摘要

[目的]获得大孔吸附树脂法纯化红皮云杉多酚的最佳工艺条件.[方法]通过静态试验筛选大孔吸附树脂,然后再进行动态试验,通过单因素和正交试验,得到纯化红皮云杉多酚的最佳工艺条件.[结果]以D-101为固定相介质,红皮云杉多酚的最佳纯化工艺条件为上样浓度1.5 mg/mL、上样量25 mL、径长比1∶25、洗脱剂浓度70%、洗脱流速2.0 mL/min,多酚得率为56.88%.[结论]该研究为红皮云彩的开发利用提供了理论依据.

Abstract

[Objective] To obtain the optimal purification conditions of polyphenol from Picea koraiensis Nakai by macroporous adsorption resin.[Method] Macroporous adsorption resin was screened by static experiment.Dynamic experiment was carried out.Single factor and orthogonal tests obtained the optimal technology of purified polyphenol from P.koraiensis.[Result] With D-101 as stationary phase medium,the optimal purification technology was as follows:1.5 mg/mL sample concentration,25 mL sample volume,1 ∶ 25 diameter-length ratio,70%eluent concentration,and 2.0 mL/min elution velocity.The extraction rate of polyphenol was 56.88%.[Conclusion] This research provides theoretical foundation for the development and utilization of P.koraiensis.
