摘要
[目的]解决罗汉果和山药破璧草本DNA提取困难,而无法用分子生物学鉴定的问题.[方法]通过比较常用DNA提取试剂盒法和改良的CTAB法,利用sbA-trnH片段鉴定罗汉果和山药破壁草本.[结果]改良的CTAB法可从罗汉果和山药的药材、破壁粉末和破壁草本3种形态中成功地提取出基因组DNA,并可进一步利用DNA条形码的psbA-trnH片段进行分子鉴定.[结论]该方法稳定可行,可用于罗汉果和山药的药材、破壁粉末和破壁草本的DNA条形码鉴别.
Abstract
[Objective] To solve the problems of difficult extraction of DNA extraction from ultra-fine granular powers of SIRAITIAE FRUCTUS and DIOSCOREAE RHIZOMA.[Method] Ultra-fine granular powers of SIRAITIAE FRUCTUS and DIOSCOREAE RHIZOMA were identified by psbA-trnH fragment by comparing the traditional DNA extraction kit and improved CTAB method.[Result] The improved CTAB method could successfully extract the medicinal materials,ultra-fine powders and ultra-fine herbs of SIRAITIAE FRUCTUS and DIOSCOREAE RHIZOMA.psbA-trnH fragment in DNA barcode was used for molecular identification.[Conclusion] This method is stable and feasible,and can be used for the DNA barcode identification of medicinal materials,ultra-fine powders and ultra-fine herbs of SIRAITIAE FRUCTUS and DIOSCOREAE RHIZOMA.
