摘要

[目的]研究永福杜鹃组织培养离体再生体系.[方法]以永福杜鹃的带腋芽幼嫩茎段、带顶芽茎尖和不带顶芽茎尖为外植体,采用不同激素配比进行组培快繁研究,建立永福杜鹃离体快繁技术体系.[结果]适宜永福杜鹃离体培养的外植体为带顶芽茎尖,外植体消毒以0.1%氯化汞消毒5 min为宜,芽诱导最适培养基为1/2MS +2-ip 3.0 mg/L+ NAA0.10 mg/L+活性炭0.5 g/L,生根最适培养基为1/2MS+ NAA 0.8 mg/L.[结论]建立了永福杜鹃组织培养离体再生体系,为永福杜鹃离体快繁技术研究提供理论依据.

Abstract

[Objective] The aim was to study in vitro regeneration culture system of Yongfu azalea.[Method] The tissue rapid propagation was conducted on stem segments with axillary buds, stem apex with top buds, and stem shoot without top buds as explants, using various hormone ratios, in vitro rapid propagation technique system was established.[Result] Using stem apex as explant was appropriate for in vitro culture of azalea, sterilization of explants with 0.1% mercuric chloride for 5 minutes was suitable, the optimum bud induction medium was 1/2MS + 2-ip 3.0 mg/L + NAA 0.10 mg/L + activated carbon 0.5 g/L, the optimum rooting medium was 1/2MS + NAA 0.8 mg/L.[Conclusion] The in vitro regeneration culture system for Yongfu azalea is established, which can provide theoretical basis for research of Yongfu azalea in vitro rapid propagation technology.
