摘要

[目的]建立高效的发根农杆菌诱导草莓生根再生体系.[方法]以适合安徽省栽培的草莓优良品种丰香为试材,利用发根农杆菌R1601侵染植物细胞,观察发根农杆菌诱导草莓离体叶片产生毛状根的效果.[结果]发根农杆菌R1601培养成功;Carb有效除菌浓度为500 mg/L;当发根农杆菌R1601的菌液OD600 =0.6时,草莓叶片毛状根的诱导率最高,而高于或低于该密度,诱导率均呈下降趋势.[结论]建立了发根农杆菌诱导草莓毛状根再生体系,为草莓利用发根农杆菌进行抗除草剂bar基因转化研究奠定基础.

Abstract

[Objective] To establish high-efficient root regeneration system of Fragaria ananassa Duch by.Agrobacterium rhizogene.[Method] With cultivar Toyonoka suitably cultivated in Anhui Province as the research material,plant cell was infected by A.rhizogene R1601.Grwoth of Agrobacterium-induced hairy roots from F.ananassa leaves were observed.[Result] A.rhizogene R1601 was successfully cultivated.The effective sterilization concentration of Carb was 500 mg/L.When the OD600 of A.rhizogene R1601 was 0.6,the induced frequency of hairy roots was the highest.Otherwise,the induction rate all reduced.[Conclusion] Ahigh-efficient root regeneration system of F.anarassa by A.rhizogene is established,which lays foundation for the researches on bar gene transformation in F.ananassa by using A.rhizagene.
