摘要
[目的]采用改进的双波长法测定混合糖液中葡萄糖和木糖含量.[方法]采用分光光度法(Douglas法)对标准糖溶液的糖含量进行了测定,并对传统的双波长法的波长选择和标准曲线拟合进行了改进:通过误差分析确定测定波长,对木糖显色反应在550nm处的标准曲线进行二次方程拟合,使其更适于低浓度木糖的检测.[结果]该方法可同时测定混合体系中葡萄糖和木糖的含量,总糖、木糖和葡萄糖测定的回收率均为94.00%~101.60%,准确度较高.[结论]该方法能满足植物纤维水解液中糖含量的常规分析要求.
Abstract
[Objective] To detect the accharide contents of glucose and xylose in duplex mixture by improved dual-wavelength spectrophotometry.[Method] Saccharide content in standard solution was detected by spectrophotometry (Douglas method).The wavelength selection and standard curve fitting were improved in traditional dual-wavelength method : Detection wavelength was detected by error analysis.Quadratíc equation fitting of standard curve was carried out at 550 nm, which was more suitable for the detection of low-concentration xylose.[Result] This method could simultaneously detect the glucose and xylose contents in the mixed system.The recovery rates of total sugar, xylose and glucose were all between 94.00% and 101.60%, which had relatively high accuracy.[Conclusion] This method meets the demand of saccharide determination for plant fiber hydrolysis.
