摘要
[目的]提高油茶籽饼粕多糖的提取率.[方法]以榨油后的油茶籽饼粕为原料,利用酶水解与传统热水浸提相结合的方法提取油茶籽饼粕多糖.考察了中性蛋白酶、纤维素酶、半纤维素酶及果胶酶对油茶籽饼粕多糖提取率的影响,对提取工艺中酶解温度、酶解时间、酶添加量分别进行单因素试验,在此基础上利用3因素3水平的响应面法优化工艺参数.[结果]中性蛋白酶法提取油茶籽饼粕多糖的酶解最佳工艺条件为酶解温度40℃、酶解时间120 min、加酶量2.0%,此工艺条件下粗多糖得率为15.66%,是优化前得率7.65%的2.05倍.[结论]该方法提取率高,是一种有效的油茶籽饼粕多糖提取方法.
Abstract
[Objective] To improve the extraction rate of the polysaccharides from camellia seed cake.[Method] With camellia seed cake after oil manufacture as the research material,polysaccharides were extracted by pectinase hydrolysis combined with traditional hot water extraction.Effects of neutral protease,cellulase,hemicellulase and pectinas on the polysaccharide extraction rate were researched.Single factor test was carried out to research the effects of three factors (enzymolysis temperature,enzymolysis time and enzyme dosage) on the polysaccharides extraction rate.Response surface methodology was applied to optimize the optimal conditions.[Result] The optimal extraction technology was as follows:40 ℃ enzymolysis temperature,120 min enzymolysis time and 2.0% enzyme dosage.Under these conditions,the average extraction rate of polysaccharides was 15.66%,which was 2.05 times before optimization (7.65%).[Conclusion] This method has high extraction rate of polysaccaride,and is a kind of effective method for polysaccharide extraction.
