摘要

[目的]提高罗伊氏乳杆菌在冷冻于燥过程中的存活率.[方法]采用响应曲面法,对罗伊氏乳杆菌冻干保护剂中低聚果糖、脱脂乳粉和海藻糖这3种主要成分进行了研究,探讨罗伊氏乳杆菌最优化的冻干保护剂配方.[结果]建立的回归模型达到极显著水平,无失拟因素存在.优化的保护剂配方为:低聚果糖2.66%、脱脂乳粉10.11%、海藻糖5.90%,冻干存活率达到89.43%,较优化前提高了13.76%.[结论]响应曲面法可用于罗伊氏乳杆菌冷冻干燥保护剂的工艺参数配比优化,为其工业化生产奠定了基础.

Abstract

[Objective] To increase the survival rate of Lactobacillus reuteri after lyophilization.[Method] Response surface methodology was used to investigate the three main components,such as fructo-oligosaccharide,dried skim milk and trehalose.The optimum formula of lyophilizing protectants for L.reuteri was obtained.[Result] The regression model reached extremely significant level with no factor of lack of fit.The optimized formula was 2.66% fructo-oligosaccharide,10.11% dried skim milk and 5.90% trehalose.The survival rate of L.reuteri after lyophilization was 89.43%,which increased by 13.76%.[Conclusion] Response Surface Method can be used for the optimization of formula of lyophilizing protectants of L.reuteri,which lays foundation for the industrialized production.
