不同组培条件对芦笋试管苗玻璃化的影响

摘要
为有效解决芦笋组培快繁继代增殖培养4~6代时,由于玻璃化试管苗的严重出现而影响增殖效率的问题,对6-BA浓度、培养温度、光照强度、蔗糖浓度、琼脂浓度等因素对芦笋组培玻璃化及增殖效果的影响进行研究.结果显示,玻璃化率随着培养温度和6-BA浓度的降低而有所降低,随光照强度、蔗糖浓度和琼脂浓度的增强而减弱.最终选择MS培养基,附加蔗糖50 g/L,琼脂粉8g/L,光照强度4200 lx,先6-BA 0.3 mg/L的培养基上培养一代,再在6-BA 0.5mg/L的培养基上培养,每周期先在20℃培养15 d,再在25℃培养10 d的方式来度过芦笋增殖培养玻璃化高峰期.
Abstract
Factors affecting the vitrification and proliferation during the 4-6 generations of Asparagus officinalis L.were studied in tissue culture,such as 6-BA concentration,temperature,light intensity,sucrose concentration,agar concentration and other factors.Results showed that vitrification percentage reduced as the culture temperature and 6-BA concentration decreased,and weakened when the light intensity,sucrose concentration and agar concentration enhanced.MS culture medium adding 50 g/L sucrose and 8 g/L agar powder was selected with the light intensity being 4 200 lx.The first generation was cultured on medium with 0.3 mg/L 6-BA,and then transferred to medium with 0.5 mg/L 6-BA.In each cycle,A.officinalis was firstly cultured at 20 ℃ for 15 days,and then at 25 ℃ for 10 days.By this way,A.officinalis could go through its vitrification peak.
