响应曲面法优化罗伊氏乳杆菌发酵培养基

摘要
[目的]优化罗伊氏乳杆菌发酵培养基,提高该菌发酵活菌数.[方法]采用Plackett-Burman以及响应曲面法对罗伊氏乳杆菌发酵培养基进行研究,确定罗伊氏乳杆菌发酵培养基中显著影响发酵活菌数的因素,并通过响应曲面法进行优化,以确定最优发酵培养基组分.[结果]试验表明,所得二次回归模型达到极显著水平,无失拟因素存在.优化后的罗伊氏乳杆菌发酵培养基组成为:葡萄糖3.09%、酵母粉4.19%、果蔬汁10.03%,在此条件下发酵液活菌数可以达到8.25×109 CFU/mL.[结论]研究证实了响应曲面法可用于优化提高罗伊氏乳杆菌发酵活菌数,为其工业化生产奠定了基础.
Abstract
[Objective] To increase the viable count of submerged fermentation of Lactobacillus reuteri.[Method] Plackett-Burman design and response surface methodology were used to investigate the fermentation medium of L.reuteri.Factors significantly affected the viable count of L.reuteri were detected in fermentation medium.Optimization was carried out by response surface method.The combination of optimal fermentation medium was detected.[Result] The second-order regression models reached extremely significant level without the lack of fit.The optimized culture medium was 3.09% glucose,4.19% yeast extract powder and 10.03% fruit and vegetable juice.Under this condition,the viable count of fermentation broth was 8.25 × 109 CFU/mL.[Conclusion] Response surface method is suitable for the enhancement of viable count of L.reuteri fermentation broth,and lays foundation for the industrialized production.
