摘要
[目的]评价水稻品种(系)种子休眠特性,为选育具有合适休眠程度的水稻品种、解决穗萌问题提供理论依据和亲本资源.[方法]通过萌发试验对不同类型水稻品种或品系新鲜种子的休眠特性进行评价,并通过荧光定量PCR检测种子休眠相关基因OsVP1、Sdr-4和TRAB1在休眠程度不同的水稻种子中的表达差异.[结果]不同类型水稻品种(系)的休眠特性存在明显差异,新采收的茶陵野生稻种子不能萌发,18.5%的两用核不育系水稻品种(系)的种子萌发率不高于5.0%,13.3%的资源品系的种子萌发率不高于40.0%;Os-VP1 、Sdr-4和TRAB1普遍存在于不同水稻品种(系)中,且表达量差异明显,但不与休眠程度成比例.[结论]水稻种子休眠特性可通过品种改良失去或重新获得.
Abstract
[Objective] To evaluate seed dormancy of various rice varieties (lines),and provide theoretical basis and parent resources for breeding of rice varieties with suitable degree of dormancy and solving the problem of spike sprouting.[Method] By means of germination test,the dormancy of fresh mature seeds of different types of rice varieties or lines were evaluated.Meanwhile,by fluorescence quantitative PCR method,the expression of seed dormancy-related genes OsVP1,Sdr-4 and TRAB1 were measured in seeds of different rice varieties or lines.[Result] Seeds of different rice varieties or lines exhibited remarkable difference in dormancy degree,fresh matured seeds of chaling wild rice did not exhibit germinability,the germination percentage of seeds of 18.5% dual-purpose genic male sterile lines was not more than 5.0%,the germination percentage of seeds of 13.3% resource lines for rice breeding was less or equal to 40.0%;OsVP1,Sdr-4 and TRAB1 were widely identified and showed significant difference in expression in various types of rice seeds,but did not exhibit linear relationship with dormancy degree.[Conclusion] Seed dormancy of rice varieties can be regained or weakened by breed improvement.
