多棘海盘车酶解多肽体外抗氧化活性研究

摘要
[目的]探讨多棘海盘车多肽的抗氧化活性.[方法]时多棘海盘车中水溶性蛋白进行提取,再采用酶解法,以胰蛋白酶和木瓜蛋白酶分别酶解水溶性蛋白,经离心、透析、冻干后分别得到多肽样品T-P、P-P.通过分析T-P、P-P对羟基自由基(·OH)、超氧阴离子自由基(O2-·)、1,1-二苯基-2-三-硝基苯肼自由基(DPPH·)的清除能力及在脂质体系中的抗氧化能力来表征其体外抗氧化活性.[结果]T-P、P-P对3种自由基均具有一定的清除作用,对脂质体系的氧化也有一定的抑制作用;P-P的抗氧化活性优于T-P;T-P、P-P的抗氧化活性均弱于水溶性蛋白.[结论]研究可为多棘海盘车多肽的深入研究及开发利用提供理论依据.
Abstract
[Objective] To discuss the antioxidant activity in vitro of the enzymolysis peptide from Asterias amurensis.[Method] Water-soluble protein was extracted from A.amurensis,and then was enzymolyzed by trypsin and papain using enzyme hydrolysis method.After centrifugation,dialysis and freeze drying,peptide samples T-P and P-P were obtained.Antioxidant activity in vitro was characterized based on analyzing the scavenging activities of T-P and P-P to hydroxyl radical (· OH),superoxide anion free radical (O2-·),and 1,1 diphenyl-2-trinitrobenzene hydrazine free radicals (DPPH ·).[Result] T-P and P-P had certain scavenging action to the three free radicals,and had certain inhibitory effects on the oxidation of lipid system.[Conclusion] Antioxidant activity of P-P is superior to that of T-P;antioxidant activities of T-P and P-P were poorer to that of water-soluble protein.
