPPH radical scavenging method and β-carotene-linoleate inhibition method;cytotoxic activity to three tumor cell lines (BGC-823,A-549 and HeLa) was determined by SRB method.[Result] Forty components were identified from the volatile oil,amounted to 88.65% of the oil,γ-cadinene (19.43%),αt-cubebene (10.24%),epizonarene (8.99%) and α-pinene (7.22%) were the main components.The IC50 value of DPPH radical scavenging was 18.25 mg/mL;inhibition ratio of the volatile oil (1.8 mg/mL) was 25.08% in β-carotene-linoleate inhibition method.The cytotoxic activity of the volatile oil to BGC-823,A-549 and HeLa cell lines with IC50 value of 37.597,46.733 and 48.656 μg/mL,respectively.[Conclusion] The volatile oil of Amorpha fruticosa fruits has less antioxidant activity,however,it has excellent cytotoxic activity to HeLa cell lines.
